Özet

Amaç: Bu çalışmada, Liquorilactobacillus hordei SK-6, Lactiplantibacillus plantarum VB-29 ve Lp. plantarum Y48 suşlarından elde edilen postbiyotiklerin antibakteriyel ve antioksidan aktiviteleri ile toplam fenolik madde içeriklerinin belirlenmesi; Lq. hordei SK-6 (A), Lp. plantarum VB-29 (B) ve A+B kombinasyonu (AB) postbiyotikleri kullanılarak fonksiyonel beyaz peynir üretilmesi; postbiyotikler ile üretilen beyaz peynirlerin depolama süresince bazı kalite parametreleri ile biyokimyasal özellikleri üzerindeki etkilerinin değerlendirilmesi; beyaz peynirlerde oluşan suda çözünür peptit profillerinin belirlenmesi, depolama süresince değişiminin izlenmesi ve biyoaktivitelerinin in vitro ve in silico olarak belirlenmesi; ayrıca antibakteriyel aktiviteye sahip Lp. plantarum Y48 postbiyotiği ilavesiyle üretilen beyaz peynirde, postbiyotiğin Escherichia coli ve Listeria monocytogenes üzerinde biyokoruyucu etkinliğinin incelenmesi amaçlanmıştır. Yöntem: Araştırma kapsamında, A ve B postbiyotiklerinin peynirin fonksiyonel özellikleri üzerindeki etkilerini değerlendirmek amacıyla, postbiyotikler %1 ve %3 oranlarında ayrı ayrı ve kombinasyon halinde kullanılarak beyaz peynir üretilmiştir. Üretilen peynirlerin mikrobiyolojik, fizikokimyasal ve tekstürel özellikleri, organik asit ve uçucu bileşik profilleri ve duyusal özellikleri incelenmiş; pH 4,6'da suda çözünebilir peptit ekstraktlarında kazein (KN) fraksiyonlarına ait αs1-, αs2-, β- ve κ-KN ile β-lactoglobulin türevli peptitler LC-QTOF/MS analizi ile tanımlanmıştır. Peptit ekstraktlarının antioksidan aktiviteleri DPPH, ABTS ve FRAP yöntemleri ile, antibakteriyel aktiviteleri ise mikrodilüsyon yöntemiyle belirlenmiş ve elde edilen sonuçlar in silico analizlerle desteklenmiştir. Ayrıca, patojenlerle kontamine edilmiş beyaz peynirlerde Lp. plantarum Y48 postbiyotiğinin biyokoruyucu etkisi seçici besiyerlerlerine ekim yapılarak belirlenmiştir. Bulgular: A, B ve Lp. plantarum Y48 postbiyotiklerinin antioksidan kapasitesi 465-597 mg TE/100 g, toplam fenolik madde (TPC) değerleri ise 1195-1353 mg GAE/100 g arasında bulunmuştur. Liyofilize postbiyotikler, 30 mg/mL konsantrasyonda 17-27 mm çapında inhibisyon zonları oluşturarak patojen mikroorganizmalar üzerinde güçlü antibakteriyel etki göstermiştir. Peynirde postbiyotik kullanımı, LAB sayısını azaltmıştır; artan postbiyotik konsantrasyonu ile pH, kuru madde, protein ve L* değerleri azalırken, titrasyon asitliği (TA), tuz, suda çözünür azota göre olgunlaşma indeksi ile a* ve b* değerlerinde artış gözlenmiştir. Tekstür analizleri, yüksek postbiyotik düzeylerinde sertlik, esneklik, iç yapışkanlık, sakızımsılık ve çiğnenebilirlik değerlerinin azaldığını, dış yapışkanlık değerlerinin ise arttığını göstermiştir. Organik asit profili incelendiğinde, postbiyotik kullanımı sitrik, laktik, asetik ve propiyonik asit miktarlarını artırmış; depolama süresince sitrik, pirüvik ve laktik asit azalmış, bütirik asit ise artmıştır. Uçucu bileşik analizinde 41 bileşik (9 aldehit, 