Özet
Fotoakustik tomografi (FAT), optik uyarım ile ultrasonik algılamanın birbirini tamamlayan avantajlarını birleştiren, gelişmekte olan bir biyomedikal görüntüleme tekniğidir. Yüksek optik kontrastı, ultrason görüntülemenin sahip olduğu derin doku özelliklerini bir araya getiren FAT, biyolojik dokuların invaziv olmayan görüntülenmesine olanak tanımakta ve özellikle kanserin tespiti, tanısı ve tedavi sürecinin izlenmesinde büyük ölçüde potansiyel sergilemektedir. Yöntem, incelenen dokuya lazer ışını uygulanarak ısıtılmasına ve ısınma-soğuma işlemleri sonucu oluşan genleşme-sıkışma hareketi ile ses dalgası üretimi prensibi ile çalışmaktadır. Bu süreç sonucunda oluşan geniş bantlı akustik dalgalar ultrasonik dönüştürücüler tarafından algılanarak dokunun yüksek çözünürlüklü görüntülenmesi gerçekleştirilmektedir. FAT görüntüleme, yapısal, fonksiyonel ve moleküler seviyede bilgi sağlayabilmesine rağmen, elde edilen görüntü kalitesi ve tanısal doğruluk halen geleneksel görüntüleme yöntemleriyle tam anlamıyla karşılaştırılabilir düzeyde değildir. Bu nedenle yöntem, günümüzde yoğun bir şekilde geliştirme ve optimizasyon çalışmalarına konu olmaya devam etmektedir. FAT sistemlerinin duyarlılık ve özgüllüğünü artırmak amacıyla, özellikle nanoparçacıklar başta olmak üzere eksojen kontrast ajanlarının kullanımı artmıştır. Nanoparçacıklar, düşük sistemik toksisite, hedef dokularda seçici birikim ve boyut, yüzey kimyasının hassas bir biçimde kontrol edilebilmesiyle optik özelliklerin ayarlanabilmesi gibi önemli avantajlar sunmaktadır. Genişleyen nanoteknoloji ve nanotıp alanı doğrultusunda, altın nanoküreler biyouyumlulukları, görünür spektral bölgede güçlü optik soğurma özellikleri ve yüzey fonksiyonelleştirme olanakları sayesinde fotoakustik görüntüleme ve fototermal terapi uygulamaları için oldukça umut verici kontrast ajanları olarak öne çıkmaktadır. Bu bağlamda, söz konusu nanoparçacıkların ve kaplama ajanlarının fizikokimyasal özelliklerinin fotoakustik sinyal üretimi üzerindeki etkilerinin incelenmesi, biyomedikal uygulamalardaki etkinliklerinin artırılması açısından kritik önem taşımaktadır. Bu çalışmada, biyouyumlu ve toksik olmayan altın nanoküreler, fotoakustik tomografi için kontrast artırıcı ajanlar olarak incelenmiştir. Çapları 20 nm, 50 nm ve 80 nm olan nanoküreler içeren sulu çözeltiler hazırlanmış ve her bir boyut için 0,05 mg/mL ve 5 mg/mL optik yoğunluğa sahip örnekler polipropilen tüpler içerisinde ölçülüp değerlendirilmiştir. Yüzey özelliklerinin etkisini incelemek amacıyla, 50 nm çapındaki nanoküreler setiltrimetilamonyum bromür (CTAB) veya sitrat (CIT) ile kaplanmış, santrifüj işlemi sonrasında cam lamlar üzerine fikse edilmiştir. Numuneler, altın nanokürelerin yaklaşık olarak maksimum soğurma tepe noktalarına yakın olan 532 nm dalga boyunda çalışan bir lazer kaynağına sahip FAT sistemi kullanılarak fotoakustik sinyal işaretleri görüntülenmiştir. Deneysel düzenekten elde edilen fotoakustik sinyallerin yoğunluğu Matlab programında kodlama yardımı ile sayısallaştırılıp, parçacık boyutu, konsantrasyon ve kaplama ajanının sinyal genliği üzerindeki etkilerini belirlemek amacıyla grafiksel olarak analiz edilmiştir. Elde edilen bulgular, fotoakustik sinyal oluşumuna etki eden karmaşık mekanizmaların daha iyi anlaşılmasına katkı sağlamakta ve literatürdeki mevcut bilgi boşluklarını gidermeye yönelik önemli veriler sunmaktadır. Genel olarak bu çalışma, özellikle kanser araştırmaları ve nanotıp uygulamaları kapsamında, fotoakustik görüntüleme ile fototermal terapinin birlikte ele alındığı gelecekteki in vitro çalışmalara katkı sağlayacak bir zemin oluşturacaktır.

