Özet
Bitki paraziti nematodlar (BPN'lar) küresel tarım üretimini önemli ölçüde etkilemekte ve her yıl yüz milyarlarca ABD doları kayba neden olmaktadır. Dünya nüfusunda öngörülen artışla birlikte, bu kayıpları önlemek için bitkisel ürünlerin sağlığını ve verimini korumak çok önemlidir. Çeşitli BPN mücadele metodları mevcut olsa da, bunlar öncelikle nematisitlere dayanmaktadır. Ancak, nematisitlerin insanlara ve çevreye toksisitesi, toprak biyotası üzerindeki hedef dışı etkileri ve artan kullanım kısıtlamaları nedeniyle, daha güvenli alternatiflere ve yenilikçi yaklaşımlara çok önemli ihtiyaç vardır. Bu çalışmada, dört nematod antagonisti fungusun, Ijuhya vitellina, Niesslia gamsii, Polydomus karssenii ve JKI72728 suşunun, iki konukçuda, üç BPN, Heterodera filipjevi, Heterodera schachtii ve Pratylenchus thornei'ye karşı biyolojik mücadele potansiyeli araştırılmıştır. Çalışmada ayrıca, bu fungusların hem BPN stresi varken hem de yokken konukçu ile endofitik etkileşimi araştırılmıştır. Nematod üremesini, fungusların nematod yumurtalar parazitliğini ve nematod penetrasyonu üzerindeki etkisini değerlendirmek için çeşitli in vitro deneyler yapılmıştır. Ayrıca fubgusların BPN enfeksiyonu olan ve olmayan bitki konukçusunun morfolojik ve morfometrik özellikleri üzerindeki etkisi de incelenmiştir. Dört nematod antagonisti fungal suş, BPN enfeksiyonunu baskılamada ve çeşitli bitki büyüme parametrelerini iyileştirmede değişen seviyelerde başarı göstermiştir.

Plant parasitic nematodes (PPNs) significantly impact global agriculture production, causing hundreds of billions of USD in losses annually. With the projected increase in world population, maintaining crop health and yield to prevent these losses is crucial. While various PPN control methods exist, they primarily rely on nematicides. However, due to the toxicity of nematicides, their non-target effects on soil biota, and increasing use restrictions, there is a pressing need for safer alternatives and innovative approaches. This study investigated the biocontrol potential of four nematode antagonistic fungi, Ijuhya vitellina, Niesslia gamsii, Polydomus karssenii, and strain JKI72728, against three PPN, Heterodera filipjevi, Heterodera schachtii and Pratylenchus thornei, in the presence of a plant host. Additionally, the study explored the endophytic interaction of these fungi with the host, both with and without PPN stress. Several in vitro experiments were conducted to assess nematode reproduction, fungi parasitism of nematode eggs, and effect on nematode penetration. The influence of the fungi on the morphological, morphometric, and agronomic characteristics of the plant host, with and without PPN infection, was also examined. The four nematode antagonistic fungal strains demonstrated varying levels of success in managing PPN infection and improving several plant growth parameters. This is the first report of a dark septate endophyte (Polydomus karssenii) reducing and delaying penetration of a PPN in planta, explicitly a migratory endoparasitic nematode, the root lesion nematode P. thornei. This study highlights the biocontrol potential of the four nematode antagonistic fungi sustainable biological agents against PPNs and emphasizes their role in integrated pest management strategies for wheat production.