9 keton, 4 alkol, 3 ester, 5 hidrokarbon, 5 benzen türevi ve 6 diğer bileşik) tespit edilmiş, A, B ve AB postbiyotikleri uçucu aroma bileşenlerinin oluşumunu artırmış ve özellikle etanol miktarı kontrol gruplarına göre belirgin olarak yüksek bulunmuştur. Duyusal değerlendirmede genel kabul edilebilirlik sıralaması %1AB > %1A > %1B > %3B > %3A > %3AB olarak belirlenmiştir. Peynirlerin 1., 60. ve 100. günlerdeki suda çözünür peptit profilleri LC-QTOF/MS ile belirlenmiş; analizler, 1. günde %3A grubunda αs2-KN, kontrol ve %1A, %3B ve %3AB gruplarında β-KN, %1B ve %1AB gruplarında ise αs1-KN türevli peptitlerin baskın olduğunu göstermiştir. 60. ve 100. günlerde, kontrol grubunda β-KN türevleri baskınlığını korurken, postbiyotik ilaveli tüm gruplarda αs1-KN türevleri baskın fraksiyon haline gelmiştir. Postbiyotik ilavesi peptit ekstraktlarının antioksidan aktivitesini artırmış, özellikle 60. günde belirgin bir artış gözlenmiştir. Antibakteriyel aktivite açısından %1 postbiyotik uygulamaları E. coli ve S. aureus üzerinde daha yüksek etki gösterirken, %3 postbiyotik kullanımında aktivite azalmıştır. In silico analizler, tüm postbiyotik içeren ekstraktlarda LLRF ve LRF peptitlerinin hem antioksidan hem de antibakteriyel aktiviteye sahip olduğunu ortaya koymuştur. 100. günde L. monocytogenes inhibisyonundaki artış, LLRF, LRF, RPKHPIKHQGLPQEVLNENLLRF, HLPLPLL, VVVPPF, FVAPFP, LKKYKVPQL, KKYKVPQL ve LFR dahil antibakteriyel peptitlerle ilişkilendirilmiştir. Patojen inoküle edilmiş peynirlerde %3 Y48 postbiyotik uygulaması, L. monocytogenes sayısını 0,13 log kob g⁻¹, E. coli sayısını ise 0,74 log kob g⁻¹ azaltmıştır. Depolama süresince L. monocytogenes sayısı 7. gün hariç 28. güne kadar artarken, E. coli sayısı düzenli olarak azalmış ve 28. gün sonunda 0,41 log kob g⁻¹ düşüş göstermiştir. Sonuç: Genel olarak, fonksiyonel beyaz peynirde postbiyotik ilavesi pH'ı düşürmüş, proteolizi artırmış ve bu etkiler yüksek konsantrasyonlarda daha belirgin olmuştur. Postbiyotik konsantrasyonunun artışı L* değerini düşürürken a* ve b* değerlerini artırmış, LAB sayısını azaltmış ve maya gelişimini desteklemiştir. Uygulama, peynirin yumuşamasına ve depolama süresince sertliğinin azalmasına yol açmıştır. Ayrıca, postbiyotik ilavesi organik asit ve uçucu bileşik üretimini artırmıştır. %1 postbiyotik uygulaması antioksidan kapasite ve patojen kontrolünde daha etkili olurken, %3 konsantrasyon antibakteriyel aktiviteyi sınırlandırmıştır. Peptidomik analizler, postbiyotik çeşidi ve konsantrasyonu ile depolama süresi değişkenlerinin KN türevli biyoaktif peptit oluşumunu ve proteoliz dinamiğini modüle ettiğini göstermiştir. Y48 postbiyotiği depolama boyunca E. coli ve L. monocytogenes üzerinde sınırlı etki göstermiş, disk difüzyon yönteminde yüksek antibakteriyel aktivite göstermesine rağmen gıda matrisindeki etkinliği belirgin şekilde azalmıştır. Bu çalışma, farklı postbiyotiklerin beyaz peynirde depolama süresi boyunca etkilerini değerlendirmek için önemli bir model sunmaktadır.

