Özet

Günümüzde yenilenebilir enerji üretimi ve yüksek değerli biyokütle geri kazanımını tek süreçte bütünleştiren entegre yaklaşımlara olan ihtiyacı giderek artmaktadır. Mikroalgler, yüksek ve çok yönlü biyokütle üretim kapasiteleriyle bu yaklaşımın merkezinde yer almaktadır. Bununla birlikte, monokültür sistemlerin çevresel kırılganlığı, farklı atık kaynakları türlerinin karşılaştırmalı değerlendirmesindeki yetersizlik ve çevresel koşulların yağ asidi profili üzerindeki etkilerinin sistematik olarak ortaya konmamış olması literatürde kritik boşluklar oluşturmaktadır. Bu çalışma, söz konusu boşlukları kapatmak amacıyla ABD'nin Houston, Teksas eyaletindeki Brays Bayou Nehri'nden örneklenen doğal bir mikroalg konsorsiyumunun büyüme dinamikleri, besin giderim kapasitesi, lipit üretim potansiyeli ve yağ asidi profili performansını iki aşamalı deneysel bir tasarımla sistematik biçimde incelemiş ve karşılaştırmalı olarak değerlendirmiştir. Birinci aşamada, Bold's Basal Medium (BBM) temel besi ortamına farklı oranlarda büyükbaş hayvan gübresi solüsyonu eklenerek hazırlanan kültür ortamlarında önce optimal zenginleştirme oranı belirlenmiş; klorofil floresansı, amonyum ve fosfat giderim verimleri birlikte değerlendirildiğinde 1:20 zenginleştirme oranının en yüksek büyüme performansını- klorofil floresansı 450,56 RFU, %99,78 amonyum ve %96,00 fosfat giderimi- sağladığı tespit edilmiştir. Bu optimal ortamda pH (7,5-8,5-9,5), sıcaklık (15-22,5-30°C), CO₂ konsantrasyonu (%0-2,5-5) ve ışık şiddeti (50-125-200 µmol foton m⁻² s⁻¹) parametrelerini kapsayan 11 farklı fizikokimyasal koşul kombinasyonu test edilmiştir. Fosfat giderimi tüm koşullarda %99'un üzerine çıkmış; amonyum giderimi ise %89,7–94,5 aralığında gerçekleşerek tüm koşullarda ulusal/uluslararası deşarj standartları karşılanmıştır. Klorofil floresansı ve kuru ağırlık verileri, koşullar arasında belirgin büyüme farklılıkları ortaya koymuş; en yüksek spesifik büyüme hızı Koşul 10'da (µ̄ = 0,3214 gün⁻¹), en yüksek kuru biyokütle Koşul 2'de (0,158 g) ve en yüksek lipit miktarı ile lipit verimliliği ise Koşul 9'da (0,05075 g; %35 kuru ağırlık) elde edilmiştir. Nile Red floresans analizleri, Koşul 3 ve Koşul 5'in lipit birikimini en güçlü biçimde teşvik ettiğini ortaya koymuştur. Tür kompozisyonu analizleri, konsorsiyumun fizikokimyasal koşullara yanıt olarak belirgin bir yönlendirilmiş seleksiyon sergilediğini göstermiştir. Yüksek performanslı koşullarda Sphaerocystis sp. ve Scenedesmus opoliensis baskın türler olarak öne çıkarken, düşük performanslı stres koşullarında Selenastrum bibraianum dominantlığı (%42,0-49,9) belirginleşmiş; Scenedesmus opoliensis ise geniş bir koşul yelpazesinde (%8,4–33,4) istikrarlı varlığını koruyarak konsorsiyumun omurga türü işlevi görmüştür. Koşul 4'te diyatom Nitzschia palea'nin (%25,2) öne çıkması, düşük sıcaklık ve ışık kısıtlamasının tür seçimi üzerindeki belirleyici etkisini somutlaştırmıştır. TOPSIS ile seçilen üst performanslı altı koşulda (Koşul 1, 2, 3, 5, 9, 