-42- SUMMARY It has been known for a long time that microorganisms contain ç>(. -D-galactos idase. C\ -D-galactosidase enzyme is a fundamental material for sugar industry during recent years. Because of this reason it is very important that the isolation and purification of 0\ -D-galakc tosidase economically from an appropriate resource. In this study, 0\ -D-galactosidase was isolated from Lactobac i 1 lus brevis and then we have worked on the characte rization of this obtained preparation. A series of experiments were carred out such as;the examination of different isolation and purification techniques, determination of optimum pH and temperature and the removal of impurities. As an evaluation of the data of the above mentioned experiments, a highly active soluble C\ -D-galactosidase preparation was obtained. The fer- mantation time, propertion of inoculation and the Content of Sugar in the medium was studied primarily and it was Found that the medium, that contains molasses with a propertion of 2 '/. inculation, at 3*t hours fermantation time has the highest enzymatic activity. At the end of the fermantation, microorga nisms were removed by centr i f ugat ion at 3OO0 g for 15 minutes and were washed with 0,85 V, salt solution. Then the cells were sonicated and the resudues of cells were removed by centrifugate were precipitated at 70 '/. ammonium Sulfate satu ration and this precipitate was dialized against water.
