Methylene blue (MB), also known as methylthioninium chloride, is a salt commonly used as an antioxidant, antimalarial, antidepressant, acid-base indicator, and cardioprotective compound. This study was conducted to determine the embryotoxic effect of MB. A total of 360 fertile eggs were used in the study. The study was conducted using two different experiments. The lethal dose of MB was evaluated in the first experiment, and the second experiment, the embryotoxic activity was assessed. In the lethal dose experiment of the study, MB was injected into the air chamber at doses of 1, 2, 4, and 8; in the embryotoxicity experiment, it was injected into the air chamber at doses of 0.5, 0.25, 0.125, and 0.067 mg/kg. Sham and positive control groups were formed in both experiments. In lethal dose and embryotoxicity experiments, the developmental stages of the embryos on the 7th and 14th days of incubation were determined according to the Hamburger-Hamilton scale, and the distribution of embryonic deaths according to this scale was determined. The dead, alive, and relative embryo weights of the groups and the embryos' crown-rump length (CRL) were determined. The numerical data obtained were analyzed with statistical methods. In the lethal experiment, it was determined that the embryo weights were low and the mortality was high in the groups given MB (p<0.001). It was observed that MB doses of 4 mg/kg and above killed all embryos. The embryotoxicity experiment showed no negative effects on embryo development and CRL at MB doses of 0.067 - 0.5 mg/kg.The current research showed that the levels of MB employed in treatment were significantly harmful to embryonic development.
