Özet

VPA, gebelikte maruz kalındığında nörogelişimsel bozukluklara ve otizm riskinin artmasına yol açabilen bir antiepileptiktir. OSB, sosyal iletişim güçlükleri ve yineleyici davranışlarla karakterizedir. Ergenlik dönemi ise hormonal değişimler nedeniyle OSB belirtilerinin şekil ve şiddetinin değişebildiği kritik bir evredir. VPA ile oluşturulan OSB hayvan modelleri, bu döneme özgü duyusal ve sosyal davranış farklılıklarını nesnel olarak değerlendirmeyi sağlar. Bu kapsamda çalışmamızda prenatal VPA maruziyetinin dişi sıçanlarda ergenlik dönemine özgü duyusal işleme ve sosyal davranışlar üzerindeki etkileri incelenmiştir. VPA ve kontrol grubu için ayrı ayrı 8 adet sıçan gebe bırakılmış gebeliğin 12.5 günü 500 mg/kg VPA uygulanmış, kontrol grubuna ise SF uygulanmıştır. Her gebe sıçandan 4 adet yavru alınmış ve n=32 VPA, n=32 kontrol grubu kullanılmıştır. Bu gruplar ise n=16, 25 günlük puberte öncesi ve n=16, 60 günlük puberte sonrası olmak üzere ikiye ayrılmıştır. Sıçanlara davranış değerlendirmesi için 25. ve 60. günlerde duyusal hassasiyeti ölçmek için akustik irkilme-habitüasyon, 28. ve 63. günlerde sosyal tercihi incelemek için sosyal etkileşim testi, 31. ve 66. günlerde keşif eğilimlerini incelemek için Y-Labirent testi uygulanmıştır. Habitüasyon yaş ve VPA maruziyeti faktörlerinden etkilenmeden tüm gruplarda gerçekleşmiştir. Sosyal etkileşim testinde puberte öncesi kontrol grubu (p=0,001) ve puburte sonrası kontrol grubunun (p= 0,015) yabancı sıçan tercihi daha yüksek bulunurken diğer gruplarda bu tercih görülmemiştir. Sosyal etkileşimde toplam frekansın VPA maruziyetinin bölmelere giriş frekansını düşürdüğü görülmüştür (p=0,02). Sosyal yenilik testinde bölmelere toplam giriş frekansı VPA (p=0,006) ve yaş durumundan (p=0,02) etkilenmiştir. VPA gruplarının ve puberte sonrası grupların daha düşük keşif eğilimine sahip olduğu görülmüştür. Y-Labirent testinde yeni kola giriş sayısının yaştan (p=0,014), VPA maruziyetinden (p=0,001) etkilendiği görülmüştür. VPA ve puberte durumu yeni kola giriş sayılarını azaltmıştır. Puberte öncesi gruplar puberte sonrası gruplara göre daha fazla alternasyon sağlamıştır (p=0,034). Puberte sonrası gruplar puberte öncesi gruplara göre daha fazla yeni kola giriş oranı göstermiştir (p=0,031). VPA gruplarında benzer şekilde yeni kola girme oranı azalmıştır (p=0,002). Sonuç olarak VPA maruziyeti habitüasyon yanıtlarını etkilememesine rağmen kısa süreli bellekte yaş bağımlı etki gösterdiği görülmüştür. Bu sonuçlar VPA modelininin yaşa göre etkilerinin azalabileceğini göstermektedir.

Valproic acid (VPA) is an antiepileptic drug that, when exposure occurs during pregnancy, can induce neurodevelopmental abnormalities and increase the risk of autism spectrum disorder (ASD). ASD is marked by impairments in social communication and repetitive behaviors, and adolescence represents a critical period wherein hormonal changes can alter symptom expression. VPA-induced ASD models allow objective evaluation of sensory and social behavioral alterations specific to this stage. In this study, we examined the effects of prenatal VPA exposure on sensory processing and social behavior in adolescent female rats. Eight pregnant rats per condition were used; dams received either 500 mg/kg VPA or saline on gestational day 12.5, yielding n=32 VPA-exposed and n=32 control offspring. These were divided into prepubertal (day 25) and postpubertal (day 60) groups (n=16 each). Behavioral assessments included acoustic startle–habituation, the social interaction/novelty tests, and the Y-maze test. Habituation occurred in all groups and was not influenced by age or VPA exposure. In the social interaction test, the prepubertal (p=0.001) and postpubertal (p=0.015) control groups displayed a significantly stronger preference for the unfamiliar rat, a preference not observed in the VPA groups. In terms of total exploration frequency, VPA exposure reduced the number of entries into the social compartments (p=0.02). In the social novelty test, the total compartment entry frequency was affected by both VPA exposure (p=0.006) and age (p=0.02), with VPA-exposed and postpubertal animals exhibiting reduced exploratory tendencies. In the Y-maze test, the number of entries into the novel arm was influenced by age (p=0.014) and VPA exposure (p=0.001). Both VPA exposure and puberty status reduced new arm enrollment numbers. Prepubertal groups exhibited a higher alternation rate compared to postpubertal groups (p=0.034), while postpubertal rats showed a higher proportion of novel arm entries than prepubertal rats (p=0.031). Similarly, VPA-exposed rats showed reduced novel arm entry ratios (p=0.002). Overall, while VPA exposure did not alter habituation responses, it exerted age-dependent effects on short-term memory. These findings suggest that the behavioral impact of the VPA model may diminish with age.
