Özet
Çalışmamızda Wistar albino türü erkek sıçan kullanıldı. Deney hayvanları rastgele 3 eşit gruba (n=8) ayrıldı. Birinci gruba (Grup K) serum fizyolojik, ikinci gruba (Grup T10) 10mg/kg ve (Grup T20) 20mg/kg tramadol, üçüncü gruba (Grup D6) 6µg/kg ve (Grup D8) 8µg/kg deksketoprofen uygulandı. İlaçlar ratlara 0,1cc volümde intraperitonel yolla injekte edildi. Her hayvan için 0.dk, 15.dk, 30.dk, 45.dk, 60.dk ve 120. dakikalarda ağrı ölçüm testleri (Hot plate testi ve Von Frey testi) ayrı ayrı yapıldı ve veriler kaydedildi. Deney hayvanları optimize edilmiş ortamda barındırıldılar. Verilerin analizinde normal dağılım gösteren parametrelerin gruplar arası karşılaştırmalarında tek yönlü Anova testi ve farklılığa neden olan grubun tespitinde Post Hoc Tukey testi kullanıldı. Normal dağılım gösteren parametrelerin grup içi karşılaştırmalarında Paired-Samples T testi kullanıldı ve p<0,05 istatistiksel açıdan anlamlı kabul edildi.Hot Plate yönteminde, Grup T10'da kontrol grubuna göre 60. ve 120. dakikalarda, Grup T20'de 45.dakikadan itibaren, Grup D6 ve Grup D8'de, kontrole göre 45. ve 60. dakikalarda istatistiksel olarak anlamlı farklılıklar bulunmuştur (p<0,05). Von Frey metodunda T20 grubunda 30.dk'dan sonra istatistiksel açıdan anlamlı farklılık bulundu (p<0,05). D6 grubunda 30.dk ve 45.dk'larda kontrole göre anlamlı farklılık bulunurken D8 grubunda kontrole göre anlamlılık 15.dk'dan itibaren bulunmuştur (p<0,05).Sonuç olarak; termal ağrıda tramadolün, mekanik ağrıda ise deksketoprofenin uygun analjezik olabileceği düşünülmektedir.

Today, the opioit and NSAIDs are often used for acute pain relief. In this experimental study we aimed to determine the analgesic effects of tramadol, which is a synthetic opioid and dexketoprofen, a kind of NSAIDs.In our study Wistar albino male rats were used. The experimental animals (n = 8) were randomly seperated 3 equal groups. It was given normal saline for the first group (Group K), tramadol (10mg/kg and 20mg/kg) for the second group (group T), deksketoprofen (6?g/kg and 8?g/kg) for the third group (group D). Drugs was injected 0,1cc volume to rats intraperitoneally. Pain measurement tests (Hot Plate test and Von Frey test) of 0th, 15th, 30th, 45th, 60th and 120th minutes were separately performed for each animal and datas were recorded. The experimental animals were kept in optimized conditions. One-way ANOVA test was used for comparing between groups at the data showing normal distribution parameters and Tukey's post hoc test was used for the determination of the differences between the groups in data analysis. Paired-samples t test was used for within the group comparisons at the data showing normal distribution parameters and p<0.05 value was considered statistically significant.In the Hot Plate method, they were found statistically significant differences on the Group T10 in 60th and 120th minutes, on the Group T20 from 45th minutes and on the Group D6 and the Group D8 at the same time (45th and 60th minutes) to the control group (p<0.05). In the Von Frey's method it was found statistically significant difference on the Group T20 from 30th minutes the control group (p<0.05). Statistically significant differences were found on the Group D6 in 30th and 45th minutes, on the Group D8 from 15th minutes to the control group (p<0.05).As a result, tramadol and dexketoprofen may be appropriate analgesics for the thermal pain and the mechanical pain respectively.
