Type 1 Diabetes (T1D) is an autoimmune disease that results from T cell-mediated destruction of insulin-producing beta cells of pancreas. Although genetic factors are believed to be a major component of disease susceptibility, the rapidly rising in the incidence of the disease suggested that environmental factors, especially viruses, have a role in the disease development. Considerable evidence coming from animal models and patients supported the link between viral infection and T1D. In this study, one group of enteroviruses, coxsackie B viruses (CVB), which have been largely suggested as a potential inducer of T1D were investigated. In order to understand if different serotypes of CVB might have role in T1D, I cloned several recombinant proteins of CVB antigens, developed immunoassays to detect antibodies against these, and used these to interrogate the presence of antibodies against the virus antigens in the collection of T1D patients and healthy controls. My results establish valid antibody assays against eight viral antigens from three CVB, and provide evidence for an increased antibody response to the CVB4 VP1 antigen in patients with type 1 diabetes as compared to controls.
