Özet
Kanser, hücrelerin kontrolsüz çoğalmasıyla karakterize edilen ve tümör oluşumuna yol açabilen ciddi bir hastalık grubudur. Meme kanseri, kadınlarda en sık görülen kanser türlerinden biri olup, erken tanı ve doğru tedavi ile yaşam kalitesi artırılabilir. Histopatoloji, biyopsi ile alınan doku örneklerinin mikroskop altında incelenmesiyle kanser tanısında kritik rol oynar. Bu süreçte H&E tüm slayt görüntüleri, hücre ve doku yapılarının detaylı analizini sağlayarak, tanı ve hastalığın derecesinin belirlenmesi için temel bir kaynak sunar. Ancak el ile analiz zaman alıcı, zahmetli ve sübjektif olduğu için bilgisayar destekli yöntemlere ihtiyaç duyulmaktadır. Bu tez çalışmasında meme kanseri tanısında histopatoloji görüntüleri üzerinde mitoz analizi ve meme kanseri tespiti için yenilikçi bir metodoloji önerilmiştir. Önerilen metodoloji: 1) Veri seti oluşturma; 2) Önişleme; 3) Mitoz tespiti ve sınıflandırılması; 4) Mitoz segmentasyonu; 5) Kanserli alanın tespiti, 6) Kanser derecelendirmesi ve 7) Değerlendirme metrikleri olmak üzere yedi adım içermektedir. İlk adımda MiDeSeC veri setleri oluşturulmuştur. İkinci adım kapsamında renk normalizasyonu için CD-SNMF yöntemi önerilmiştir. Üçüncü adımda mitoz tespiti ve sınıflandırılması için YOLOv8 ile bulanık mantık tekniklerinin kullanıldığı bir yöntem önerilmiştir. Ayrıca, bu adım kapsamında mitoz tespiti performansını artırmak amacıyla derin evrişimli segmentasyon mimarisi olan DeepLabv3+ modeli iyileştirilerek kullanılmıştır. Dördüncü adımda, mitoz segmentasyonu için KANs tabanlı model önerilmiştir. Beşinci adımda kanserli alan tespiti için MST algoritması önerilmiştir. Altıncı adımda kanser derecelendirmesi için Mitoz Alan Oranı yaklaşımı önerilmiştir. Sonuncu adımda ise önceki adımlarda elde edilecek sonuçları değerlendirmek için kullanılacak metrikler özetlenmiştir. Önerilen metodoloji, tez kapsamında oluşturulan MiDeSeC ve literatürde bilinen MIDOG21, ICPR12 ve ICPR14 veri setleri üzerinde uygulanmış ve elde edilen sonuçlar metodolojinin başarılı olduğunu göstermiştir. Bu çalışma, tanı süreçlerini objektif ve tekrarlanabilir hale getirerek, patologların iş yükünü azaltmakta ve tanıda güvenilirliği artırmaktadır.

Cancer is a serious group of diseases characterized by the uncontrolled proliferation of cells, which can lead to tumor formation. Breast cancer is the most common type of cancer in women, and early diagnosis and appropriate treatment can improve quality of life. Histopathology plays a critical role in cancer diagnosis by examining tissue samples obtained through biopsy under a microscope. In this process, H&E-stained whole slide images provide detailed analysis of cellular and tissue structures, serving as a fundamental resource for diagnosis and determining the grade of the disease. However, manual analysis is time-consuming, labor-intensive, and subjective, creating a need for computer-aided methods. In this thesis, an innovative methodology is proposed for mitosis analysis and cancerous region detection in histopathology images for breast cancer diagnosis. The proposed methodology consists of seven steps: 1) Dataset creation; 2) Preprocessing; 3) Mitosis detection and classification; 4) Mitosis segmentation; 5) Cancerous region detection; 6) Cancer grading; and 7) Evaluation metrics. In the first step, MiDeSeC datasets were created. In the second step, the CD-SNMF method was proposed for color normalization. In the third step, a method utilizing YOLOv8 and fuzzy logic techniques was proposed for mitosis detection and classification. Additionally, to enhance mitosis detection performance, the deep convolutional segmentation architecture DeepLabv3+ was improved and employed. In the fourth step, a KANs-based model was proposed for mitosis segmentation. In the fifth step, the MST algorithm was proposed for detecting cancerous regions. In the sixth step, the Mitosis Area Rate approach was proposed for cancer grading. In the final step, the metrics to be used for evaluating the results obtained in the previous steps were summarized. The proposed methodology was applied to the MiDeSeC dataset created within the scope of the thesis, as well as to publicly available datasets such as MIDOG21, ICPR12, and ICPR14, and the results demonstrated the success of the methodology. This study makes diagnostic processes objective and reproducible, reduces the workload of pathologists, and enhances reliability in diagnosis.
