This doctoral thesis aims to develop, characterize, and evaluate the mechanical performance of collagen-based surgical sutures with biodegradable and biocompatible properties for use in surgical applications. Within this scope, collagen surgical sutures were fabricated at different collagen concentrations (4%, 5%, and 6% w/v) using the wet spinning method with a coaxial needle configuration. To enhance the structural and mechanical properties of the produced sutures, post-processing treatments were applied using different secondary washing conditions, namely no washing (X), acetone (A), and formaldehyde (F). In addition, based on the non-mineralized sutures exhibiting the highest mechanical performance, mineralized sutures were produced using three different mineralization approaches, and these methods were comparatively evaluated. The morphological characteristics of the produced sutures were investigated using scanning electron microscopy (SEM), while the presence and distribution of mineral phases were confirmed by SEM–EDX elemental analysis and mapping. The chemical structure of the sutures and the collagen–mineral interactions were assessed using Fourier transform infrared (FTIR) and Raman spectroscopy. Mechanical performance was evaluated through tensile strength, elongation at break, Young's modulus, and fatigue tests. Furthermore, the water retention capacity of the sutures was analyzed based on diameter changes measured under dry and wet conditions. Based on the obtained results, the optimum collagen concentration, secondary washing condition, and mineralization method were determined. To assess the biocompatibility of the produced collagen-based surgical sutures and their effects on cellular responses, in vitro biological evaluations were conducted. In this context, the effects of the sutures on cell viability were examined using cell viability assays, while their influence on cell behavior and wound healing potential was investigated using scratch (wound closure) assays. In conclusion, this thesis elucidates the structure–property relationships of collagen-based surgical sutures produced by the wet spinning method by systematically investigating the effects of processing parameters, secondary treatments, and controlled mineralization. The findings provide a scientific foundation for the development of biomimetic and clinically adaptable surgical suture prototypes.