10) gerçekleştirilen FAME analizleri, koşula özgü yağ asidi profillerinin oluştuğunu ortaya koymuştur. Palmitik asit (C16:0) tüm koşullarda baskın veya nadiren alt baskın yağ asidi olarak belirlenmiş (%25,62–31,22) ve oleik asit (C18:1) çoğunlukla ikinci sırada yer almıştır (%21,42–30,16). Koşul 3, yüksek oleik asit içeriği (%30,16) ve yalnızca bu koşulda tespit edilen palmitoleik asit (7-Heksadekanoik Asit Metil Esteri C16:1, %10,88) ile uluslararası biyodizel standartlarını tam karşılayan tek koşul olmuştur (setan sayısı: 53,4; iyot değeri: 97,8 g I₂/100g; CFPP: −2,0°C). Koşul 5, en yüksek linolenik asit (%21,24), en yüksek omega-3 içeriği (%25,41), en düşük omega6/omega-3 oranı (0,42) ve EPA (C20:5, %1,26) içeriğiyle fonksiyonel gıda, farmasötik ve akuakültür yemi uygulamaları için öne çıkmıştır. Koşul 9, yalnızca bu koşula özgü α-linolenik asit (C18:3n3, %10,84) ve yüksek C16:2 (%10,34) birikimi ile en iyi soğuk akış performansını (CFPP: −7,0°C) sergilemiş; EPA ise Koşul 1 (%0,99), Koşul 2 (%1,09) ve Koşul 5'te (%1,26) tespit edilerek bu koşulların akuakültür yemi potansiyelini ortaya koymuştur. Yağ asidi sınıf dağılımı incelendiğinde, SFA içeriğinin %30,30–36,43, MUFA içeriğinin %21,42–41,04 ve PUFA içeriğinin %23,60–48,28 aralığında değiştiği görülmüştür; en yüksek PUFA içeriği Koşul 9'da (%48,28), en yüksek MUFA içeriği ise Koşul 3'te (%41,04) elde edilmiştir. Omega6/omega-3 oranı tüm koşullarda 1,11'in altında kalarak WHO/FAO önerilen ≤5:1 sınırının çok altında gerçekleşmiştir. Kuru ağırlık (w=0,25), lipit miktarı (w=0,30), lipit verimi (w=0,30) ve spesifik büyüme hızı (w=0,15) kriterlerini farklı ağırlık katsayılarıyla eş zamanlı değerlendiren TOPSIS çok kriterli karar analizi, biyokütle, lipit (lipit miktarı ve % lipit verimi) ve büyüme performansını en dengeli biçimde optimize eden koşul olarak Koşul 9'u (Cᵢ* = 0,8267) birinci, Koşul 2'yi (Cᵢ* = 0,7981) ikinci, Koşul 10'u (Cᵢ* = 0,6402) üçüncü ve Koşul 3'ü (Cᵢ* = 0,5836) dördüncü sıraya yerleştirmiştir. İkinci aşamada, birinci aşamada TOPSIS ile belirlenen en iyi dört koşulun performans aralığını temsil eden iki uç koşul (Koşul 3: pH 9,5; 15°C; %0 CO₂; 200 µmol ve Koşul 9: pH 8,5; 22,5°C; %2,5 CO₂; 125 µmol) altında, BBM'ye sırasıyla büyükbaş hayvan gübresi (1:20 seyreltme), kümes hayvanı gübresi (1:50 seyreltme; optimal oran ayrıca belirlenmiştir) katılarak hazırlanan zenginleştirilmiş besi ortamları ve atıksu karşılaştırmalı olarak test edilmiştir. Besin giderimi açısından tüm ortam-koşul kombinasyonları deşarj standartlarını karşılamıştır: fosfat giderimi tüm koşullarda %99'un üzerinde gerçekleşmiş; amonyum gideriminde evsel atıksu ortamı W9 (%99,77) ve W3 (%99,73) ile öne çıkmış, kümes hayvanı gübresi P3 ve P9 (%98,34) ve büyükbaş gübresi C3 ve C9 (%97,89) bunu izlemiştir. KOİ gideriminde kümes hayvanı gübresi P3 koşulu (%96,61) en yüksek verimi sağlarken, TOK gideriminde P9 koşulu (%80,61) öne çıkmıştır. Büyüme ve biyokütle üretimi açısından Koşul 9, tüm ortamlarda Koşul 3'e kıyasla belirgin