İNCİ KEFALİNİN [ALBURNUS TARICHI (GULDENSTAEDTII, 1814)] GÖZ, SOLUNGAÇ VE DERİ DOKULARINDA ÜREME GÖÇÜ SONRASI OLUŞABİLECEK DEĞİŞİMLERİN MAKROSKOBİK, HİSTOLOJİK VE TARAMALI ELEKTRON MİKROSKOBİK OLARAK İNCELENMESİ Yıldırım ÇELİK Erciyes Üniversitesi, Sağlık Bilimleri Enstitüsü Veterinerlik Anatomisi Anabilim Dalı Doktora Tezi, Haziran 2026 Danışman: Prof. Dr. Aydın ALAN ÖZET Dünyanın en büyük sodalı gölü olan Van Gölü'nde yayılış gösteren endemik inci kefali (Alburnus tarichi), üreme döneminde yüksek alkaliteye sahip göl ortamından tatlı su karakterindeki akarsulara (Bendimahi Çayı) anadrom bir göç gerçekleştirmektedir. Bu süreçte balığın maruz kaldığı ekstrem ozmotik değişimler ve çevresel stres faktörleri, dış ortamla doğrudan temas halinde olan dokularda önemli adaptif yanıtlar meydana getirmektedir. Çalışmamızda göl ve akarsudan cinsiyet ayırımı yapılmadan 60'ar adet ve toplamda 120 adet cinsel olgunluğa ulaşmış (+3 yaş) İnci kefalleri incelendi. Morfometrik ölçümler, histolojik incelemeler için dokular Crosman'ın üçlü boyaması ile boyandı ve Taramalı Elektron Mikroskop (SEM) ile yüzey incelemeleri yapıldı. Göl ve akarsu habitatlarındaki bireylerin karşılaştırmalı morfometrik analizinde, total ağırlığın göl örneklerinde (111,23 ± 15,71 g) akarsu örneklerine (100,06 ± 19,77 g) oranla istatistiksel olarak anlamlı derecede yüksek olduğu saptandı (P < 0,05). Akarsu örneklerindeki bu ağırlık kaybı; çevresel stres faktörleri, göç sürecindeki yüksek enerji tüketimi ve beslenmenin durmasıyla ilişkilendirildi. Bulbus oculi'nin histolojik değerlendirmesinde, akarsu habitatına geçişle birlikte cornea epitel kalınlığında (129,87 ± 26,61 µm) ve stroma kalınlığında (161,53 ± 46,26 µm) göl örneklerine kıyasla istatistiksel olarak anlamlı bir artış gözlendi (P < 0,05). Bu kalınlaşma, tatlı suyun düşük ozmotik basıncına karşı geliştirilen bir koruma mekanizması olarak değerlendirildi. SEM incelemeleri, akarsu örneklerinde, cornea yüzeyinde hücre yenilenmesine bağlı deformasyonlar ve epitel katmanında hiperplazi bulgularını ortaya koydu. Solungaç dokusunda, tatlı su ortamındaki bireylerin serbest sekonder lamel uzunluklarında (SSL UZ) kısalma (152,42 ± 37,20 µm) ve interlameller hücre kitlesinde (ILCM) belirgin bir artış (57,92 ± 14,25 µm) tespit edildi (P < 0,05). ILCM miktarındaki bu artışın, oksijen yönünden zengin olan akarsu ortamında solunum yüzey alanını azaltarak ozmotik yükü dengelemeye yönelik bir adaptasyon olduğu düşünüldü. Ayrıca akarsu örneklerinde lameller füsyon ve epitel kaldırma gibi stres kaynaklı histopatolojik değişimler gözlenirken, anevrizma bulgusuna sadece göl örneklerinde (%10) rastlandı. Deri dokusu incelemelerinde; göl örneklerinde hem epidermis (207,30 ± 27,99 µm) hem de hipodermis (136,38 ± 33,39 µm) tabakalarının akarsu örneklerine oranla anlamlı derecede kalın olduğu belirlendi (P < 0,05). Göl örneklerinin epidermal kalınlığının fazla olması, Van Gölü'nün yüksek ozmotik yoğunluğuna karşı bir bariyer görevi görmesi ve derinin solunum için ek bir organ olması ile ilişkilendirildi; akarsu örneklerindeki hipodermis tabakasında saptanan incelme ise göç sırasında enerji deposu olan yağ dokusunun tüketilmesine bağlandı. Çalışma verileri, inci kefalinin habitat geçişi sırasında ozmotik dengeyi korumak ve değişen çevresel parametrelere uyum sağlamak amacıyla göz, solungaç ve deri dokularında hücresel düzeyde kapsamlı modifikasyonlar gerçekleştirdiğini bilimsel olarak kanıtlandı. Özellikle SEM analizleri ile desteklenen yüzeysel morfolojik değişimler, deformasyonlar ve yenilenme bulguları, türün ekstrem koşullara adaptasyonunu kanıtlar nitelikteydi.