Photoacoustic tomography (PAT) is a rapidly advancing biomedical imaging modality that synergistically combines the strengths of optical excitation and ultrasonic detection to overcome many of the inherent limitations of conventional imaging techniques. By exploiting the high contrast associated with optical absorption and the superior depth penetration of ultrasound, PAT enables non invasive visualization of biological tissues with enhanced spatial resolution. This hybrid approach has attracted considerable attention in recent years, particularly for applications in oncology, where accurate detection, diagnosis, and monitoring of tumors remain critical clinical challenges. The fundamental operating principle of PAT is based on the photoacoustic effect. When short duration laser pulses irradiate biological tissue, optical energy is selectively absorbed by endogenous or exogenous chromophores. This absorption leads to a rapid but localized temperature rise, which in turn induces transient thermoelastic expansion. This process generates broadband ultrasonic waves that propagate through the tissue and can be detected by transducers integrated within the ultrasound probe positioned at the tissue surface. By collecting these acoustic signals from multiple spatial locations and applying appropriate reconstruction algorithms, high resolution tomographic images representing the optical absorption distribution within the tissue can be obtained. One of the major advantages of PAT is its ability to provide multi scale and multi contrast information. Depending on the excitation wavelength and detection configuration, PAT can reveal not only structural features but also functional parameters such as blood oxygenation, hemoglobin concentration, and vascular architecture. Furthermore, when combined with molecularly targeted contrast agents, PAT can achieve molecular level imaging, enabling visualization of specific biological processes associated with disease progression. Despite these capabilities, however, the image quality and diagnostic accuracy of PAT systems do not yet consistently match those of established imaging modalities such as magnetic resonance imaging (MRI), computed tomography (CT), or positron emission tomography (PET). As a result, significant research efforts continue to focus on improving system sensitivity, contrast, spatial resolution, and overall reliability. A major strategy for enhancing PAT performance involves the use of exogenous contrast agents. Among these, nanoparticles have emerged as particularly attractive candidates due to their unique physicochemical properties. Nanoparticles can be engineered to exhibit strong optical absorption at specific wavelengths, allowing efficient photoacoustic signal generation. Additionally, they often demonstrate low systemic toxicity, favorable biocompatibility, and the ability to selectively accumulate in target tissues through passive or active targeting mechanisms. Importantly, nanoparticle size, shape, composition, and surface chemistry can be precisely controlled during synthesis, enabling fine tuning of their optical and biological behavior. Within the broader fields of nanotechnology and nanomedicine, gold nanospheres have gained widespread recognition as promising contrast agents for photoacoustic imaging as well as for photothermal therapy. Gold nanoparticles exhibit strong localized surface plasmon resonance, resulting in intense optical absorption in the visible and near infrared spectral regions. This property makes them particularly effective at converting absorbed optical energy into localized thermal and acoustic responses. Moreover, gold nanospheres are generally considered biocompatible and chemically stable, and their surfaces can be readily functionalized with a wide range of biomolecules, polymers, or surfactants. Such surface modifications allow improved colloidal stability, enhanced targeting capability, and tailored interactions with biological environments. Given these advantages, understanding how the physicochemical characteristics of gold nanospheres influence photoacoustic signal generation is of critical importance. Parameters such as particle size, concentration, and surface coating can significantly affect optical absorption efficiency, heat transfer dynamics, and acoustic emission. However, despite the growing body of literature on nanoparticle enhanced photoacoustic imaging, systematic investigations addressing these factors remain relatively limited. Addressing these knowledge gaps is essential for optimizing nanoparticle design and maximizing their effectiveness in biomedical applications. In the present study, biocompatible and non toxic gold nanospheres were systematically evaluated as contrast enhancing agents for photoacoustic tomography. Aqueous solutions containing gold nanospheres with diameters of 20 nm, 50 nm, and 80 nm were used to assess the influence of particle size on photoacoustic signal generation. For each particle size, two different optical concentrations (0.05 mg/mL and 5 mg/mL) were investigated to examine concentration dependent effects. The nanoparticle solutions were placed inside polypropylene tubes for imaging. In addition to particle size and concentration, the role of surface chemistry was also examined. To this end, 50 nm gold nanospheres were coated with either namely the cationic cetyltrimethylammonium bromide (CTAB) and the anionic citrate (CIT), two commonly used surface stabilizing agents with distinct chemical and electrostatic properties were used, subsequently centrifuged, and fixed onto glass slides. This preparation allowed direct comparison of photoacoustic signal behavior associated with different surface coatings under controlled conditions. All samples were imaged using a photoacoustic tomography system equipped with a pulsed laser source operating at a wavelength of 532 nm. This wavelength was selected because it closely corresponds to the approximate maximum absorption peak of gold nanospheres, thereby ensuring efficient optical excitation and robust photoacoustic signal generation. The detected acoustic signals were captured by ultrasonic transducers and digitized for subsequent analysis. Evaluation of the photoacoustic signals was performed using MATLAB. Signal amplitudes were extracted, processed, visually and graphically analyzed to assess the effects of nanoparticle size, concentration, and surface coating on photoacoustic response. Comparative analysis revealed clear trends in signal intensity as a function of these parameters, providing insight into the underlying mechanisms governing photoacoustic signal formation in nanoparticle based systems. The results of this study contribute to a more comprehensive understanding of how gold nanosphere properties influence photoacoustic imaging performance. The systematic investigation of the effects of size, concentration, and surface chemistry enables comparison with previous studies and provides experimentally grounded data that can guide future nanoparticle design. Overall, this study establishes a foundational basis for future in vitro investigations that integrate photoacoustic imaging with photothermal therapy, particularly in the context of cancer research and nanomedicine applications.
