Özet

Yara iyileşme sürecinde yara yatağını koruyan, cilt rejenerasyonunu destekleyen ve iyileşmeyi hızlandıran bir yara örtüsüne ihtiyaç duyulur. Bu yara örtülerinin yara iyileşmesini destekleyecek şekilde yarayı nemli tutması, oksijen geçirgenliğini sağlaması, ciltle biyouyumlu olması ve enfeksiyon oluşumunu engellemek için antibakteriyel olması gerekmektedir. Maliyet açısından ucuz olan ve kolay erişilebilen, bu nedenle günümüzde de hala yaygın olarak kullanılan geleneksel yara örtüleri, çeşitli dezavantajları ve artan ihtiyacı karşılamaktaki yetersizlikleri sebebiyle, yeni modern yara örtülerinin geliştirilmesi için yapılan çalışmalar hız kazanmıştır. Biyouyumlu ve biyobozunur yapıları, antimikrobiyal etkinlikleri, doku rejenerasyonunu teşvik etmeleri ve kontrollü ilaç salınımına olanak tanıyan özelleştirilebilir mekanik özellikleri sayesinde hidrojeller, modern yara örtüsü uygulamalarında yaygın olarak tercih edilmektedir. Yaklaşık %90 oranında su içeren bu yüksek hidratlı yapılar, yara yatağında ideal nem dengesini koruyarak iyileşme sürecini belirgin ölçüde hızlandırmaktadır. Hidrojellerin biyofonksiyonel kapasitesini artırmak amacıyla yapıya peptit entegrasyonu sağlanması ve bu doğrultuda peptit-bazlı kompozit hidrojellerin geliştirilmesi, güncel literatürde stratejik bir araştırma alanı haline gelmiştir. Bu tür hibrit sistemlerde peptitler; hücresel yapışmanın (adhezyon) teşvik edilmesi, mekanik özelliklerin modülasyonu veya matrise antimikrobiyal işlevsellik kazandırılması gibi kritik roller üstlenmektedir. Biyofonksiyonel özelliklerin yanı sıra, bu hidrojellerin hastanın spesifik ihtiyaçlarına göre kişiselleştirilmesi süreci ivme kazanmış ve bu amaçla 3B yazıcı teknolojileri temel üretim aracı haline gelmiştir. Özellikle Dijital Işık İşleme (DLP) tekniği; yüksek baskı hızı, mikro ölçekteki çözünürlük hassasiyeti ve karmaşık geometrileri aslına uygun şekilde üretme kabiliyeti ile bu alandaki en kritik yöntemlerden biri olarak öne çıkmaktadır. Söz konusu teknolojiyle, hidrojel sisteminin mekanik özelliklerinin ve geometrik tasarımının, baskı parametreleri üzerinden kişiye ve yara tipine özgü olarak modüle edilmesi amaçlanmıştır. Bu sayede, farklı boyut ve morfolojilere sahip lezyonlar için vücudun çeşitli anatomik bölgelerine uyumlu, özelleştirilmiş yara örtülerinin üretilmesi hedeflenmektedir. Bu doğrultuda, katı faz peptit sentezi (SPPS) yöntemi ile sentezlenen peptit, kendiliğinden birleşme özelliği sayesinde hidrojel matrisi oluşturmak için kullanılmıştır. Sentezlenen peptidin HPLC metodu ile saflaştırılmasının ardından peptidin yapısı LC-MS analizi ile doğrulanmıştır. Ayrıca 3B yazıcıda basılabilmesi için gerekli olan akrilat ucundan yararlanılan ve peptit ile kompozit yapıldığında hidrojelin mekanik dayanımını artıran GelMA sentezlenmiştir. Hazırlanan solüsyon, hücre dostu ortamların oluşturulmasına imkan sağlayan ve hassas yapılar üretilebilen 3B DLP teknolojili yazıcıda basıma uygun hale getirmek için koşullar ve parametreler optimize edilmiş, sonrasında fotobaşlatıcı ilavesiyle beraber UV ışını ile basılmıştır. Baskı sonrası elde edilen hidrojeller, Taramalı Elektron Mikroskobu (SEM) ve Fourier Dönüşümlü Kızılötesi Spektroskopisi (FTIR) ile karakterize edilmiştir. Elde ettiğimiz sonuçlar, peptit/GelMA kompozit hidrojeli kullanılarak geliştirilen formülasyon, biyouyumlu ve antimikrobiyal özelliğiyle yara iyileştirmesini destekleyici ve hızlandırıcı etkinliğe sahip olduğunu göstermektedir. Malzemenin sahip olduğu bu fonksiyonel özellikler, biyobasım teknolojisiyle birleştiğinde, yara tedavisinde yenilikçi ve kişiye özel yara örtülerinin geliştirilmesine olanak sağlayacak güçlü bir potansiyel ortaya koymaktadır.

