Özet

Hücre mekanobiyolojisi; hücrelerin mekanik sinyalleri algılama ve bunlara yanıt verme yeteneğini inceleyerek prolıferasyon (çoğalma), adhezyon (yapışma), migrasyon (göç) ve motilite (hareketlilik) gibi temel hücresel süreçlerin moleküler düzeyde nasıl düzenlendiğini ortaya koymaktadır. Bu bağlamda, hücre içi sertlik (stiffness) dağılımının yüksek hassasiyetle haritalanması, hücresel homeostazın ve patolojik değişimlerin anlaşılması için elzem bir parametre haline gelmiştir. Özellikle kanserin yayılması, malign transformasyon ve metastatik potansiyelin belirlenmesinde, hücrenin mekanik sertliğindeki değişimler biyobelirteç niteliği taşımaktadır. Günümüzde hücre sertliği ölçümlerinde Atomik Kuvvet Mikroskobu (AFM) ve optik cımbızlama (optical trapping) teknikleri“altın standart”olarak kabul edilse de; bu yöntemlerin düşük örnekleme hızı, temas noktası belirlemedeki belirsizlikler, hücre üzerinde yarattıkları mekanik stres ve yüksek hacimli (high-throughput) analizlere izin vermeyen doğası, klinik ve geniş çaplı araştırmalarda kullanımlarını kısıtlamaktadır. Bu çalışma kapsamında, mevcut yöntemlerin kısıtlamalarını aşmak amacıyla; akustik uyarım (acoustic stimulation) ve dijital holografik görüntüleme (holographic imaging) prensiplerine dayanan; hızlı, invaziv olmayan, etiketsiz (label-free) ve yüksek çözünürlüklü yeni bir teknik sunulmaktadır. Geliştirilen bu akusto-holografik yöntem, tek hücre düzeyinde elastisite modülü ve sertlik dağılımını üç boyutlu olarak yeniden yapılandırabilmektedir. Deneysel validasyon sürecinde, standart HCT116 kolon kanseri hücre hattı ile metastatik potansiyeli temsil eden CTC-mimicking (dolaşımdaki tümör hücresini taklit eden) HCT116 hücrelerinin sertlik profilleri çıkarılmış ve bu iki grup arasındaki biyomekanik farklılıklar başarıyla ayırt edilmiştir. Elde edilen bulgular, bu sistemin tüm hücre sertliği ölçümlerini saniyeler mertebesinde gerçekleştirebildiğini ve yüksek doğruluk sunduğunu göstermektedir. Bu teknoloji; kanser biyolojisi araştırmaları, hastalık modelleme süreçleri, farmakolojik ilaç taramaları ve rutin klinik teşhis uygulamalarında mekanobiyolojik verinin kullanımını kolaylaştırma potansiyeline sahiptir. Sonuç olarak, bu tez çalışmasında sunulan yöntem, biyomedikal mühendisliği ve onkoloji alanında hücre mekaniğinin analizine yönelik düşük maliyetli ve yüksek verimli bir alternatif sunmaktadır.

Cell mechanobiology investigates the ability of cells to detect mechanical signals and respond to them, revealing how fundamental cellular processes such as proliferation, adhesion, migration, and motility are regulated at the molecular level. In this context, high-precision mapping of intracellular stiffness distribution has become an essential parameter for understanding cellular homeostasis and pathological changes. Changes in cellular mechanical stiffness serve as biomarkers, particularly in determining cancer spread, malignant transformation, and metastatic potential. Although Atomic Force Microscopy (AFM) and optical trapping techniques are currently accepted as the 'gold standard' for measuring cell stiffness, the low sampling rate of these methods, uncertainties in determining the contact point, the mechanical stress they create on the cell, and their nature that does not allow high-throughput analysis, limit their use in clinical and large-scale research. Within the scope of this study, in order to overcome the limitations of existing methods, a new technique is presented that is fast, non-invasive, label-free, and high-resolution, based on the principles of acoustic stimulation and holographic imaging. This developed acousto-holographic method can reconstruct the elasticity modulus and stiffness distribution at the single-cell level in three dimensions. During the experimental validation process, the stiffness profiles of the standard HCT116 colon cancer cell line and CTC-mimicking HCT116 cells representing metastatic potential were extracted, and the biomechanical differences between these two groups were successfully distinguished. The findings demonstrate that this system can perform all cell stiffness measurements in a matter of seconds and offers high accuracy. This technology has the potential to facilitate the use of mechanobiological data in cancer biology research, disease modelling processes, pharmacological drug screening, and routine clinical diagnostic applications. In conclusion, the method presented in this thesis offers a low-cost and highly efficient alternative for the analysis of cell mechanics in the fields of biomedical engineering and oncology.
