Sıçanlarda indometazin ile oluşturulan gastrik ülser modelinde tiyamin pirofosfatın antiülser ve hepatoprotektif etkileri

Özet
Bu çalışmada indometazin ile indüklenen sıçan ülser modelinde tiyamin pirofosfatın antiülser ve hepatoprotektif etkileri araştırıldı. Araştırmada toplam 32 adet dişi sıçan, dört gruba (randomize 8 sıçan) ayrıldı. Kontrol grubu (K), tiyamin pirofosfat grubu (TPP, 20 mg/kg, i.p intraperiotaneal), indometazin grubu (İndo, 25 mg/kg, gavaj), ve tiyamin pirofosfat+indometazin (TPP+İndo, tiyamin pirofosfat 20 mg/kg, i.p, indometazin 25mg/kg gavaj) olarak uygulandı. 6 saat sonra anastezi altında sıçanların mide ve karaciğerleri alındı. Bu dokularda makroskopik (Ülser skorları), biyokimyasal (MDA, GSH, GSH-Px ve SOD), histopatolojik analizleri gerçekleştirildi. Makroskopik analizde, midede indometazin grubunun kontrol grubuna göre anlamlı derecede ülseratif lezyonlar oluşturduğu saptandı. TPP verilen TPP+İndo grubunda, indometazin grubuna göre; ülser, nekroz, kanama odaklarını azalttığı saptandı. Biyokimyasal analizlerde İndo grubunun kontrol grubuna göre, mide ve karaciğer dokusu MDA değerlerinde istatistik anlamda artış (p˂0,05), mide ve karaciğer dokularının GSH, GSH-Px, SOD değerlerinde anlamlı derecede azalma saptandı (p˂0,05). TPP+İndo grubunun İndo grubuna göre karşılaştırılmasında; mide ve karaciğer dokusu MDA düzeylerinde istatistik anlamlılıkta azaltma (p˂0,05), mide ve karaciğer dokusunda GSH, GSH-Px SOD değerlerinde ise istatistik anlamlılıkta artış saptandı. İndo grubunda kontrol grubuna göre mide dokusu histopatolojik değerlendirilmesinde; eroziv, yangısal, hemorajik bulgular, karaciğer dokusunda ise pasif konjeksiyon, dejeneratif ve apoptotik değişiklikler saptandı. TPP+İndo grubunun İndo grubu ile mide ve karaciğer doku hasarlarının karşılaştırılmasında TPP verilmesinin doku hasarlarını istatistik derecede azalttığı saptandı. Sonuçta TPP'ın sindirim sisteminde ülser oluşumuna, bununla ilişkili karaciğer hasarına karşı koruyucu özellikte olabileceği kanaatine varıldı.

This study investigated the antiulcer and hepatoprotective effects of thiamine pyrophosphate (TPP) in a rat ulcer model induced by indomethacin. A total of 32 female rats were randomly divided into four groups (n=8 per group): Control (C), Thiamine Pyrophosphate (TPP, 20 mg/kg, intraperitoneally), Indomethacin (Indo, 25 mg/kg, orally via gavage), and Thiamine Pyrophosphate + Indomethacin (TPP+Indo; TPP 20 mg/kg, i.p. + Indomethacin 25 mg/kg, gavage). Six hours after administration, the rats were sacrificed under anesthesia, and their stomach and liver tissues were collected. Macroscopic (ulcer scores), biochemical (MDA, GSH, GSH-Px, and SOD), and histopathological analyses were performed. Macroscopic analysis revealed that the indomethacin group developed significantly more ulcerative lesions in the stomach compared to the control group. In the TPP+Indo group, the number of ulcers, necrotic areas, and hemorrhagic foci were reduced compared to the indomethacin group. Biochemically, MDA levels were significantly increased (p<0.05), and GSH, GSHPx, and SOD levels were significantly decreased (p<0.05) in the stomach and liver tissues of the indomethacin group compared to the control. In contrast, the TPP+Indo group showed a significant decrease in MDA levels and a significant increase in GSH, GSH-Px, and SOD levels in both stomach and liver tissues compared to the indomethacin group (p<0.05). Histopathological examination of the stomach tissues in the indomethacin group revealed erosive, inflammatory, and hemorrhagic findings, while the liver tissues showed passive congestion, degenerative, and apoptotic changes compared to the control group. In comparison with the indomethacin group, the TPP+Indo group showed a statistically significant reduction in tissue damage. In conclusion, TPP may have a protective effect against ulcer formation in the gastrointestinal system and related liver damage.
