Özet
Fibromiyalji, yaygın kas ağrısı, yorgunluk ve uyku bozukluklarıyla seyreden kronik bir hastalıktır. Hastalığın tam nedeni bilinmemekle birlikte, oksidatif stres ve inflamasyonun önemli rol oynadığı düşünülmektedir. Oksidatif stres, hücre hasarına yol açarak semptomları artırabilir. Fibromiyalji hastalarında oksidatif stresin artması, inflamasyonu tetikleyerek semptomların şiddetlenmesine katkıda bulunabilir. Koenzim Q10 (CoQ10), mitokondrial enerji üretiminde görev alan güçlü bir antioksidandır ve fibromiyalji hastalarında eksikliği semptomları kötüleştirebilir. Bu nedenle CoQ10 takviyesinin, oksidatif stres ve inflamasyonu azaltarak semptomları hafifletebileceği öne sürülmektedir. Çalışmada 24 adet dişi Wistar sıçan kullanıldı. Sıçanlar; Kontrol grubu (n=6), Fibromiyalji grubu (n=6), Koenzim Q10 (n=6), Fibromiyalji+Koenzim Q10 grubu (n=6) olmak üzere 4 gruba ayrıldı. Kontrol grubuna çalışma boyunca herhangi bir uygulama yapılmadı. Fibromiyalji gruplarına cilt altından rezerpin 1 mg/kg/gün 3 gün uygulandı. Koenzim Q10 gruplarına rezerpin verilmeden 1 gün önce mısır yağında çözülmüş koenzim Q10 150 mg/kg oral olarak verildi. Çalışmanın 0, 4 ve 6. Günler de von Frey ve soğuk allodini testi uygulandı. Çalışmanın sonunda, sıçanlar anestezi altında sakrifiye edilerek trapezius kas dokusu örnekleri alındı. Alınan kas dokusunda MDA, GSH Seviyeleri ve SOD aktivitesi ELISA, NF-kB değeri ise RT-PCR ile analiz edildi. Bulgular doğrultusunda SOD, MDA ve GSH konsantrasyonları açısından gruplar arası istatistiksel olarak anlamlı fark bulunmadı (p>0,05). İnflamasyon belirteci olan NF-kB gruplar arası anlamlı fark bulundu (p<0,05). Von frey testinde grup içi ve gruplar arası anlamlı fark bulunurken soğuk allodini testinde grup içi ve gruplar arası anlamlı bir fark bulunmadı. Çalışmamız, CoQ10 takviyesinin inflamasyon ve oksidatif stres parametreleri üzerinde belirgin bir etkisinin olmadığını göstermektedir.

Fibromyalgia is a chronic condition characterised by widespread muscle pain, fatigue, and sleep disturbances. Although the exact cause of the condition is unknown, oxidative stress and inflammation are thought to play a significant role. Oxidative stress can exacerbate symptoms by causing cellular damage. Increased oxidative stress in fibromyalgia patients may contribute to worsening symptoms by triggering inflammation. Coenzyme Q10 (CoQ10) is a powerful antioxidant involved in mitochondrial energy production, and its deficiency in fibromyalgia patients may worsen symptoms. Therefore, it has been suggested that CoQ10 supplementation may alleviate symptoms by reducing oxidative stress and inflammation. In the study, 24 female Wistar rats were used. The rats were divided into 4 groups as Control group (n=6), Fibromyalgia group (n=6), Coenzyme Q10 (n=6), Fibromyalgia+Coenzyme Q10 group (n=6). The control group did not receive any treatment during the study. Fibromyalgia groups received subcutaneous reserpine 1 mg/kg/day for 3 days. Coenzyme Q10 groups received 150 mg/kg of coenzyme Q10 dissolved in corn oil orally 1 day before reserpine administration. On days 0, 4 and 6 of the study, von Frey and cold allodynia tests were performed. At the end of the study, rats were sacrificed under anaesthesia and trapezius muscle tissue samples were collected. MDA, GSH levels and SOD activity in the collected muscle tissue were analysed by ELISA, while the NF-kB value was analysed by RT-PCR. According to the findings, there was no statistically significant difference between the groups in terms of SOD, MDA, and GSH concentrations (p>0.05). A significant difference was found between the groups in NF-kB, an inflammation marker (p<0.05). Significant differences were found within and between groups in the von Frey test, while no significant differences were found within or between groups in the cold allodynia test. Our study demonstrates that CoQ10 supplementation has no significant effect on inflammation and oxidative stress parameters.
