Özet

Bu çalışmada, Erzincan ilinde üretilen ve satışa sunulan, Erzincan Tulum Peynirlerinin, Listeria monocytogenes varlığı yönünden konvansiyonel ve moleküler yöntemlerle incelenmesi amaçlanmıştır.Bu amaçla 100 adet tulum peyniri materyal olarak kullanılmıştır. Listeria spp. izolasyonunda ISO 11290-1/A1-2004 metodu kullanılmıştır. Elde edilen Listeria spp. izolatlarının genus ve tür düzeyinde identifikasyonu ise sırasıyla iap gen bölgesi ve listeriolysine gen bölgesine spesifik primerler kullanılarak yapılan Polimeraz Zincir Reaksiyonu ile gerçekleştirilmiştir. Çalışma sonucunda tulum peynir örneklerinin 3'ü (% 3) Listeria spp. yönünden pozitif olarak saptanmış ve izolatların tamamı PZR ile L. monoytogenes olarak identifiye edilmiştir.Bu çalışmada, Erzincan ilinde, önemli bir tüketim potansiyeline sahip Erzincan Tulum Peynirlerinde L. monocytogenes varlığının saptanması sebebiyle bölgede etkenle bulaşık tulum peyniri tüketiminin, halk sağlığı açısından risk oluşturabileceği kanısına varılmıştır.

The aim of this study is to investigate Listeria monocytogenes existence in Erzincan Tulum Cheese produced and sold in Erzincan city by conventional and molecular methodsFor this purpose, 100 Tulum Cheese specimens are used as a starting material. The isolation of Listeria spp. was done by using ISO 11290-1/A1-2004 method. The genus and species level identification of obtained Listeria spp. isolates were performed by Polimerase Chain Reaction using specific primers in iap and listeiolysine gene region, respectively. In the light of the search results, 3 % of the analyzed Tulum Cheese specimens were found as Listeria spp. positive and all of these isolates were identified as L. monoytogenes by using PCR.In this work, the existence of L. monocytogenes were determined in Erzincan Tulum Cheese which has high consumption potential in Erzincan territory, therefore it is concluded that the consumption of infection carrying Tulum Cheese can generate risk in public health.
