Several commercial marine fisheries have strongly declined or even collapsed in the last decades. To ensure sustainable exploitation of fish stocks, it is crucial to match management units to the biological populations. Brill (Scophthalmus rhombus) is a flatfish of high commercial value and is caught as a by-catch in the trawl fishery, which primarily target sole and plaice. Being a by-catch species, brill has never been analytically assessed and the status of the stocks are unknown. Moreover, basic life-history information is still lacking which hampers sustainable exploitation of this species. In this study we investigate the evolutionary significant population units of brill in the Northeast Atlantic Ocean. Samples were collected at 16 different locations with replicates available for some locations. The level of genetic differentiation is determined with 19 neutral microsatellite loci. In general, the genetic variation is high and does not differ much between the populations. For the majority of the populations, the observed heterozygosity is lower than expected under the Hardy-Weinberg equilibrium. The most likely explanation for this deficit of heterozygotes is the Wahlund effect. Strongest divergence was found between the Atlantic and Mediterranean Sea, the latter being represented by a single sample from the Adriatic Sea. Within the Northeast Atlantic the observed global genetic differentiation is low, suggesting a certain genetic homogeneity and at least some dispersion between populations. Nevertheless, when temporal replicates are pooled some populations show indications for genetic differentiation. The samples from the Kattegat and Skagerrak in the North might be genetically differentiated from those of the Bay of Biscay and Irish Sea in the South- Southwest. In studies on other flatfish species such as plaice, turbot and sole, this differentiation has been confirmed. Our data on neutral genetic markers will serve as a baseline for future studies examining the effects of selection. Although neutral microsatellites suggest genetic homogeneity, significant heterogeneity might still exist in adaptive variation. One of the primary future objectives should be to search for evidence of local adaptation in brill.
