Özet
Polifosfat (poliP) tüm organizmalarda bulunan önemli bir enerji ve fosfat depolama polimeridir. Streptomyces'te poliP kinaz (PPK) geninin silinmesi, antibiyotik aşırı üreten bir fenotiple sonuçlanmıştır. Bununla birlikte, yüksek antibiyotik üreticisi Streptomyces ∆ppk suşlarının endüstriyel kullanımı, oksidatif strese karşı artan duyarlılıkları nedeniyle mümkün olamamaktadır. Demir, çeşitli hücresel süreçler için gerekli bir elementtir, ancak aşırı demir, özellikle Fenton reaksiyonu yoluyla oksidatif stres oluşturur. İlginç bir şekilde, poliP'nin demire bağlanarak biyolojik kullanımını dolayısıyla oksidatif stresi azaltabildiği bilinmektedir. Bu çalışmanın amacı Streptomyces coelicolor 'da poliP metabolizması, demir homeostazı ve oksidatif stres arasındaki ilişkileri aydınlatmaktır. Bunun için, demir içeren ve demir içermeyen ortamlarda büyütülen yabani tip (WT) ve ∆ppk suşlarının üç biyolojik tekrarlı olacak şekilde karşılaştırmalı proteomik analizleri yapılmıştır. Sonuçlar, S. coelicolor'da ppk geninin silinmesinin demir yokluğunda bile toplam proteomu önemli ölçüde değiştirdiğini ve PPK'nın hücresel metabolizmada ne kadar önemli olduğunu göstermiştir. Özellikle enerji metabolizması ve protein katlanmasında rol oynayan proteinler ∆ppk mutantında WT'ye göre daha bol miktarda bulunmuştur. Demir varlığında ise fosfat metabolizması, oksidatif stres yanıtı ve translasyonda yer alan proteinlerin bolluğunun ∆ppk mutant suşunda WT'ye göre daha yüksek olduğu tespit edilmiştir. Proteom analizi dışında, çalışma kapsamında yüksek antibiyotik üreticisi ∆ppk suşunun oksidatif stres hassasiyetini gidermek için katalaz ve alkil hidroperoksit reduktaz genlerinin yüksek düzeyde ifade edilmesine yönelik çalışmalar da yürütülmüştür. Bildiğimiz kadarıyla, bu çalışma Streptomyces'te demir homeostazı, oksidatif stres ve polifosfat arasındaki ilişkiyi aydınlatan ilk çalışmadır.

Polyphosphate (polyP) is an important energy and phosphate storage polymer present in all organisms. In Streptomyces, the deletion of the polyP kinase (PPK) gene resulted in an antibiotic-overproducing phenotype. However, the industrial use of highly antibiotic producing Streptomyces ∆ppk strain is not feasible due to their increased susceptibility to oxidative stress. Iron is an essential element for various cellular processes, yet excess iron generates oxidative stress, particularly via the Fenton reaction. Interestingly, polyP can sequester iron, reducing its bioavailability and thus oxidative stress. The aim of this study was to elucidate the relationships between polyP metabolism, iron homeostasis and oxidative stress in Streptomyces coelicolor. To achieve this, comparative proteomic analyses were conducted on three biological replicates of wild-type (WT) and ∆ppk strains grown in iron-containing and iron-free media. The results indicated that even in the absence of iron, the deletion of ppk gene of S. coelicolor significantly altered the total proteome, thereby confirming the importance of PPK in the cellular metabolism. For instance, proteins involved in energy metabolism and protein folding were more abundant in the ∆ppk mutant than in the WT strain. In the presence of iron the abundance of the proteins involved in phosphate metabolism, oxidative stress response, and translation was found to be higher in the ∆ppk mutant strain than in the WT. In addition to the proteome analysis, the study sought to overcome the oxidative stress susceptibility of the ∆ppk strain, a high antibiotic producer, by overexpression of catalase and alkyl hydroperoxide reductase genes. To the best of our knowledge, this is the first study to elucidate the relationship between iron homeostasis, oxidative stress, and polyphosphate in Streptomyces.
