Due to the level and duration of high altitude, lack of appetite, weight loss and oxidative damage in tissues can lead to form atrophy in skeletal muscles. Increase in the heat shock 72 protein level, mainly formed with heat, is involved in the protection of cells from different stresses. As a matter of fact, augmentation of HSP72 has been shown to decrease not only skeletal muscle athrophy but also oxidative protein damage. Therefore, the aim of this investigation is to determine that whether heat stress is effective to prevent from hypoxia inducible athrophy and oxidative tissue damage in rat skeletal muscle. Subjects were four month old thirty-five Wistar Albino male rats assigned to four groups: Hypoxia (HN=9), hypoxia with heat stress (HS=10), normoxia (NN=8) and normoxia with heat stress (NS=8). The hypoxia group was kept in simulated altitude of 6000m (%9.7 O2, 90.3 N2) for up to 15 days. Heat stress was applied two times with one day interval before the experiment and 8th day of experiment at the level of 41 0C during 60 minutes. At the end of the experiment all the rats were anesthetized with sodium pentobarbütirat (50mg/kg) and then killed via servical dislocation. Soleus (SOL),Plantaris (PLA) and Extensör digitorum longus (EDL) muscles were cut and removed. The soleus muscle was used to evaulate atrophy for this purpose its water content, dry weight and protein content were determined. PLA and EDL muscles were used for analyzing protein carbonil (PCO), advance oxidation protein products (AOPP), lipid hidroperoxide(LHP), total thiol (T-SH) and protein thiol (P-SH) groups. Results indicated that fifthteen days hypoxia application has had no effect on soleus muscle body weight ratio (p>0.05), but it decreased in protein content (p<0.05) while heat stress was preventing this effect (p<0.05). Hypoxia has raised the PCO and AOPP levels while causing decline in P-SH level in the PLA and EDL muscles (p<0.05) besides, the changes in PCO, AOPP and P-SH have been prevented by the heat stress practice (p<0.05). In addition, hypoxia application reduced the T-SH (p<0.05) and again heat shock stress has repressed a fall in T-SH level in the PLA muscle (p<0.05). On the other hand, hypoxia has not created any change in LHP level. In conclusion, in spite of the decline in protein content and oxidative protein damage in the rat skeletal muscle, heat stress application and consequently Hsp72 induction has a protective role.
