Sperm cell cryopreservation (KP) is one of the methods frequently used during fertility preservation and reproductive aids. However, factors such as temperature changes, osmotic shock, formation of Reactive Oxygen Species (ROT), and Lipid Peroxidation cause irreversible damage to spermatozoa. Due to the decrease in mitochondrial membrane potential and membrane damage, there are changes in the function of channels and membrane receptors such as CNG, voltage dependent channels, CATSPER, which provide the transmission of Na +, K +, H +, HCO3- and Ca2 + in the cell. The most common negative effect of KP is the decrease in sperm viability and motility. In our study, an increase in sperm motility was achieved by inhibition of the destruction of cAMP and cGMP in the cell using Papaverine (PAP), a specific inhibitor of Phosphodiesterase10A (FDE10A). Cells were frozen by determining the motility, Acrosome Integrity (AB), Phosphatidyl Serine Translocation (FST), viability and DNA damage rate of 20 normozoospermic sperm. PAP was applied to the thawed sperm after KP, and motility activation, Acrosome Reaction (AR), viability rate, apoptosis and DNA damage formation were examined at 30 and 60 minutes. For determining PAP effectiveness, Flow Cytometry method was used after imaging under fluorescent microscopy by immunofluorescence. Our findings show that KP treatment leads to a decrease in viability and motility of sperm and an increase in DNA damage. There was no change in FST rates, which are among the markers of AB and early apoptosis formation after KP. Compared with the control group of sperm administered with PAP, total sperm motility was 30 (32.14 ± 6.90% and 25 ± 6.15%; p <0.001) and 60 minutes (31.84 ± 7.32 and 25.72 ± 6.23; p = 0.001). In the presence of PAP, there was no change in the rates of AB, FST and DNA damage between 30-60 minutes. However, in the presence of PAP, compared to the 30th minute, in the 60th minute, AnneksinV (-) / Propidium Iodide (-) cells decreased (32.64 ± 12.88% and 29.29 ± 11.78; p = 0.019) and although it is not statistically significant (p = 0.066) the number of PI (+) cells slightly increased.
