Özet
Serapias vomeracea, Orchidaceae familyasına ait yaygın salep orkidelerinden biridir. Bu tür, geleneksel sıcak içeceklerde ve dondurmada kıvam artırıcı olarak kullanılan salep üretimi için sıklıkla kullanılmaktadır. Ancak, kontrolsüz toplama ve yetersiz üretim oranları, tohumlarında endosperm bulunmamasından dolayı türün mikorizal mantarlara bağımlı olmasıyla birleşince, sürdürülebilirliği tehdit altındadır. Bu çalışma, yumru, sürgün ve tohum eksplantlarını kullanarak direkt ve indirekt organogenez teknikleri ile S. vomeracea türünü in vitro koşullar altında çoğaltmayı amaçlamıştır. Eksplantların en yüksek yüzey sterilizasyon başarısı %0,1 (w/v) HgCl2 ile yumrularda %67, sürgünlerde %93,75 ve tohum eksplantlarında %66,7 olarak elde edilmiştir. Üç farklı fitohormon denemesinde, sürgün eksplantlarından direkt organogenez SHM V ortamında %12,5, SHM IX ve NTM I ortamlarında ise %6,25 başarı oranı ile gerçekleşmiştir. Bu ortamlar, sırasıyla, 0.5 mg/L thidiazuron (TDZ) + 0.5 mg/L naftalen asetik asit (NAA), 1 mg/L NAA ve 1.5 mg/L TDZ içeren Murashige & Skoog's (MS) besi yeri temel alınarak hazırlanmıştır. Her üç hormon denemesinde de altıncı alt kültürde ana eksplantlarda yeşil sürgün benzeri yapılar gözlenmiştir. Sürgün eksplantlarının çoğalma oranları (eksplant başına düşen kardeşlenme sayısı) SHM IX için 2x, SHM V için 3,5x ve NTM I ortamı için 6x olarak tespit edilmiştir. Ayrıca S. vomeracea türüne ait tohum eksplantlarıyla doğrudan organogenezde %46,5 rejenerasyon oranı, 0.25 mg/L TDZ ve 2 mg/L indol bütirik asit (IBA) ilaveli MS ortamında başarıyla elde edilmiştir. Öte yandan, tohum eksplantlarından indirekt organogenez yoluyla kallus indüksiyonu sağlanmıştır. Kallus oluşum oranı 0.25 mg/L TDZ ve 0.5 mg/L NAA içeren MS ortamında %3,33 oranında bulunmuştur. Yumru eksplantlarında ise, in vitro koşullarda dokuların kararması ve yumuşaması nedeniyle hem direkt hem de indirekt organogenez sonuçlarında başarısız olunmuştur. Bilgimiz dahilinde, bu çalışma, S. vomeracea'nın sürgün eksplantlarının direkt organogenez ve tohum eksplantlarının indirekt organogenez yoluyla kullanıldığı ilk çalışmadır ve sürgün çoğaltımı ile kallus oluşumu başarıyla gerçekleştirilmiştir. Bu sonuçlar, salep orkidesinin ticari üretimi ile genetik kaynaklarının kontrollü kullanımı ve korunması açısından büyük önem taşımaktadır. Bu araştırmadan elde edilen bulgulara dayalı olarak yapılacak gelecekteki iyileştirme çalışmaları, klonal bitki sayısı ve verim artışını sağlayacaktır.

Serapias vomeracea is one of the common salep orchids belonging to the Orchidaceae family. This species is commonly harvested for salep, utilized as a thickening agent in ice cream and traditional hot beverages. Nonetheless, unregulated harvesting and low multiplication rates, combined with the reliance of the species on mycorrhizal fungi due to seed endosperm absence, threaten its sustainability. This study aimed to propagate the S. vomeracea species under in vitro conditions using direct and indirect organogenesis techniques via tuber, shoot, and seed explants. The highest surface sterilization success of explants was achieved with 0.1% (w/v) HgCl2; 67% for tubers, 93.75% for shoots, and 66.7% for seed explants. Direct organogenesis of shoot explants in three different phytohormone trials exhibited success rates of 12.5% in SHM V medium, and 6.25% in both SHM IX and NTM I media, Murashige & Skoog's Medium (MS) supplemented with 0.5 mg/L thidiazuron (TDZ) + 0.5 mg/L naphthalene acetic acid (NAA), 1 mg/L NAA, and 1.5 mg/L TDZ, respectively. Green shoot-like structures were observed on the mother explants in each of the three hormone trials at the sixth subculture of the shoot explants. Their multiplication rates (sister plantlets per explant) were found to be 2x in SHM IX, 3.5x for SHM V, and 6x with NTM I media trials. Moreover, the seed explants of the S. vomeracea species were grown on MS medium supplemented with 0.25 mg/L TDZ and 2 mg/L indole butyric acid with 46.5% of regeneration rate with direct organogenesis. On the other hand, callus induction was achieved from seed explants with indirect organogenesis. Callus induction rate was assessed as 3.33% with MS medium supplemented with 0.25 mg/L TDZ and 0.5 mg/L NAA. Indirect and direct organogenesis results for tuber explants were unsuccessful due to darkening and softening of the tissues under in vitro conditions. To the best of our knowledge, this study was to first utilize the shoot explants of Serapias vomeracea for direct organogenesis and seed explants for indirect organogenesis, leading to successful in vitro propagation with shoot multiplication and callus induction. These results are of significant importance for the commercial production of the salep orchid and controlled utilization and conservation of its genetic resources. Future improvement studies based on the findings of this research may increase the number of regenerated plantlets, thereby enhancing yield.