biçimde daha yüksek spesifik büyüme hızı sağlamış; en yüksek büyüme hızı C9 koşulunda (µ̄ = 0,3456 gün⁻¹) elde edilmiştir. En yüksek biyokütle üretimi (kuru ağırlık) evsel atıksu ortamlarında gerçekleşmiş; W3 (0,406 g) ve W9 (0,402 g) tüm koşullar arasında en yüksek değerlere ulaşmıştır. Lipit birikimi açısından ise Koşul 3 grubu tüm ortamlarda Koşul 9 grubuna kıyasla belirgin biçimde daha yüksek Nile Red floresans (RFU) değerleri sergilemiş; en yüksek mutlak lipit miktarı W3 koşulunda (0,09744 g), en yüksek lipit yüzdesi ise C9 koşulunda (%29) elde edilmiştir. İkinci aşama tür kompozisyonu analizleri, konsorsiyumun ortama özgü dinamik tür değişimleri sergilediğini ortaya koymuştur: evsel atıksu ortamında Golenkiniopsis parvula (%31,4) ve Scenedesmus acutus (%20,7) baskın türler olarak belirlenirken, büyükbaş gübresi ortamında Selenastrum bibraianum (%36,9) dominantlığını korumuş; W9 koşulu ise tüm koşullar arasında en dengeli tür dağılımını sergileyerek Golenkiniopsis parvula (%16,1), Scenedesmus bicaudatus (%15,8) ve Closteriopsis acicularis (%15,4) türlerinin neredeyse eşit oranlarda temsil edildiği bir konsorsiyum yapısı ortaya koymuştur. İkinci aşama GC-MS tabanlı FAME analizleri, besi ortamının yağ asidi sınıf dağılımını köklü biçimde yeniden şekillendirdiğini göstermiştir. SFA içeriği %14,10 (W9) ile %41,61 (C3) arasında, MUFA içeriği %0,00 (W9) ile %27,48 (P3) arasında ve PUFA içeriği %28,42 (C9) ile %51,18 (W3) arasında değişmiştir. Biyodizel kalitesi açısından C9 ve W9 koşulları EN 14214 uluslararası biyodizel standartlarını karşılamıştır. Fonksiyonel gıda ve omega-3 takviyesi açısından W3 (omega 3: %25,03; linolenik asit: %22,84; omega6/omega-3: 0,78) ve P3 (omega-3: %23,90 linolenik asit: %21,57; omega-6/omega-3: 0,60) öne çıkmış; C3 koşulunda tespit edilen EPA (C20:5, %1,17) akuakültür yemi potansiyelini ortaya koymuştur. Kozmetik ve biyolubrikant uygulamaları için P3 ve P9 (oleik asit: %27,48 ve %25,78) en uygun profili sunmuştur. Aynı dört kriter ve ağırlık yapısıyla uygulanan TOPSIS analizi, biyokütle üretimi, besin giderimi ve lipit verimliliğini en dengeli biçimde bir araya getiren sistem olarak W3 koşulunu birinci sıraya (Cᵢ* = 0,8988) yerleştirmiş; C9 (Cᵢ* = 0,4909) ve W9 (Cᵢ* = 0,4857) koşulları orta performans kategorisinde yer almıştır. Monokültür sistemlerin aksine, doğal konsorsiyumların gübre katkılı ve/veya atıksu ortamlarında yüksek dayanıklılık göstermesi, türler arası tamamlayıcılığın biyokütle ve lipit verimliliğini artırması ve farklı koşullar altında yüksek değerli yağ asitlerinin üretilmesi, konsorsiyum yaklaşımının belirgin üstünlüklerini ortaya koymaktadır. Çevresel koşulların tür kompozisyonunu ve dolayısıyla ürün profilini yönlendirme kapasitesi, mikroalg tabanlı sistemlerin hedef ürüne göre optimize edilebileceğini göstermekte; TOPSIS çok kriterli karar analizi ise bu çok boyutlu optimizasyon probleminin nesnel, sayısal ve tekrarlanabilir biçimde çözümünde güçlü bir metodolojik araç olarak öne çıkmaktadır.

