Özet
Amaç: Bu tez çalışmasında antikanser etkisi kanıtlanmış olan vulpinik asit'in (VA), meme (MCF7), akciğer (A549) ve karaciğer (HepG2) hücrelerinde oksidatif stres parametrelerini etkileyip etkilemediği; süperoksit dismutaz (SOD), katalaz (CAT) ve glutatyon sistemi üzerindeki etkisinin gen, protein ve enzim aktivitesi seviyelerinde araştırılması amaçlanmıştır. Yöntem: Bu çalışma kapsamında, belirtilen hücre hatları ile VA'nın muamelesi daha önce grubumuz tarafından belirlenen IC50 değeri ve süresince yapıldı. Oksidatif stres belirteci olan indirgenmiş glutatyon (GSH), malondialdehit (MDA) ve reaktif oksijen türleri (ROS) seviyeleri ölçüldü. SOD, CAT ve glutatyon sistem genlerinin kantitatif ekspresyonuna Real-Time PCR (qPCR) yöntemi ile bakıldı. Bu genlerin kantitatif protein ekspresyonu Western Blot yöntemiyle ve enzimatik aktiviteleri spektroskopik yöntemlerle tespit edildi. Bulgular: VA, her üç hücre hattında da oksidatif stres parametreleri olan MDA ve ROS seviyelerini artırırken GSH seviyelerini azalttığı tespit edildi. Her üç hücre hattında da gen ve protein seviyelerinde VA kaynaklı değişimler gözlendi. Ayrıca VA'nın her üç hücre hattında SOD, CAT ve glutatyon sistemi ile ilişkili tüm enzimlerin aktivitesini azalttığı belirlendi. Sonuç: VA, MCF7, A549 ve HepG2 kanser hücrelerinde oksidatif stresi artırarak, antioksidan sistemleri fonksiyonel boyutta inhibe ederek, detoksifikasyon sürecini bozarak ve ferroptosis tetiğini çekerek antikanser etki göstermiştir.

Purpose: The central objective of this thesis is to investigate the impact of vulpinic acid (VA), a substance that has demonstrated anticancer efficacy, on oxidative stress parameters in breast (MCF7), lung (A549), and liver (HepG2) cells. Additionally, the study will examine its effects on superoxide dismutase (SOD), catalase (CAT), and the glutathione system (GPx, GR, and GST) at the gene, protein, and enzyme activity levels. Method: In this study, the specified cell lines were treated with VA at the IC50 value and duration previously determined by the present group. Levels of reduced glutathione (GSH), malondialdehyde (MDA), and reactive oxygen species (ROS), which are markers of oxidative stress, were measured. The quantitative expression of the genes encoding SOD, CAT, and the glutathione system (GPx, GR, and GST) was determined using Real-Time PCR. The quantitative protein expression of these genes was determined by Western Blot, and their enzymatic activities were determined by spectroscopic methods. Findings: The present study demonstrates that, in accordance with the data obtained, the administration of VA increased the levels of the oxidative stress parameters MDA and ROS, concomitant with a diminution of GSH levels in all three cell lines. Changes at the gene and protein levels were observed in all three cell lines due to VA. Furthermore, VA was determined to reduce the activity of all enzymes associated with the SOD, CAT, and glutathione systems in all three cell lines. Results: In the present study, the anticancer effect of VA was demonstrated in MCF7, A549, and HepG2 cancer cells. This effect was characterized by oxidative stress induction, inhibition of antioxidant systems, disruption of detoxification, and triggering of ferroptosis.
