The objective of this study was to determine the optimal level of dietary soybean oil (SBO) in high-concentrate diet for maximal ruminal escape of unsaturated fatty acid (USFA) when fed to sheep, and to determine duodenal and ileal flows of FA and other nutrients in response to supplemental SBO. Eight wether lambs (39.9 ±3.0 kg BW) fitted with ruminal, duodenal, and ileal cannulae were used in a replicated 4x4 Latin square designed experiment. Diets were isonitrogenous (13.8% CP) and included bromegrass hay, cracked corn, SBO, corn gluten meal, urea, and limestone. Soybean oil was added to the diets at 0, 3.2, 6.3, and 9.4% of dietary DM, and dietary forage was 18.4%. The diets were fed at maintenance (1.4% of BW) in two equal allotments at 0630 and 1830. Wethers had ad libitum access to fresh water and trace mineralized salt. After 14 d of dietary adaptation, Cr203 (2.5 g) was dosed at each feeding for 7 d followed by ruminal, duodenal, ileal, and fecal sample collections for 4 d. Duodenal flow of most individual and total FA increased linearly with increasing dietary SBO. Duodenal appearance (g/d) of IS:!“?15”11 increased linearly (P < 0.01) with increasing dietary SBO. The diet containing 9.4% SBO provided the greatest (P < 0.01) duodenal flow of 18:2 and 18:3. Biohydrogenation of 18:2 and 18:3 increased linearly (/* < 0.01) with increasing dietary SBO. However, biohydrogenation of 18:2 and 18:3 with greater dietary SBO was not high enough (90-93%) to prevent greater (P < 0.01) flows of these fatty acids to the duodenum. Greater dietary SBO resulted in no changes in duodenal appearance of cis-9, trans-ll CLA (P = 0.89), but appearance of trans-W, cis-12 and c/s-10, cis-\2 CLA was greater (P < 0.01) with increasing level of SBO. Increasing dietary SBO resulted in a linear increase (P < 0.05) in net flows of total FA, TSFA, and MUFA, whereas a linear decrease (P < 0.01) in net flows of PUFA and TUFA occurred. Increasing dietary SBO supplementation depressed total tract digestibility of OM (P = 0.09), lower tract and total tract digestibility of starch (P < 0.10), total tract digestibility of NDF (P = 0.07), as well as lower tract and total tract digestibility of total FA (P = 0.01). However, SBO supplementation had no effects on N digestibility at any site of the intestinal tract (P > 0.10). The diet containing 6.3% SBO provided the greatest concentration of total ruminal VFA and propionate. It was concluded that 6.3% SBO was the most benefit in terms of appearance of 18:2, 18:ltrans"n, and CLA in the duodenum, and no real benefit, biological or financial, would be observed by feeding 9.4% SBO.