In recent years, the need for integrated approaches that combine renewable energy production and high-value biomass recovery within a single process has grown considerably. Microalgae occupy a central role in this approach owing to their high and versatile biomass production capacities. Nevertheless, the environmental fragility of monoculture systems, the insufficiency of comparative evaluations of different waste source types, and the lack of systematic characterization of the effects of environmental conditions on fatty acid profiles represent critical gaps in the literature. To address these gaps, this study systematically investigated and comparatively evaluated the growth dynamics, nutrient removal capacity, lipid production potential, and fatty acid profile performance of a natural microalgal consortium sampled from Brays Bayou River in Houston, Texas, USA, using a two-phase experimental design. In the first phase, the optimal enrichment ratio was first determined in culture media prepared by supplementing Bold's Basal Medium (BBM) with cow manure solution at varying ratios; when chlorophyll fluorescence, ammonium and phosphate removal efficiencies were evaluated together, the 1:20 dilution ratio was found to provide the highest growth performance chlorophyll fluorescence (450.56 RFU), 99.78% ammonium removal, and 96.00% phosphate removal. In this optimal medium, 11 different physicochemical condition combinations encompassing pH (7.5–8.5–9.5), temperature (15–22.5–30°C), CO₂ concentration (0–2.5–5%), and light intensity (50– 125–200 µmol photons m⁻² s⁻¹) were tested. Phosphate removal exceeded 99% under all conditions; ammonium removal ranged from 89.7% to 94.5%, meeting national/international discharge standards under all conditions. Chlorophyll fluorescence and dry weight data revealed pronounced growth differences among conditions; the highest specific growth rate was obtained under Condition 10 (µ̄=0.3214 day⁻¹), the highest dry biomass under Condition 2 (0.158 g), and the highest lipid content and lipid yield under Condition 9 (0.05075 g; 35% dry weight). Nile Red fluorescence analyses revealed that Conditions 3 and 5 most strongly promoted lipid accumulation. Species composition analyses demonstrated that the consortium exhibited pronounced directed selection in response to physicochemical conditions. While Sphaerocystis sp. and Scenedesmus opoliensis emerged as dominant species under high-performing conditions, Selenastrum bibraianum dominance (42.0–49.9%) became pronounced under low-performing stress conditions; Scenedesmus opoliensis maintained stable representation across a wide range of conditions (8.4–33.4%), functioning as the backbone species of the consortium. The emergence of the diatom Nitzschia palea (25.2%) under Condition 4 concretely demonstrated the decisive role of low temperature and light limitation on species selection. FAME analyses conducted on the top six high-performing conditions selected by TOPSIS (Conditions 1, 2, 3, 5, 9, and 10) revealed the formation of condition-specific fatty acid profiles. Palmitic acid (C16:0) was identified as the dominant or rarely subdominant fatty acid across all conditions (25.62–31.22%), with oleic acid (C18:1) most frequently ranking second (21.42–30.16%). Condition 3 was the sole condition fully meeting international biodiesel standards (cetane number: 53.4; iodine value: 97.8 g I₂/100g; CFPP: −2.0°C), distinguished by high oleic acid content (30,16%) and palmitoleic acid (7-Hexadecenoic Acid Methyl Ester, C16:1; 10.88%) the latter detected exclusively under this condition. Condition 5 stood out for functional food, pharmaceutical, and aquaculture feed applications with the highest linolenic acid (21.24%), the highest omega-3 content (25.41%), the lowest omega-6/omega-3 ratio (0.42), and EPA (C20:5, 1.26%). Condition 9 exhibited the best cold flow performance (CFPP: -7.0°C) with a uniquely condition-specific accumulation of α-linolenic acid (C18:3n3, 10.84%) and elevated C16:2 (10.34%); EPA was additionally detected under Conditions 1 (0.99%), 2 (1.09%), and 5 (1.26%), highlighting the aquaculture feed potential of these conditions. Regarding fatty acid class distribution, SFA content ranged from 30.30% to 36.43%, MUFA from 21.42% to 41.04%, and PUFA from 23.60% to 48.28%; the highest PUFA content was obtained under Condition 9 (48.28%) and the highest MUFA content under Condition 3 (41.04%). The omega6/omega-3 ratio remained below 1.11 across all conditions, well within the WHO/FAO recommended ≤5:1 threshold. TOPSIS