Özet

ÖZET Bu çalışmada koyun ve kuzu bronkopneumonilerinde Haemophilus/Histophilus grubu mikroorganizmaların varlığı incelendi. Haemophilus/Histophilus grubu mikroorganizmaların izolasyonu için, 1997 yılı Şubat-Nisan ayları arasında Bursa bölgesindeki üç ayrı kesimhaneden (Çalı, Çimet ve Akçalar) alınan 87 adet lezyonlu akciğer örneğinin selektif besiyerlerine (brain-heart infüzyon agar ve Fildes zenginleştirme agar) ekimleri yapıldı. İzole edilen bakterilerin koloni morfolojisi ve bireysel morfolojileri incelendi. Haemophilus/Histophilus grubu bakteriler ile uyumlu olan izolatlara cins ve tür düzeyinde identifikasyon amacı ile biyokimyasal testler uygulandı. Biyokimyasal testler referens suşlar {Haemophilus somnus, Histophilus ovis, Haemophilus agni) ile karşılaştırmalı olarak uygulandı. Cins düzeyinde identifikasyon için oksidaz, nitrat redüksiyonu, arjinin dihidrolaz, CAMP testi, X faktörü ihtiyacı, ornitin dekarboksilaz, eskulin hidrolizasyonu ve karbonhidrat fermentasyon testleri (sukroz, ramnoz, laktoz, ksiloz, mannitol, glukoz, arabinoz, sorbitol, mannoz, fruktoz) yapıldı. İncelenen 87 adet lezyonlu akciğer örneğinin 14 (%16) adetinden Haemophilus/Histophilus grubu bakteriler izole edildi. Tür düzeyinde identifikasyon için hippurat -1-hidrolizi, ONPG, p-glukosidaz, nitrit redaksiyonu, indol, üreaz, katalaz, lizin dekarboksilaz, alkali fosfataz ve karbonhidrat fermentasyon testleri (trehaloz, inositol, dulcitol) uygulandı. Haemophilus/Histophilus grubu bakteriler olarak tanımlanan 14 adet izolatın 10 adeti (%11.4) H.somnus, 3 adeti (%3.4) H.ovis, 1 adeti (%1.14) H.agni olarak identifiye edildi. Haemophilus/Histophilus grubu bakteri izole edilen hayvanların 6-8 aylık oldukları saptandı. Şubat-Mart ayları döneminde daha yüksek oranda (%65) Haemophilus/Histophilus grubu bakteri izolasyonu yapıldı. Haemophilus/Histophilus grubu bakteri izole edilen akciğer örneklerinin histopatolojik incelemelerinde, fokal, mononüklear hücre infiltrasyonları ile başlayıp, daha sonra nekrotik ve apseli görünüm alan bronkopneumoni tablosu gözlendi. Summary Bacteriological Detection of Haemophilus Species from Bronchopneumonia of Sheep. This study was undertaken to determine the presence of Haemophilus/Histophilus group in the bronchopneumonia of sheep and lambs, to isolate the Haemophilus/Histophilus grup, 87 lung samples taken from abattoirs in Bursa (Çalı, Çim e t and Akçalar) were cultured on selective media (brain-heart infusion agar, Fildes enrichment agar). The colony and microscopic morphology of the isolated bacteria were examined. Biochemical tests were performed to identify Haemophilus species and strains. The results of the biochemical tests were compared with those of reference strains (H.somnus, H.agni and Histophilus ovis). Oxidase, nitrate reduction, arginine dihydrolase, CAMP reaction, X-factor requirement, ornithine decarboxylase, esculin hydrolisation and carbonhydrate fermentation tests (sucrose, rhamnose, lactose, xylose, manitol, glucose, arabinose, sorbitole, mannose and fructose) were used to identify Haemophilus sp. In 14 (16%) out of 87 samples, Haemophilus sp. were isolated. To identify Haemophilus spp. hippurate hydrolysis, -3-ONPG test, p-glucosidase, nitrite reduction, indole production, urease, catalase, lysine decarboxylase, alkaline phosphatase and carbonhydrate fermentation tests (trehalose, inositol and dulcitol) were applied. Of the 14 isolates, 10 (11.4%) were identified as H.somnus; 3 (3.4%) as H.ovis; and 1 (1.14%) as H.agni. The animals which were found to be infected with Haemophilus/Histophilus group were between six and eight months old. The isolation rate was at its highest level during February and March. In the histopathological examination of the lung samples in which Haemophilus/Histophilus group were isolated, the appearance of the lesions ranged from focal, mononuclear cell infiltrations to necrosis and abscess formation, typical for bronchopneumonia.