The wound healing process requires a wound dressing that protects the wound bed, supports skin regeneration, and accelerates the healing. In this context, the effectiveness of the healing process is strongly dependent on the ability of the wound dressing to provide a suitable microenvironment at the wound interface. Such wound dressings should maintain a moist environment, allow oxygen permeability, be biocompatible with the skin, and exhibit antibacterial properties to prevent infection. Additionally, they are expected to minimize pain during application and removal, reduce scar formation, and support cellular activities such as proliferation and migration. Traditional wound dressings, which are inexpensive, easily accessible, and therefore still widely used today, have various disadvantages and are insufficient to meet the increasing demands of modern wound care. These materials generally act as passive protective barriers and lack the ability to actively contribute to or enhance the healing process. Conventional materials such as gauze and bandages, while cost-effective, fail to maintain adequate moisture levels within the wound environment. Moreover, they are prone to adhering to the wound surface, which can cause secondary tissue damage and pain during removal. Furthermore, they offer no inherent antimicrobial activity, leaving the wound susceptible to infection. This has led to accelerated research into the development of new modern wound dressings. Hydrogels are widely preferred in modern wound dressing applications due to their biocompatible and biodegradable structures, antimicrobial activity, ability to promote tissue regeneration, and customizable mechanical properties that allow for controlled drug release. Furthermore, their structural similarity to the natural extracellular matrix provides a favorable environment for cell proliferation. These highly hydrated structures, containing approximately 90% water, maintain an ideal moisture balance within the wound bed, thereby significantly accelerating the healing process. In addition, their soft and flexible nature enables them to adapt to irregular wound geometries, improving contact with the wound surface and enhancing patient comfort. To enhance the biofunctional capacity of hydrogels, the integration of peptides into the structure and the development of peptide-based composite hydrogels have increasingly become a strategic research area in the current literature. This approach allows for the effective combination of structural support with biological functionality within a single system. In such types of hybrid systems, peptides play critical roles such as promoting cellular adhesion, modulating mechanical properties, or imparting antimicrobial functionality to the matrix. In particular, antimicrobial peptides (AMPs) can be incorporated into the hydrogel structure to provide inherent antimicrobial activity by disrupting microbial cell membranes and preventing infection at the wound site. Moreover, certain peptides can undergo self-assembly, contributing to the formation of stable and organized nanostructures within the hydrogel network. In addition to biofunctional properties, the process of personalizing these hydrogels according to the specific needs of the patient has gained momentum, and three-dimensional (3D) printing technologies have become the primary production tool for this purpose. The application of 3D printing technologies enables the fabrication of patient-specific structures with high precision and reproducibility. In particular, Digital Light Processing (DLP) technology stands out as one of the most critical methods in this field due to its high printing speed, micro-scale resolution accuracy, and ability to accurately reproduce complex geometries, making it highly suitable for wound dressing applications. With this technology, the aim is to modulate the mechanical properties and geometric design of the hydrogel system according to the individual and wound type via printing parameters. In this way, the aim is to produce customized wound dressings compatible with various anatomical regions of the body for lesions with different sizes and morphologies. In this context, the peptide synthesized via the solid-phase peptide synthesis (SPPS) method was used to form a hydrogel matrix due to its self-assembly property. After purification of the synthesized peptide by High-Performance Liquid Chromatography (HPLC), its molecular weight and chemical structure were confirmed by Liquid Chromatography-Mass Spectrometry (LC-MS) analysis. Additionally, Gelatin Methacrylate (GelMA), which utilizes acrylate functional groups necessary for photopolymerization during 3D printing and increases the mechanical strength of the hydrogel when composited with the peptide, was synthesized, thereby providing enhanced structural stability and improved printability of the resulting hydrogel system. The prepared solution was optimized for printing on a 3D DLP printer, which allows for the creation of cell-friendly environments and the production of precise structures, and then printed with ultraviolet (UV) light after the addition of a photoinitiator. During this process, photo-crosslinking reactions occur, leading to the formation of a stable three-dimensional network. The hydrogels obtained after printing were characterized by Scanning Electron Microscopy (SEM) and Fourier Transform Infrared Spectroscopy (FTIR), enabling the evaluation of their morphological features and chemical structure. Our results indicate that the formulation developed using the peptide/GelMA composite hydrogel has biocompatible and antimicrobial properties, and has an activity that supports and accelerates wound healing. These findings suggest that the developed material successfully meets the fundamental requirements of an effective wound dressing. When combined with bioprinting technology, these functional properties of the material demonstrate a strong potential for the development of innovative and personalized wound dressings for wound treatment.
