SUMMARY LEVELS OF HOMOCYSTEIN, S-ADENOSYLMETHIONINE AND ANTIOXIDANTS IN PATIENTS WITH HYPER- AND HYPOTHYROIDISM This study was carried out in 22 newly diagnosed hypothyroid (12 men, 10 women) and 30 newly diagnosed hyperthyroidism (10 men, 20 women) patients from the Endocrinology Department of Atatürk University Yakutiye Research Hospital. Fifteen similary age- and sex-matched healthy volunteers (8 men and 7 women) were taken as control group. In these patient and control groups, plasma homocystein (Hey), S- adenosylmethionine (SAM), serum malondialdehyde (MDA) and nitic oxide (NO) levels, superoxide dismutase (SOD) and glutathione peroxidase (GSHPx) activities were determined and the relations of these parameters were investigated. In the patients with hyperthroidism the mean plasma Hey levels (X ± SD; 8.4 ± 1.9 umol/L) was lower than those of the control group (9.1±2.8 umol/L), but this was statistically insignificant. It was observed that SAM levels of the same patient group (150.6 ± 29.6 nmol/L) were low in comparison to the control group (187.1 ± 20.8 nmol/L, p<0.05). It was also seen that plasma Hey (17.9±4.9 umol/L) and SAM (237.4±34.5 nmol/L) levels were higher in the hypothyroid group than in the control group (p<0.01 for Hey and SAM). MDA levels of the control group (23.5±7.4 mmol/L) were lower than those to both the patient groups (for hyperthyroid group: 27.7±7.8 mmol/L, p<0.01 and for hypothyroid group: 26.5±9.9 mmol/L, p<0.05). While there was not any significant difference between the control group (27.7±8.4 U/mL) and hyperthyroid group (28.12±7.6 U/mL) for the means of NO levels, the difference between the control group and hypothyroid group (17.2±8.7 U/mL) was statistically significant (p<0.01).SOD activities of patients with hyper- and hypothyroidism (1.8±0.1 U/mL, 237±0.7 U/mL, respectively) were lower than those of the control group (3.4±0.7 U/mL), (p<0.01 for hyperthyroid, p<0.05 for hypothyroid). There was no significant difference in GSHPx activities between control group (1927±534.8 U/mL) and patient groups (hyperthyroid: 1967±263.6 U/mL, hypothyroid: 2026.2±587.5 U/mL). In the correlation analysis, it was found that there were negative correlation between FT4 and SAM levels (r = -0.409, p<0.05) and positive correlation between FT3 and NO levels (r = 0.431, p<0.05) in hypothyroid group. There were negative correlation both between TSH and MDA levels (r = -0,482, p<0.05) and between FT3 levels and GSHPx activities (r = -0.479, p<0.05) in hyperthyroid group. In conclusion, it was not observed any direct correlation not only between increased Hey and SAM levels and the levels of thyroid hormones, but also between increased oxidative stress and decreased antioxidant capacity in the patients with hypothyroidism. In the patients with hyperthyroidism, it was found that Hey and SAM levels were low as compared with the control group independent of thyroid hormones. In these group in which oxidative stress was high, there was not any significant correlation between Hey and SAM levels and antioxidants. Key World: Homocystein, S-adenosylmethionine, hyperthyroidi, hypothyroidi And oxidatif stress
