Yerel Mavidil virusu izolatlarının Plak Redüksiyon Nötralizasyon (PRN) ve Reverz Transkriptaz Polimeraz Zincir Reaksiyonu (RT-PZR) teknikleri ile serotipik identifikasyonu

Özet
Bu çalışmada 1998-2005 yılları arasında izole edilen yerel BT viruslarının serotipik identifikasyonu amaçlanmıştır. Bu amaç için jenerik ve serotip spesifik RT-PZR sistemleri ile PRN testi tasarlanmıştır. Jenerik RT-PZR segment 10 düzeyinde daha önce bildirilen (Nikolakaki ve ark., 2005) primerler kullanılarak gerçekleştirilirken, sRT-PZR segment 2 düzeyinde ve serotip 2 ve 4 için daha önce Mertens ve ark. (2007) tarafından, serotip 9 ve 16 için ise bu çalışma kapsamında tasarlanan primerler kullanılarak gerçekleştirildijRT-PZR ile araştırma kapsamında çalışılan 26 adet BTV izolatının tamamında Seg-10 spesifik DNA ürünleri elde edilebilmiştir. Diğer taraftan sRT-PZR uygulaması sonunda ise izolatlardan 19 adedi serotip 9 ve 1 adedi serotip 16 olarak identifiye edildi. sRT-PZR sonunda serotip 2 ve 4 primerleri ile herhangi bir DNA ürünü elde edilemediMoleküler identifikasyon verileri kullanılarak PRN testinin ana komponenti olan tip spesifik hiperimmun serumların elde edilmesi için 3 referans virus (BTV-4, BTV-9 ve BTV-16) tavşanlara inokule edildi. İnokulasyon takvimi sonunda hiperimmun serumlarda her 3 virusa karşı 1:10 düzeyinde nötralizan antikor titresi belirlendi. Bu serumlar kullanılarak yapılan PRN testi sonunda 19 izolat BTV-9, 1'er izolat BTV-4 ve BTV-16 olarak identifiye edildiSonuç olarak RT-PZR direkt olarak laboratuar ve kullanıcı profillerinden etkilense de sRT-PZR ve PRN testleri arasında BT virusu serotiplerinin tanısı açısından benzer duyarlılık tespit edildi.

In this study it is aimed to serotypic identification of local Bluetongue viruses (BTV) isolated between 1998 and 2005. For this purpose, generic (g) and serotype specific (s) RT-PCR systems and Plaque Reduction Neutralization Assay (PRNA) were designed. Generic RT-PCR was performed on segment 10 level using primers designed previously (Nikolakaki et al.). On the other hand, sRT-PCR was carried on segment 2 with primers sets two of which were designed previously by Mertens et al. (2007) while remaining two (BTV-9 and BTV-16) were designed within this studyAt the end of gRT-PCR applications, specific DNA product (822 bp in lenght) was amplified in all of 26 BTV field isolates. On the other hand, based on sRT-PCR, 19 BTV isolates were identified as serotype 9 and one isolates was found as serotype 16. There was no specific DNA amplification as a result of sRT-PCRUsing molecular identification data as a main component of PRNA hyperimmun serums aganist three serotypes (BTV-4, BTV-9 and BTV-16) were produced in rabbits. Neutralizing antibody level in hyperimmun serums for all of three viruses were detected as 1:10. PRNA of field isolates of BTV using hyperimmune serums revealed nineteen BTV-9, one BTV-4 and one BTV-16 serotypic distribution among the isolatesIn conclusion, although RT-PCR is directly influenced by laboratory or user profile, sRT-PCR and PRNA were found in almost similar sensivity in diagnosing BTV serotypes.
