Özet

Bir ya da daha fazla hidroksil grubu taşıyan -hidroksi--amino asit örnekleri gösterdikleri farmakalojik etkiden dolayı araştırmacıların ilgisini çekmektedir. Sekiz üyeli halka sistemlerinde -amino asit örneklerinin sayısı oldukça azdır. Bu nedenle siklooktanon'dan çıkılarak, prokiral ve kiral merkeze sahip klorosiyanosiklookten bileşiği sentezlendi. Klor'un (S)-b-metilbenzilamin ile yerdeğiştirmesi ile siklooktan ana iskelet yapısı içeren -amino asit öncüsü diastreomerler elde edildi. Bu diastereomerlerin hidrojenasjonunda istenilen seçimlilik sağlanamadı. Klorosiyanosiklookten bileşiğinden klorun eliminasyonu ile siyanosiklooktadien, bu bileşiğin oksidasyonu ile epoksisiyanosiklookten sentezlendi. Epoksisiyanosiklookten'in (S)-b-metilbenzilamin ile allilik pozisyondan açılma reaksiyonu başarısızlıkla sonuçlandı. Epoksit'in lewis asidi, kuvvetli ve zayıf bazik şartlarda açılmasıyla prokiral ve kiral merkezlere sahip hidroksisiyanosiklooktadien sentezlendi. Hidroksisiyanosiklooktadien'in Cl3CCN ve DBU eşliğinde gerçekleştirilen reaksiyondan g-hidroksi b-amino asit öncüleri elde edildi. Epoksit'in, metanolik HCl ile açılmasıyla, hidroksiklorosiyanosiklookten bileşiği sentezlendi. Elde edilen bileşiğin (S)-b-metilbenzilamin ile reaksiyonundan g-hidroksi b-amino asit öncüsü diastereomerler elde edildi. Enantiyosaf olarak sentezlenen diastereomerlerin mutlak stereokimyası X-Ray analizi ile belirlendi.

Özet (Çeviri)

-hydroxy--amino acid derivatives carrying one or more hydroxyl groups are of great interests to scientists due to their pharmacological activities. Since there are very few examples of eight membered cyclic systems containing -amino acids, we synthesized a chlorocyanocyclooctene compound containing both a prochiral and chiral center. The diastereomers of the -amino acid precursors containing the general cyclooctane structure were obtained via the substitution of (S)-b-methyllbenzylamine with chlorine. The expected selectivity was not obtained through the hydrogenation of these diastereomers. The elimination of chlorine from chlorocyanocyclooctene yielded cyanocyclooctadiene and this was further oxidized to epoxycyanocyclooctene. The epoxide ring opening from the allylic position of epoxycyanocyclooctene with (S)-b-methyllbenzylamine was unsuccessful. Hydroxycyanocyclooctadiene containing a prochiral and chiral center was synthesized via the opening of the epoxide's lewis acid in both strong and weak base conditions. The g-hydroxy b-amino acid precursors were obtained from the reaction of hydroxycyanocyclooctadiene in the presence of Cl3CCN and DBU. The hydroxychlorocyanocyclooctene compound was synthesized via the ring opening of the epoxide with methanolic HCl. Diastereomers of the g-hydroxy b-amino acid precursors were obtained from the reaction of (S)-b-methyllbenzylamine with the previously synthesized compound. The absolute stereochemistry of the synthesized enantiopure diastereomers were confirmed with X-Ray analysis.