MACROSCOPIC, HISTOLOGICAL, AND SCANNING ELECTRON MICROSCOPIC INVESTIGATION OF POSSIBLE POST-SPAWNING MIGRATION CHANGES IN THE EYE, GILL, AND SKIN TISSUES OF PEARL MULLET [ALBURNUS TARICHI (GÜLDENSTÄDT, 1814)] Yıldırım ÇELİK Erciyes University, Institute of Healthy Sciences Department of Veterinary Anatomy PhD. Thesis, June 2026 Supervisor: Prof. Dr. Aydın ALAN ABSTRACT The endemic Pearl Mullet (Alburnus tarichi), which is distributed in Lake Van the largest soda lake in the World performs an anadromous migration from the highly alkaline lake environment to freshwater streams (Bendimahi Creek) during its reproductive period. The extreme osmotic changes and environmental stress factors to which the fish is exposed during this process induce significant adaptive responses in tissues that are in direct contact with the external environment. In our study, a total of 120 sexually mature Pearl Mullets (aged three years and older) were examined, with 60 specimens collected from the lake and 60 from the stream, without gender differentiation. Morphometric measurements were recorded, tissues were stained with Crossmon's trichrome for histological analysis, and surface examinations were performed using Scanning Electron Microscopy (SEM). In the comparative morphometric analysis of individuals from the lake and stream habitats, the total weight was found to be statistically significantly higher in the lake specimens (111.23 ± 15.71 g) compared to the stream specimens (100.06 ± 19.77 g) (P < 0.05). This weight loss observed in the stream specimens is associated with environmental stress factors, high energy consumption during the migration process, and the cessation of feeding. In the histological evaluation of the bulbus oculi, a statistically significant increase was observed in corneal epithelial thickness (129.87 ± 26.61 µm) and stromal thickness (161.53 ± 46.26 µm) upon transition to the stream habitat compared to the lake specimens (P < 0.05). This thickening is evaluated as a protective mechanism developed against the low osmotic pressure of freshwater. SEM examinations revealed deformations associated with cell renewal on the corneal surface and findings of hyperplasia in the epithelial layer of the stream specimens. In the gill tissue, a shortening in the free secondary lamellar length (FSLL) (152.42 ± 37.20 µm) and a distinct increase in the interlamellar cell mass (ILCM) (57.92 ± 14.25 µm) were detected in individuals from the freshwater environment (P < 0.05). This increase in the amount of ILCM is considered to be an adaptation aimed at balancing the osmotic load by reducing the respiratory surface area in the oxygen-rich stream environment. Furthermore, while stress-induced histopathological changes such as lamellar fusion and epithelial lifting were observed in the stream specimens, the finding of aneurysm was encountered exclusively in the lake specimens (10%). In the examinations of the skin tissue, both the epidermis (207.30 ± 27.99 µm) and hypodermis (136.38 ± 33.39 µm) layers were determined to be significantly thicker in the lake specimens compared to the stream specimens (P < 0.05). The greater epidermal thickness of the lake specimens was associated with its function as a barrier against the high osmotic density of Lake Van and the skin acting as an auxiliary organ for respiration; meanwhile, the thinning detected in the hypodermis layer of the stream specimens is attributed to the consumption of adipose tissue, which serves as an energy reserve during migration. The study data scientifically demonstrate that the pearl mullet undergoes comprehensive modifications at the cellular level in the eye, gill, and skin tissues to maintain osmotic balance and adapt to changing environmental parameters during habitat transition. In particular, the superficial morphological changes, deformations, and regeneration findings supported by SEM analyses confirm the adaptation of the species to extreme conditions.