multi-criteria decision analysis simultaneously evaluating dry weight (w=0.25), lipid content (w=0.30), lipid yield (w=0.30), and specific growth rate (w=0.15) under differentiated weighting coefficients ranked Condition 9 (Cᵢ*=0.8267) first, Condition 2 (Cᵢ*=0.7981) second, Condition 10 (Cᵢ*=0.6402) third, and Condition 3 (Cᵢ*=0.5836) fourth as the conditions most comprehensively optimizing biomass, lipid (lipid content and % lipid yield), and growth performance in a balanced manner. In the second phase, two extreme conditions representing the performance range of the best four conditions identified by TOPSIS in the first phase (Condition 3: pH 9.5; 15°C; 0% CO₂; 200 µmol and Condition 9: pH 8.5; 22.5°C; 2.5% CO₂; 125 µmol) were applied, and BBM-enriched media prepared by supplementing with cow manure (1:20 dilution) and poultry manure (1:50 dilution; optimal ratio separately determined), as well as municipal wastewater, were comparatively tested. In terms of nutrient removal, all medium-condition combinations met discharge standards: phosphate removal exceeded 99% under all conditions; in ammonium removal, municipal wastewater medium stood out with W9 (99.77%) and W3 (99.73%), followed by poultry manure P3 and P9 (98.34%) and cow manure C3 and C9 (97.89%). For COD removal, the poultry manure P3 condition achieved the highest efficiency (96.61%), while the P9 condition led in TOC removal (80.61%). In terms of growth and biomass production, Condition 9 consistently provided significantly higher specific growth rates than Condition 3 across all media; the highest growth rate was obtained under the C9 condition (µ̄=0.3456 day⁻¹). The highest biomass production (dry weight) was achieved in municipal wastewater media; W3 (0.406 g) and W9 (0.402 g) reached the highest values among all conditions. In terms of lipid accumulation, the Condition 3 group consistently exhibited markedly higher Nile Red fluorescence (RFU) values than the Condition 9 group across all media; the highest absolute lipid content was obtained under the W3 condition (0.09744 g), and the highest lipid percentage under the C9 condition (29%). Second-phase species composition analyses revealed that the consortium exhibited medium-specific dynamic community shifts: Golenkiniopsis parvula (31.4%) and Scenedesmus acutus (20.7%) were identified as dominant species in municipal wastewater medium, while Selenastrum bibraianum (36.9%) maintained its dominance in cow manure medium; the W9 condition exhibited the most balanced species distribution among all conditions, revealing a consortium structure in which Golenkiniopsis parvula (16.1%), Scenedesmus bicaudatus (15.8%), and Closteriopsis acicularis (15.4%) were represented at nearly equal proportions. Second-phase GC-MS-based FAME analyses demonstrated that the growth medium fundamentally reshaped fatty acid class distribution. SFA content ranged from 14.10% (W9) to 41.61% (C3), MUFA from 0.00% (W9) to 27.48% (P3), and PUFA from 28.42% (C9) to 51.18% (W3). In terms of biodiesel quality, C9 and W9 conditions met EN 14214 international biodiesel standards. For functional food and omega-3 supplementation, W3 (omega-3: 25.03%; linolenic acid: 22.84%; omega-6/omega-3: 0.78) and P3 (omega-3: 23.90%; linolenic acid: 21.57%; omega-6/omega-3: 0.60) stood out; EPA detected under the C3 condition (C20:5, 1.17%) highlighted its aquaculture feed potential. For cosmetic and biolubricant applications, P3 and P9 (oleic acid: 27.48% and 25.78%) offered the most suitable profiles. Applying the same four-criteria weighted TOPSIS framework, W3 was ranked first (Cᵢ*=0.8988) as the system most comprehensively balancing biomass production, nutrient removal, and lipid yield; C9 (Cᵢ*=0.4909) and W9 (Cᵢ*=0.4857) were placed in the moderate performance category. In contrast to monoculture systems, the high resilience of natural consortia in manuresupplemented and/or wastewater environments, the enhancement of biomass and lipid productivity through interspecies complementarity, and the production of high-value fatty acids under varying conditions collectively highlight the distinct advantages of the consortium approach. The capacity of environmental conditions to direct species composition and thereby shape the product profile demonstrates that microalgae-based systems can be optimized toward target products; TOPSIS multi-criteria decision analysis emerges as a powerful methodological tool for the objective, quantitative, and reproducible resolution of these multi-dimensional optimization problems.
