Özet

91 SUMMARY Chemical mutagens intrupt DNA replication and cell division and may cause chromosome aberrations, both structural and numerical. A major group of chemical mutagens are drugs. These include alkylating agents, nucleic acid analogues, cytotoxics and antibiotics. In this study the mutagenic effect of Ifosfamide has been tested. Ifosfamide is a cytotoxic alkylating antineoplastic agent belonging to oxazaphosphorine family chromosome analysis has been performed on direct bone marrow preparations of 45 Wistar Albino rats (IS controls and 30 subjects) after in vivo application of 25 mg/kg, 100 mg/kg and 400 mg/kg of Ifosfamide. Mitotic index, chromosome structure and number changes has been compared in doses groups. Mitotic index has decreased significantly in the 100 mg/kg and 400 mg/kg groups. A significant increase in chromosome and chromatid type gaps and breaks, and numerical aberrations has been noted in all dosis groups. Increase in aberrations was proportional to the increase in dosis. As a result Ifosfamide has been found to have a highly mutagenic and especially clastogenic effect, supporting the previous findings.
