SELÇUK ÜNİVERSİTESİ SAĞLIK BİLİMLERİ ENSTİTÜSÜ Flonicamid İnsektisitin Genotoksik Etkilerinin İncelenmesi Durdane Yıldırım Kimyasal, Biyolojik, Radyolojik ve Nükleer Savunma Anabilim Dalı YÜKSEK LİSANS TEZİ / KONYA-2026 Flonicamid, tarımsal üretimde özellikle yaprak biti, beyazsinek ve trips gibi emici ağız yapısına sahip zararlılarla mücadelede yaygın olarak kullanılan sistemik özellikte bir insektisittir. Tarımsal verimliliğin artırılması ve ürün kayıplarının azaltılması açısından önemli avantajlar sağlamasına rağmen, pestisitlerin yoğun ve bilinçsiz kullanımı çevre ve insan sağlığı açısından çeşitli riskleri de beraberinde getirmektedir. Son yıllarda pestisit kalıntılarının toprak, su ve gıda zinciri yoluyla canlı organizmalara ulaşabilmesi; bu maddelerin yalnızca hedef zararlılar üzerindeki etkilerinin değil, aynı zamanda genetik materyal üzerindeki potansiyel toksik etkilerinin de araştırılmasını gerekli hale getirmiştir. Özellikle genotoksik etkiye sahip kimyasalların DNA hasarı, kromozomal bozukluklar, mutasyonlar ve hücresel düzensizliklere yol açabilmesi nedeniyle bu tür bileşiklerin güvenilirlik değerlendirmeleri büyük önem taşımaktadır. Bu çalışmada, flonicamid insektisitinin olası genotoksik etkilerinin belirlenmesi amacıyla bakteriyel geri mutasyon testi olan Ames testi ile bitki bazlı biyogöstergeler arasında yaygın olarak kullanılan Allium cepa testi uygulanmıştır. Ames testi kapsamında farklı Salmonella typhimurium TA98, TA100 ve TA102 suşları kullanılarak flonicamid'in mutajenik potansiyeli değerlendirilmiş, farklı konsantrasyonlarının geriye dönen koloni sayıları üzerindeki etkileri incelenmiştir. Elde edilen bulgulara göre, flonicamid'in yalnızca TA98 suşunda en yüksek konsantrasyonda geriye dönen koloni sayısında 2 kattan fazla artışa neden olduğu belirlenmiştir. Allium cepa L. kök ucu test sistemi kullanılarak kök uzunluğu, mitotik indeks ve kromozomal anomaliler değerlendirilmiş ve flonicamid'in sitotoksik ve genotoksik etkileri incelenmiştir. Soğan kökleri 125, 250 ve 500 ppm konsantrasyonlarındaki flonicamid'e 24, 48 ve 72 saatlik sürelerle maruz bırakılmıştır. Sitotoksisite göstergesi olan Mitotik İndeks (Mİ) değerlerinde konsantrasyon artışına paralel olarak dalgalanmalar gözlenmiş, kök uzunluğu testinde ise artan dozla birlikte büyümenin baskılandığı ve sitotoksisitenin arttığı (%66.14'e varan) belirlenmiştir. Çalışma sonucunda yapılan istatistiksel analizler flonicamid'in toplam kromozom aberasyonu (CA) üzerinde konsantrasyon bağımlı etki gösterdiğini (p < 0.05) ortaya koymuştur. Özellikle 500 ppm konsantrasyonunda c-metafaz, köprü, poliploidi ve c-anafaz gibi anomalilerin frekansında kontrol grubuna oranla önemli artışlar kaydedilmiştir. Bu artışların tespit edilmesi, flonicamid'in hücresel düzeyde genetik materyal üzerinde olumsuz etkiler oluşturabileceğini göstermektedir. Sonuç olarak flonicamid'in önerilen uygulama dozlarında (75-100ppm) ames ve Allium cepa testinde genotoksik potansiyelinin düşük olduğu, ancak doz aşımı durumundaki toksik olmayan yüksek dozlarda genotoksik potansiyel taşıdığı belirlenmiştir. Tarımsal üretimde yaygın olarak kullanılan pestisitlerin genetik toksisite potansiyelinin belirlenmesi; çevre sağlığının korunması, sürdürülebilir tarım uygulamalarının geliştirilmesi ve insan sağlığı açısından oluşabilecek risklerin önceden değerlendirilmesi bakımından büyük önem taşımaktadır. Elde edilen bulguların, pestisit kullanımına yönelik güvenlik politikalarının geliştirilmesine, kontrollü kullanım stratejilerinin oluşturulmasına ve çevresel risk analizlerine katkı sağlayacağı düşünülmektedir. Ayrıca çalışmanın gelecekte gerçekleştirilecek toksikolojik ve genotoksikolojik araştırmalara kaynak oluşturması ve benzer pestisitlerin biyolojik etkilerinin değerlendirilmesine ışık tutması amaçlanmaktadır.

