Toll-like receptors (TLRs) and retinoic acid-inducible gene (RIG)-I-like receptors (RLRs) are significant elements of the innate immune system as a Pathogen recognition receptor (PRR). Aquaculture species such as Nile tilapia encounters with potential disease-causing pathogens because of intensive aquaculture. However there is not enough information about their PRRs compared to model teleost species to understand the underlying molecular regulation of pathogen detection for effective solutions. We identified TLRs and RLRs and evaluated their evolutionary relationship to provide initial insight into PRR-driven signaling pathway in Nile tilapia. Genes that involves viral response were selected to clone and screen their expression in selected tissues. The Nile tilapia genome contains 19 TLR genes that include mammalian and fishes specific TLRs with duplicated genes. The Nile tilapia genome posses the members of RLRs: an MDA5 gene and interestingly two LGP2 genes. Phylogenetic analysis indicated that Nile tilapia TLRs and RLRs clustered with their counterparts from other teleost and mammals. TLRs and RLRs have the conserved structure and selected genes were expressed in immune-related tissues. These results mean that TLRs and RLRs expanded in Nile tilapia genome maybe because of selective pressure and they have significant roles in recognition of pathogens.
