Wire electrodes (Ni/Cr, 80 ~lln in diameter) were implanted to the duodenum and jejunum of small intestine of overnight fasted male Sprague-Dawley rats under general anesthesia (100 mg/ kg ketamine and 0.75 mg/kg chlorpromazine i.p.) to record small intestine electro myographic (EMG) activity and to obtain detailed information by analyzing the myoelectrical signal with mathematical transformations. Three wire electrodes 5 mm apart were implanted for each recording site plus one wire electrode to the duodenum as ground electrode. An infusion catheter was positioned in the fundic region of stomach. After the recording of fasted myoelectrical activity for one hour, 4 g. standard laboratory chow was given to the rat and 12 ml 5.41 % glucose solution (300 mOsmol; 0.8 ml/min) was infused in 15 min. into stomach of another rat. Two MMCs were recorded in one fasted rat with 18 and 25 min. time periods. The avarage RMS (root mean square; mean activity in a time interval) values of phase I, phase II and phase III periods of these MMCs were calculated as 17.9, 102.2 and 323.5 respectively. Ingestion of 4 g. chow distrupted this cyclic activity and produced fed pattern consisting of irregular spike activity during the ensuing 197 min. The RMS value of this activity was calculated as 153.8, which is between the phase II and phase III values. Three MMCs were recorded in another fasted rat with 10.2, 12 and 11 min. time periods. The avarage RMS values of phase I, phase II and phase III periods of these MMCs were calculated as 6.7, 65.8 and 268.6 respectively. Infusion of glucose solution distrupted this cyclic activity and produced a new pattern consisting of irregular spike activity for 197 min. until a new phase III was restored. The RMS value of this activity was calculated as 2] 4.4, which is between phase II and phase III values. This new method ofEMG recording which is established in our laboratory brings some advantages. In this method, rats which have a short MMC cycle length were used. Wire electrodes are cheaper than other types of electrodes. The electro myographic recordings can undergo various mathematical analyses without changing the main signals. We aim to answer many questions about gastrointestinal motility by using this newly established method.
