Postmortem interval (PMI) determination is essential in forensic deaths in criminal and judicial cases. In judicial cases, determination of the time of death is required especially for the allocation of heritage. The various methods have been used in present practice subjectively. There is no way that given the certain results. With the increasing use of molecular studies in forensic science, significant efforts were mentioned particularly in the field of Forensic Genetics. In this study, our objective was determining the PMI using the postmortem early concentration changes of extracellular DNA and serum HMGB-1 protein that raising especially after the cell necrosis. The study was conducted on 96 Wistar rats whose weights ranging between 230- 260 g. The rats were allowed to stand at 4°C and +24°C temperature after anesthesia and cervical dislocation. Post-mortem blood samples were collected at the hours of 0, 3, 6, 9, 12, 24, 48 and 72. Five minutes-centrifugation separated the serum at 5000 rpm. HMGB-1 concentration was measured using HMGB1-ELISA kit and the amount of extracellular free DNA measured with the luminometer optical density after the nucleic acid staining protocol of SYBR Gold Nucleic Acid Gel Stain. The results obtained in this study were converted to concentration with equations that obtained from standard samples. The amount of extracellular free DNA increased with the postmortem period at +4°C (r =0.751 p<0.001). The serum concentration of HMGB-1 increased with the postmortem period (r =0.698 p<0.001). The negative correlation was found between postmortem period and the amount of extracellular free DNA at +24°C (r = -0.213 p=0.15), and the weak positive correlation was observed between the serum concentration of HMGB-1 and postmortem period at the same temperature (r=0.313 p=0.030). In conclusion, it was found that the amount of extracellular free DNA and the concentration of serum HMGB-1 can be used for determination of PMI at +4°C.
