Özet
Bu çalışmada, gadolinyum (III) klorür (GdCl3)'ün sucul eğrelti otu Azolla caroliniana Willd. üzerindeki fizyolojik ve biyokimyasal etkileri araştırıldı. Azot içermeyen Hoagland besin solüsyonunda sera koşullarında yetiştirilen bitkilere 1, 5, 10 ve 20 ppm GdCl3 konsantrasyonları 7 gün süre ile uygulandı. Bağıl büyüme oranı (RGR), büyüme yüzdesi, klorofil ve karotenoid içerikleri, total fenolik ve flavonoid miktarları, malondialdehit (MDA) seviyeleri, toplam antioksidan kapasite, prolin ve protein içerikleri ile peroksidaz (POD) ve katalaz (CAT) enzim aktiviteleri değerlendirildi. GdCl3 doza bağlı olarak (20 ppm) RGR'de %32'ye ve büyüme yüzdesinde %40'a varan indirgenmelere neden oldu. Klorofil a içeriği 20 ppm de yaklaşık %24 artarken, klorofil b ve karotenoid seviyeleri 1 ppm GdCl3 de sırasıyla %21,7 ve %22,1 oranında azaldı. Total klorofil içeriği düşük konsantrasyonlarda (1 ve 5 ppm) çok az indirgenirken (%3,80 ve %0,39), yüksek konsantrasyonlarda (10 ve 20 ppm) %9,3 ve %11,6 oranında arttı. Total fenolik içeriği 5 ppm'de %9 artarken, 20 ppm'de %21,7 azaldı. Total flavonoid içeriği ise 5 ppm'de %132,5 arttı. MDA seviyeleri 20 ppm'de 8,2 kat artarak oksidatif stresi, prolin ve protein içeriklerinde ise sırasıyla %45,5 ve %21,6 oranında azalarak bitkinin stres toleransının zayıfladığını ortaya koydu. POD aktivitesi 20 ppm'de %113 artarken, CAT aktivitesi %84 azaldı. Bulgular, GdCl3'ün Azolla caroliniana'da büyüme inhibisyonu, fotosentetik pigment değişiklikleri ve oksidatif stres yanıtlarını tetiklediğini göstermektedir. Çalışma, Azolla'nın nadir toprak elementlerinin (NTE'ler) toksisitesini değerlendirmede biyoindikatör olarak potansiyelini vurgulayarak, sucul ekosistemlerde kirlilik yönetimi ve fitoremediasyon stratejileri için önemli veriler sunmaktadır.

In this study, the physiological and biochemical effects of gadolinium (III) chloride (GdCl3) on the aquatic fern Azolla caroliniana Willd. were investigated. Plants were grown under greenhouse conditions in nitrogen-free Hoagland nutrient solution and treated with GdCl3 concentrations of 1, 5, 10, and 20 ppm for 7 days. Relative growth rate (RGR), growth percentage, chlorophyll and carotenoid contents, total phenolic and flavonoid contents, malondialdehyde (MDA) levels, total antioxidant capacity, proline and protein contents, as well as peroxidase (POD) and catalase (CAT) enzyme activities were evaluated. GdCl3 caused dosedependent reductions in RGR (up to 32% at 20 ppm) and growth percentage (up to 40% at 20 ppm). Chlorophyll a content increased by approximately 24% at 20 ppm, while chlorophyll b and carotenoid levels decreased by 21.7% and 22.1%, respectively, at 1 ppm. Total chlorophyll content showed minimal reduction at low concentrations (3.80% at 1 ppm and 0.39% at 5 ppm) but increased by 9.3% and 11.6% at higher concentrations (10 and 20 ppm, respectively). Total phenolic content increased by 9% at 5 ppm but decreased by 21.7% at 20 ppm. Total flavonoid content increased by 132.5% at 5 ppm. MDA levels rose by 8.2-fold at 20 ppm, indicating oxidative stress, while proline and protein contents decreased by 45.5% and 21.6%, respectively, suggesting reduced stress tolerance. POD activity increased by 113% at 20 ppm, whereas CAT activity decreased by 84%. These findings demonstrate that GdCl3 induces growth inhibition, alterations in photosynthetic pigments, and oxidative stress responses in Azolla caroliniana. The study highlights the potential of Azolla as a bioindicator for assessing the toxicity of rare earth elements (REEs) and provides valuable data for pollution management and phytoremediation strategies in aquatic ecosystems.