Purpose: In this study, the antibacterial and antioxidant activities and total phenolic contents of postbiotics obtained from Liquorilactobacillus hordei SK-6, Lactiplantibacillus plantarum VB-29, and Lp. plantarum Y48 strains were determined; functional white cheeses were produced using postbiotics of Lq. hordei SK-6 (A), Lp. plantarum VB-29 (B), and their combination (AB); the effects of postbiotic incorporation on selected quality parameters and biochemical properties of white cheeses during storage were evaluated; the water-soluble peptide profiles formed in white cheeses were characterized, their changes during storage were assessed, and their bioactivities were determined through in vitro and in silico analyses; additionally, the bioprotective efficacy of white cheese produced with the antibacterial Lp. plantarum Y48 postbiotic against Escherichia coli and Listeria monocytogenes was investigated. Method: For this study, white cheeses were produced using A and B postbiotics individually and in combination at concentrations of 1% and 3% in order to evaluate their effects on the functional properties of cheese; the microbiological, physicochemical, and textural properties, organic acid and volatile compound profiles, and sensory characteristics of the cheeses were investigated; peptides derived from casein (CN) fractions, including αs1-, αs2-, β-, and κ-CN, together with β-lactoglobulin-derived peptides in water-soluble peptide extracts at pH 4.6 were identified by LC-QTOF/MS analysis; the antioxidant activities of peptide extracts were determined using DPPH, ABTS, and FRAP assays, while their antibacterial activities were evaluated by the microdilution method, and the obtained results were further supported by in silico analyses; additionally, the bioprotective effect of Lp. plantarum Y48 postbiotic in pathogen-contaminated white cheeses was determined by enumeration on selective media. Findings: The antioxidant capacity of A, B, and Lp. plantarum Y48 postbiotics was found to range between 465–597 mgTE/100 g, while total phenolic content (TPC) values were determined to be between 1195–1353 mgGAE/100 g; lyophilized postbiotics exhibited strong antibacterial activity against pathogenic microorganisms by forming inhibition zones of 17–27 mm at a concentration of 30 mg/mL; the use of postbiotics in cheese reduced lactic acid bacteria (LAB) counts, while increasing postbiotic concentration led to decreases in pH, dry matter, protein, and L* values, whereas titratable acidity (TA), salt content, ripening index based on water-soluble nitrogen, and a* and b* values increased; texture profile analysis showed that higher postbiotic levels resulted in decreased hardness, springiness, cohesiveness, gumminess, and chewiness, while adhesiveness increased; organic acid profiling revealed that postbiotic application increased citric, lactic, acetic, and propionic acid contents, whereas during storage citric, pyruvic, and lactic acids decreased and butyric acid increased; volatile compound analysis identified 41 compounds (9 aldehydes, 9 ketones, 4 alcohols, 3 esters, 5 hydrocarbons, 5 benzene derivatives, and 6 other compounds), and A, B, and AB postbiotics enhanced the formation of volatile aroma compounds, with ethanol levels being markedly higher compared to control groups; sensory evaluation indicated the overall acceptability ranking as 1%AB > 1%A > 1%B > 3%B > 3%A > 3%AB. Water-soluble peptide profiles of cheeses on days 1, 60, and 100 were determined by LC-QTOF/MS analysis, showing that at day 1, αs2-CN-derived peptides predominated in the 3%A group, β-CN-derived peptides in the control, 1%A, 3%B, and 3%AB groups, and αs1-CN-derived peptides in the 1%B and 1%AB groups; at days 60 and 100, β-CN-derived peptides remained dominant in the control group, whereas αs1-CN-derived peptides became predominant in all postbiotic-treated groups; postbiotic addition increased the antioxidant activity of peptide extracts, with a marked enhancement observed particularly on day 60; in terms of antibacterial activity, 1% postbiotic treatments showed higher efficacy against E. coli and S. aureus, whereas 3% postbiotic application resulted in reduced activity; in silico analyses revealed that LLRF and LRF peptides in all postbiotic-containing extracts possessed both antioxidant and antibacterial activities; the increased inhibition of L. monocytogenes on day 100 was associated with peptides including LLRF, LRF, RPKHPIKHQGLPQEVLNENLLRF, HLPLPLL, VVVPPF, FVAPFP, LKKYKVPQL, KKYKVPQL, and LFR; in pathogen-inoculated cheeses, 3% Y48 postbiotic application reduced L. monocytogenes counts by 0.13 log CFU/g and E. coli counts by 0.74 log CFU/g; during storage, L. monocytogenes counts increased up to day 28 except on day 7, whereas E. coli counts decreased steadily, reaching a 0.41 log CFU/g reduction by day 28. Results: Overall, the addition of postbiotics to functional white cheese decreased pH and enhanced proteolysis, with these effects being more pronounced at higher concentrations. Increasing postbiotic levels reduced L* values while increasing b* values, decreased LAB counts, and promoted yeast growth. The application also contributed to cheese softening and reduced hardness during storage. Furthermore, postbiotic supplementation enhanced the production of organic acids and volatile compounds. A 1% postbiotic application was more effective in terms of antioxidant capacity and pathogen control, whereas a 3% concentration limited antibacterial activity. Peptidomic analyses indicated that postbiotic type, concentration, and storage time modulated the formation of CN-derived bioactive peptides and proteolysis dynamics. The Y48 postbiotic exhibited limited effects on E. coli and L. monocytogenes during storage; although it showed high antibacterial activity in disk diffusion assays, its efficacy was markedly reduced within the food matrix. This study provides a relevant model for evaluating the effects of postbiotics on white cheese during storage.
