Cardiovascular disease is the most prevalent cause of human morbidity and mortality worldwide and atherosclerosis is the main underlying cause of it. Atherosclerosis is defined as the gradual thickening and hardening of large and medium size arteries and that can eventually result in blocked arteries or a break leading to a heart attack, stroke or other dysfunction. The chronic inflammatory process associated with atherosclerosis is known to be associated to various risk factors including high blood cholesterol levels, high blood pressure, smoking amongst others. There is an urgent requirement to find novel treatments for atherosclerosis. Probiotics are live microorganisms which have been shown to have beneficial effects on the host when administered in the diet. Lactic acid bacteria are amongst the most commonly microorganisms used as probiotics. They can be found as a part of fermented products such as yoghurt and in diet supplementary pills. Recent animal studies and human clinical trials have demonstrated that the regular intake of probiotics can significantly reduce plasma cholesterol levels, high blood pressure and oxidative stress, suggesting a potential beneficial use of probiotics-based diets as a cheap and effective method for treating and preventing cardiovascular disease. We hypothesised that since probiotics are known to have immunomodulatory effects on the host immune system, these may directly (or indirectly) influence the inflammatory process by which atherosclerotic plaques grow. In this study, Lactobacilli plantarum was used as the probiotic of choice and its effects on humoral and T cell mediated immunity and plasma lipid profile as well as in atherosclerotic plaque development were studied using an experimental animal model of the disease, the apoE-/- mouse. The survival of L. plantarum was confirmed after its isolation from faecal samples of treated animals. Flow cytometry analyses showed that L. plantarum was capable of increasing the proliferation of CD4+ regulatory T cells. It was also observed that the regular intake of L. plantarum significantly increases the level of IL-10 and decreases the size of atherosclerotic plaques in aortic sinus of apoE-/- mouse, without a decrease in cholesterol levels. In conclusion, the data derived from this study provide supporting data for the use of L. plantarum as potential therapeutic agent against atherosclerosis.
