Özet
Bu çalışmada, Erzincan Garnizonu'ndaki birliklerin ihtiyacı için alımı yapılan -18 0C'de dondurulmuş-poşetlenmiş tavuk ve hindi etlerinin but, deri ve göğüs kısımlarının Salmonella spp. yönünden kontaminasyon düzeyleri konvansiyonel ve moleküler yöntemlerle araştırıldı.Araştırmada, askeri birliğin soğuk hava deposuna donmuş olarak teslim edilen tavuk ve hindi etlerinden, 200 adedi 2-8 0C'de çözündürüldü. Karkasların but, deri ve göğüs kısımları selektif ve selektif olmayan ön zenginleştirmeye tabi tutuldu. Etkenin izolasyonu amacıyla konvansiyonel kültür metodu kullanıldı. İzole edilen etkenler biyokimyasal testler ve Salmonella hızlı test yöntemi ile identifiye edildi. Ayrıca, selektif zenginleştirme aşamasında Rappaport Vassiliadis Broth (RVB)'dan alınan herbir numuneye ait süspansiyon Polimeraz Zincir Reaksiyonu (PZR)'na tabi tutuldu.Kültürel yoklamalarda analiz edilen tavuk deri, but ve göğüs numunelerinden sırasıyla % 16, % 7,5 ve % 5,5'inde Salmonella spp. saptandı. Hindi but, deri ve göğüs numunelerinin hiçbirinde Salmonella spp. tespit edilemedi. Polimeraz zincir reaksiyonu ile yapılan analizler sonucu numunelerin hiçbirinden Salmonella spp. saptanamadı.Sonuç olarak, Erzincan Garnizonunda tüketime sunulan tavuk ve hindi etlerinin üretiminde ve işlenmesinde teknolojik ve hijyenik kurallara belirgin bir şekilde uyulduğu kanaatine varıldı.

In this study, the drum stick, skin and chest samples of frozen-packed, at -18 oC, chicken and turkey?s meat that had been purchased for the need of military units in Erzincan Base were investigated for the contamination levels with Salmonella Spp. by conventional and molecular methods.In the present study, 200 samples collected from the chicken and turkey carcasses that had been delivered to and kept as frozen in the military units were thawed at 2-8 oC. The drum stick, skin and chest samples taken from the carcasses were underwent selective and unselective pre-enrichment. Conventional culture method was used for isolation of the agent. The isolated agents were identified by biochemical tests and Salmonella rapid test. Furthermore, Polymerase Chain Reaction (PCR) was applied to the suspension of every sample taken from the Rappaport Vassiliadis Broth (RVB) at the selective enrichment step.In the cultural examinations, Salmonella spp. was isolated in 16 %, 7,5 % and 5,5 % of the chicken?s skin, drum stick and chest samples respectively. Salmonella spp. was not isolated in any of the culture of turkey?s drum stick, skin and chest samples. Salmonella spp. could not be detected in any of the samples by polymerase chain reaction.In conclusion, the technological and hygienic rules were strictly obeyed during the production and processing stages of the chicken and turkey?s meats that were served for the consumption in the Erzincan Base.