SUMMARY REPUBLIC OF TÜRKİYE SELÇUK UNIVERSITY HEALTH SCIENCES INSTITUTE Investıgatıon of The Genotoxıc Effects of Flonıcamıd Insectıcıde Durdane Yıldırım Department of Chemical, Biological, Radiological and Nuclear Defence MASTER THESIS / KONYA-2026 Flonicamid is a systemic insecticide widely used in agricultural production, particularly for controlling pests with sucking mouthparts such as aphids, whiteflies, and thrips. Although it offers significant advantages in terms of increasing agricultural productivity and reducing crop losses, the intensive and indiscriminate use of pesticides also poses various risks to the environment and human health. In recent years, the ability of pesticide residues to reach living organisms through soil, water, and the food chain has made it necessary to investigate not only the effects of these substances on target pests but also their potential toxic effects on genetic material. Given that chemicals with genotoxic effects can cause DNA damage, chromosomal abnormalities, mutations, and cellular dysregulation, safety assessments of such compounds are of great importance. In this study, the Ames test—a bacterial reverse mutation assay—and the Allium cepa test, a plant-based bioassay widely used for this purpose, were employed to determine the potential genotoxic effects of the insecticide flonicamid. Within the scope of the Ames test, the mutagenic potential of flonicamid was evaluated using different Salmonella typhimurium strains (TA98, TA100, and TA102), and the effects of different concentrations on the number of revertant colonies were examined. According to the findings, it was determined that flonicamid caused an increase of more than twofold in the number of revertant colonies only in the TA98 strain at the highest concentration. Root length, mitotic index, and chromosomal abnormalities were evaluated using the Allium cepa L. root tip test system, and the cytotoxic and genotoxic effects of flonicamid were investigated. Onion roots were exposed to flonicamid at concentrations of 125, 250, and 500 ppm for durations of 24, 48, and 72 hours. Fluctuations were observed in the Mitotic Index (MI) values—a marker of cytotoxicity—in parallel with increasing concentrations; in the root length test, it was determined that growth was inhibited and cytotoxicity increased (up to 66.14%) with increasing doses. Statistical analyses conducted following the study revealed that flonicamid exhibited a concentration-dependent effect on total chromosome aberration (CA) (p < 0.05). In particular, at a concentration of 500 ppm, significant increases in the frequency of abnormalities such as c-metaphase, bridges, polyploidy, and c-anaphase were recorded compared to the control group. The detection of these increases suggests that flonicamid may cause adverse effects on genetic material at the cellular level. Consequently, it was determined that flonicamid exhibits low genotoxic potential at recommended application doses (75–100 ppm) in the Ames and Allium cepa tests; however, it possesses genotoxic potential at non-toxic high doses in cases of overdose. Determining the genetic toxicity potential of pesticides widely used in agricultural production is of great importance for protecting environmental health, developing sustainable agricultural practices, and assessing potential risks to human health in advance. It is anticipated that the findings will contribute to the development of safety policies regarding pesticide use, the formulation of controlled use strategies, and environmental risk analyses. Additionally, the study aims to serve as a resource for future toxicological and genotoxicological research and to shed light on the evaluation of the biological effects of similar pesticides.
