Özet

Dünyada, çiftlerin çocuk sahibi olamama (infertilite) oranı yaklaşık % 20'ye yakındır ve günümüzün en büyük problemlerinden bir tanesi de budur. Sağlık Bakanlığı verilerine göre Türkiye'de iki milyon çift çocuk sahibi olamıyor. Çiftlerin büyük bir kısmında çocuk sahibi olamama nedenini açıklayacak sebepler bulunabilirken, yaklaşık % 15'inde infertilite açıklanamayan gruba girmektedir. Sisplatin, çeşitli genitoüriner tümörlerin tedavisi için kullanılan, en etkili ve yaygın olarak reçete edilen antikanser ilaçlardan biridir. Fakat organlar üzerindeki toksik etkileri hala net olarak bilinmemektedir. Propolis bal arıları tarafından ağaçların kozalak ve kabuklarında, bitkilerin tomurcuk ve filizlerinden toplanan çeşitli yağlar, polenler, özel reçine ve mumsu maddelerin karışımından oluşan bir arı ürünüdür. Çeşitli organ sistemleri üzerinde yapılan çalışmalarla antioksidan özellikleri gösterilmiştir. Bizde yaptığımız bu çalışmada sisplatin kaynaklı testis hasarına karşı propolisin antioksidan ve antiapoptotik özelliklerini araştırmayı amaçladık. Deneyimizin toplam süresi ondört gün olarak belirlenmiştir. Deney için toplam 36 adet Spraque Dawley cinsi erkek sıçan kullanıldı. Her grupta altı adet sıçan bulunacak şekilde altı gruba ayrıldı. Kontrol grubuna tek doz izotonik solüsyon intraperitoneal (ip) olarak uygulandı. Sisplatin grubuna sadece çalışmanın ilk günü 7 mg/kg sisplatin ip olarak uygulandı. Propolis gruplarına (propolis 50 mg/kg ve propolis 100 mg/kg) ise 14 gün boyunca propolis 50 mg/kg ve propolis 100 mg/kg gavaj yoluyla verildi. Tedavi gruplarına (sisplatin + propolis 50 mg/kg ve sisplatin + propolis 100 mg/kg) tek doz 7 mg / kg sisplatin ip olarak uygulama sonrası deney süresi boyunca gavaj yolu ile verildi. Deney süresinin sonunda anestezi altında sıçanların testisleri epididimis ile birlikte çıkarıldı ve kansızlaştırma yöntemi ile sakrifiye edildi. Çıkarılan bir testis ikiye bölündü ve bir parçası histolojik incelemeler için kullanıldı. Diğer parçası ve plazma örneği ise biyokimyasal analizler için kullanıldı

In the world, the ratio of the couples inability to have children (infertility) is close to about 20% and this is also one of today's biggest problem. According to Ministry of Health data can not have two million pairs of children in Turkey. Couples will explain the cause of the inability to have a child found in a large part of the reason is unable to enter the group described in approximately 15%. Cisplatin, various genitourinary prescribed the most effective and widely used anti-cancer drugs are for the treatment of tumors. But toxic effects on the body are still not known clearly. Propolis honey bees by trees and cones in the bark of the plant oils collected from various buds and sprouts, pollen, bee products consisting of a mixture of special resins and waxy substances. Their antioxidant affect shown with studies on various organ systems. We aim to investigate antioxidant and antiapoptotic effects of propolis against cisplatin induced testis damage. In this study that we have done, we vere intended to research the antioxidant and antiapoptotic properties of propolis against cisplatin induced testicular damage. Fourteen days was determined as the total time of experiment. In this expermient thirty six male Sprague Dawley rats were used. They were divided into six groups of six rats. Rats of control group will be injected intraperitoneally (ip) at a dose of saline with a single injection. Just for the first day of the experiment, 7 mg/kg of cisplatin were injected intraperitoneally (i.p) with a single injection to the rats of cisplatin group. Propolis groups (propolis 50 mg/kg and propolis 100 mg/kg), were given by gavage of the propolis 50 mg / kg and propolis 100 mg / kg during the 14 days.The rats of treatment groups (sisplatin + propolis 50 mg/kg and sisplatin + propolis 100 mg/kg), were injected with 7 mg/kg of cisplatin on the first day of the study and propolis were given by gavage for fourteen days. At the end of the fourteen -day experiment, testicles with epididiymis of the rats were taken out and were sacrified with the method of making bloodless. One of the each pair of the testicles were divided into two pieces and one of them were used for histological processes. The other half and blood serum were used for biochemical analyses.
