Flooding is a frequently occurring environmental stress that can severely affect plant growth. In 1993 and 1994, two Hoods occured in the United States that caused millions dollars of damage to farmers in the Midwest and the South. The floods increased the awareness of farmers to the the importance of flooding tolerant plants. In this study, the effects of flooding on Arabidopsis thaliana was studied at molecular levels. Sixty seven A. thaliana mutant pools (about 7.000 individual plants) were screened for flooding tolerance by submerging the roots of three-week old plants in water for 15 days. Seeds were harvested from individual susceptible plants that were stunted, and yellow. Kanamycin resistance tests were done to determine the presence of functional T-DNA insertions. Plants with a functional T-DNA insertion were retested for flooding susceptibility. The number of insertion were checked for 74 plantd using Southern blot and tluee plants with single insertion pattern were chosen. Flood susceptible plants with single insertion were crossed with wild type (Ws) plants, Fi plants were selfed and cosegregation analysis was done on F2 plants for kanamycin resistance and susceptibility to flooding. The three plants that showed cosegregation for the traits studied were chosen for molecular analysis. Flanldng plant genomic sequences to the T-DNA insertion were isolated by the plasmid rescue technique, and sequenced. The sequences were compared with known genes in data base and genbank. The left border of one mutant has homology with the Arabidop:iis ubiquitin conjugating enzyme (E2) whereas the right border of same mutant has homology witli a mouse Ran-GTPase gene responsible for nuclear protein import and RNA processing. In addition, one left border and one right border of tlie other two mutants show homology Wiih. Arabidopsis ribosomal RNA.
