Agricultural water quality and animal intrusion are substantial food safety concern in produce fields. Validation of metrics for agricultural water and animal intrusion is essential for produce safety in Florida. Water samples from six agricultural ponds were collected during two consecutive growing seasons (540 samples). Microbial indicator populations (total coliforms, generic Escherichia coli, and Enterococci) were enumerated. A microbial water quality profile (MWQP) was established for all ponds. Water (150 mL) was filtered for pathogen analysis by traditional and multiplex PCR. Salmonella isolates were serotyped. All ponds met the current FDA MWQP recommendations 100.0% of the time. The invA gene (Salmonella) was detected in 26/540 (4.8 %) samples, in all ponds and growing seasons. Fourteen different Salmonella serotypes were detected. Six STEC genes were detected: hly-83.3%, fliC-51.8%, eaeA-17.4%, rfbE-17.4%, stx-I-32.6% stx-II-9.4%. A STEC cocktail and generic E. coli were inoculated into 100 mL EPA Worst Case water, sterile, and non-sterile surface water at 15°C and 25°C. At day 168, E. coli populations decreased ≤0.5 Log CFU/mL in non-sterile surface water and were 2.8≤ and ≤6.5 Log CFU/mL in sterile surface water and EPA Worst Case water.Rifampicin resistant cocktails of STEC and Salmonella were inoculated into domestic cattle, pig, waterfowl, deer, and raccoon feces at room temperature. Models to predict STEC, Salmonella populations based on generic E. coli were developed. Over the 364 day storage, STEC and Salmonella populations declined by ≤2.4 Log CFU/g in the cattle and deer feces; declined to ≤2.6 Log CFU/g in pig, waterfowl, and raccoon feces over the first 56 days. The calculated predictive power (R2) of the models relating pathogen declines to generic E. coli in each fecal type ranged from 0.4362 to 0.9774. Fate of generic E. coli in cattle fecal pats under different tomato field conditions was evaluated, and Salmonella and STEC survival was predicted using the developed models. Generic E. coli concentration stayed high under field conditions for a week, and different conditions did not impact survival dramatically. The concentration differences between generic E. coli and predicted pathogen populations in fecal pats was less than Log CFU/g at each time interval.
