Özet
ÖZET Bu çalışmada, deneysel olarak tuz (NaCl) ile oluşturulan Asites Sendromu'nda immunohistokimyasal yöntemler kullanılarak, MMP-2 (matriks metalloproteinaz-2), MMP-9, matriks metalloproteinaz doku inhibitörü-1 (TEMP-1) ve TIMP-2 aktivitelerinin hastalığın oluşumundaki rollerinin incelenmesi amaçlandı. Çalışmada 1 günlük, erkek,“Avian Farm 43”hattı, 80 adet et tipi civciv ile 1 günlük, erkek,“Lohman LSL”hattı, 80 adet yumurtacı civciv olmak üzere toplam 160 hayvan kullanıldı. Et tipi ve yumurtacı civcivler kendi aralarında deneme ve kontrol olmak üzere 2 'şer gruba ayrılarak toplam 4 grup oluşturuldu. Asites oluşturmak amacı ile broyler ve yumurtacı gruplardan ilk ikisinin sularına sekizinci günden itibaren %0,6'lık NaCl eklendi. Bu grupların kontrolü olan grupların sularına herhangi bir uygulama yapılmadı. Klinik olarak asitese sadece tuz uygulanan broylerlerde rastlandı. Makroskobik olarak asites oluşan hayvanların karın boşluğunda saman şansı renkte, bazen fibrin pıhtıları da içeren sıvı birikimi dikkati çekti. Akciğer, karaciğer, böbrek ve kalpte çeşitli makroskobik ve mikroskobik lezyonlara rastlandı. Ticari olarak hazırlanmış monoklonal primer antikorlar kullanılarak Streptavidin- Biotin-Peroksidaz yöntemi ile akciğer, karaciğer, böbrek ve beyindeki MMP-2 ve 9 ile TIMP-1 ve 2 aktiviteleri değerlendirildi, immunohistokimyasal boyamaların skorlanmasında, boyanmanın dağılımı, yoğunluğu ve boyanan hücre tipleri göz önüne alınarak yan kantitatif bir değerlendirme yapıldı. MMP-2 ve MMP-9 enzim aktivitesinin normal ve asitesli hayvanlarda yaşın artışı ile birlikte belirli bir düşüş gösterdiği, ancak bu düşüşün asitesli hayvanlarda normal hayvanlara göre daha yavaş olduğu gözlendi. TIMP-2 aktivitesinin asitesli hayvanlarda normal hayvanlara göre daha yüksek olduğu görüldü. MMP-9 aktivitesinin TIMP-1 aktivitesini baskıladığı belirlendi. Sonuç olarak, et tipi tavuklarda tuzla oluşturulan deneysel Asites Sendromu'nda MMP-2 ve MMP-9 enzimlerinin kapillar damar bazal membranlarının tahribinde rolü olabileceği kanısına varıldı.

SUMMARY An Investigation on the Activities of Matrix Metalloproteinases in the Formation of Ascites Syndrome in Meat Type Chickens The aim of this study was to investigate the roles of MMP-2 (matrix metalloproteinase-2), MMP-9, TIMP-1 (tissue inhibitor of matrix metalloproteinase-1) and TIMP-2 in the formation of Ascites Syndrome in a salt (NaCl) induced experimental model. 80 male, day-old, Avian Farm 43 line, meat type chicks and 80 male, day-old, Lohman LSL line, egg type chicks were used. Meat type and egg type chicks were divided into two subgroups as experimental group and the control group, each. In order to induce ascites 0.6% NaCl was added into the drinking water of one subgroup of both meat and egg types beginning by day 8. The control groups were given normal water. Clinically ascites was observed only in the NaCl given meat type chickens. Macroscopically yellowish fluid accumulation, sometimes with fibrin clumps, was seen in the abdominal cavity of ascitic chickens. Various macroscopical and microscopical lesions were seen in the lungs, liver, kidneys and heart. The MMP-2, -9 and TIMP-1, -2 activities in the lungs, liver, kidneys and brain were evaluated with Streptavidin-Biotin-Peroxidase method using commercially available primary monoclonal antibodies. A semi-quantitative evaluation was done according to the staining of the cell types, and the distribution and intensity of the staining for immunohistochemical scoring. A decrease was seen in the MMP-2 and MMP-9 enzyme activities with the increasing age both in normal and ascitic chickens, however, the decrease in ascitic chickens was relatively slower than that in the normal chickens. TIMP- 2 enzyme activity was found higher in ascitic chickens than in normal chickens. MMP-9 enzyme activity was found to suppress the TIMP-1 enzyme activity. As a conclusion, it was suggested that MMP-2 and MMP-9 enzymes may have a role in the destruction of the basal membranes of the capillaries in the NaCl induced experimental Ascites Syndrome in meat type chickens.
