Özet
Fototermal terapi, kanser tedavisinde minimal invaziv bir strateji olarak büyük ilgi görmüştür; ancak terapötik etkinliği, ısı şoku proteinleri tarafından düzenlenen hücresel termotolerans gelişimi nedeniyle kısıtlanabilmektedir. Bu tez çalışmasında, 4T1 meme kanseri modelinde, ısı şoku proteini susturması yoluyla fototermal tedaviyi güçlendirmek amacıyla polidopamin (PDA) kaplı MXene tabanlı bir sistem kombinasyonel bir platform olarak araştırılmıştır. Çalışma, hem nanokompozit ile siRNA arasındaki fizikokimyasal etkileşimi hem de gen susturma ile MXene destekli fototermal tedaviyi birleştirmenin terapötik potansiyelini değerlendirmek üzere tasarlanmıştır. HSP90AA1 hedefli siRNA adayları taranmış ve en etkili sekans RT-qPCR ile belirlenmiştir. Hücresel alım ve hücre içi dağılım konfokal mikroskopi ve akım sitometrisi ile analiz edilmiştir. Tek başına MXene-PDA'nın ve gen susturma ile kombinasyonunun biyolojik etkileri, ışık uygulanan ve uygulanmayan koşullar altında LDH tabanlı sitotoksisite analizleri kullanılarak değerlendirilmiştir. Bulgular, nanokompozitin siRNA ile etkileşime girebildiğini ve umut vadeden bir fototermal bileşen olarak işlev gördüğünü göstermiştir. Nanoplatform tabanlı eş iletim yaklaşımı test edilen koşullar altında sınırlı bir performans gösterirken, etkili HSP90AA1 susturulmasının ardından uygulanan MXene-PDA destekli ışık uygulamasından oluşan ardışık kombinasyon daha elverişli terapötik sonuçlar vermiştir. Genel olarak sonuçlar, ısı şoku proteini susturulması ile MXene tabanlı fototermal tedavinin birleştirilmesinin kanser tedavisi için bilimsel olarak rasyonel ve potansiyel olarak etkili bir strateji olduğunu göstermektedir. Bununla birlikte, önerilen nanoplatformun siRNA iletim performansını iyileştirmek için daha fazla optimizasyona ihtiyaç duyulmaktadır.

Photothermal therapy has attracted considerable attention as a minimally invasive strategy for cancer treatment; however, its therapeutic efficacy may be limited by the development of cellular thermotolerance regulated by heat shock proteins. In this thesis, a polydopamine (PDA)-coated MXene-based system was investigated as a combinatorial platform to enhance photothermal therapy through heat shock protein silencing in the 4T1 breast cancer model. The study was designed to evaluate both the physicochemical interaction between the nanocomposite and siRNA and the therapeutic potential of combining gene silencing with MXene-assisted photothermal therapy. HSP90AA1-targeting siRNA candidates were screened, and the most effective sequence was identified by RT-qPCR. Cellular uptake and intracellular distribution were analyzed by confocal microscopy and flow cytometry. The biological effects of MXene-PDA alone and in combination with gene silencing were evaluated under irradiated and non-irradiated conditions using LDH-based cytotoxicity analyses. The findings demonstrated that the nanocomposite was capable of interacting with siRNA and functioned as a promising photothermal component. While the nanoplatform-based co-delivery approach showed limited performance under the tested conditions, the sequential combination consisting of effective HSP90AA1 silencing followed by MXene-PDA-mediated irradiation yielded more favorable therapeutic outcomes. Overall, the results indicate that combining heat shock protein silencing with MXene-based photothermal therapy represents a scientifically rational and potentially effective strategy for cancer treatment. Nevertheless, further optimization is required to improve the siRNA delivery performance of the proposed nanoplatform.
