7. ÖZET Tribulus terrestris L. (Demirdikeni) halk arasında kolik ağrılarda, anginal ağrılarda ve yüksek kolesterolün düşürülmesinde yaygın olarak kullanıl maktadır. Çalışmamızda kurutulmuş Tribulus terrestris L. (Demirdikeni) bit kisinden özel bir ekstraksiyon yöntemi ile elde edilen liyofilize saponozit karışımının düz kaslara etkisi araştırıldı. Deneylerde izole tavşan aortu, tavşan jejunumu ve koyun üreteri prepa ration kullanıldı. Preparatlarm seçiminde halkın bitkiyi kullanım amaçları gözönünde tutuldu. Önce HPLC ile saponozit karışımı içindeki sapogeninle- rin tespiti yapıldı. Muhtemelen diosgenin ve tigogeninin karışımdaki ana sapogeninler olduğu ve etkinin de bunlardan kaynaklandığı düşünüldü. Çalışmalarımızda Litchfield-Wilcoxon metodu ile İsviçre albinosu farelerde LD50 değeri bulundu. Ekstre farelere i.p. yoldan verildi. Bu değer 813 mg/kg ve % 95 güvenlik sınırları 739 mg/kg-894 mg/kg olarak bulundu.59 Saponozit karışımının izole aort preparatlarmda herhangi bir etkisi yok tu. 8. SUMMARY Tribulus terrestris L. has been commonly used in folk medicine against colic, anginal pains and to decrease the plasma cholesterol level. This study investigated the effects of liophilized material on several smooth muscles. The liophilized material was obtained from dried and powdered Tribulus terrestris L. by specific extraction method for saponins. To this end, isolated rabbit aorta, rabbit jejunum, and sheep ureter preparations were used. The reason for selecting these preparations for the experiments was due to the fact that the saponin mixture is commonly used in public medicine against colic, anginal pains and spasms. ' ' '“ ”- >- At the outset, sapogenins in saponin mixture were analyzed using HPLC. It was thought that diosgenin and tigogenin were main sapogenins and they would be responsible for the possible effects on the preparations used. Next, median lethal dose (LD50) on Swiss albino mice was obtained using Litchfield- Wilcoxon method. The extract has been administered to mice by i.p. route median lethal dose and its 95 % confidence limits were found to be 813 and61 739-894 mg/kg respectively. At the used concentrations, the extract did not have any significant effects on the isolated aorta preparation.
