Özet
Pineal bezden salgılanan melatonin hormonunun çinkonun sindirim sisteminden emilimini artırabileceğine dikkat çekilmektedir. Bu çalışmanın amacı da pineal bezleri çıkarılmış (px), pinealektomi sonrası melatonin uygulanmış (px+mel) ve pineal bezlerine dokunulmadan melatonin uygulaması yapılmış (mel) sıçanların ince bağırsak bölümlerinde (duodenum, jejunum, ileum) çinko emiliminde temel bir mekanizma olan metallotionin, ZnT2(çinko transport protein ), ZIP2 (ZRT ve IRT-benzeri protein), ZIP4 ve doku çinko düzeyleri arasındaki ilişkinin araştırılmasıdır. Bu çalışma Selçuk Üniversitesi Deneysel Tıp Araştırma ve Uygulama Merkezinden temin edilen Wistar cinsi erişkin erkek sıçanlar üzerinde aynı merkezde gerçekleştirildi. Çalışma protokolü aynı merkezin etik kurulu tarafından onaylandı. Toplam 36 adet sıçan kullanıp araştırmada gruplar şu şekilde oluşturuldu: Grup 1, (n:6) Kontrol: Hiçbir uygulamanın yapılmadığı normal diyetle beslenen grup Grup 2, (n:10) Pinealektomi: Pinealektomi yapılan ve normal diyetle beslenen grup. Grup 3, (n:10) Pinealektomili + Melatonin: Pinealektomi yapılan ve 4 hafta boyunca 5mg/kg/gün deri altı melatonin uygulanan grup. Grup 4, (n:10) Melatonin: Dört hafta süreyle 5mg/kg/gün deri altı melatonin uygulanan grup. Dört hafta süren çalışmaların bitiminde hayvanların tamamından dekapitasyonla gerekli analizlerde kullanılmak üzere ince bağırsak (duodenum, jejunum. İleum) doku örnekleri alındı. İmmünohistokimyasal çalışmalar için ayrılanlar içerisinde %10'luk formol bulunan kaplarda, doku çinko analizleri için kullanılacak örnekler ise analiz zamanına kadar -80oC'de muhafaza edildi. ZnT2, ZIP2, ZIP4 ve metallotionin yüzdeleri immünohistokimyasal yöntemle, doku çinko düzeyleri atomik absorpsiyon spektrofotometresiyle tayin edildi Duodenum, jejunum ve ileumdaki en düşük ZnT-2 düzeyleri pinealektomili grupta (grup 2) elde edildi (P<0,05). Grup 2 (Px)'nin duodenum ve ileumdaki ZIP-2 yüzdeleri diğer grupların tamamından önemli şekilde düşük bulunudu (P<0,05). Grup 2(Px)'nin jejunumdaki ZIP-2 değerleri ise kontrol grubundan (grup 1) düşük P<0,05), diğer gruplardan farklı değildi. Çalışmamızda duodenum ve jejunumdaki en düşük ZIP-4 seviyeleri pinealektomili grupta (grup 2) elde edildi (P<0,05). Gerçekleştirdiğimiz çalışmada grup 2 (Px)'nin duodenum ve ileumdaki metallotionin yüzdeleri diğer grupların tamamından önemli şekilde düşük bulundu (P<0,05). En yüksek ileum metallotionin değerleri melatonin uygulanan grup 4'de elde edildi (P<0,05). Çalışmamızın bulguları pinealektomi sonrası özellikle duodenumda azalan ZnT-2, ZIP-2, ZIP-4 ve metallotionin düzeylerinin, melatonin uygulamasıyla kontrol değerlerine yaklaştığını göstermektedir. Pineal bezden salgılanan melatonin hormonu sıçan ince bağırsağında ZnT-2, ZIP-2, ZIP-4 ve metallotionin düzeylerini etkileyerek çinkonun sindirim sisteminden emiliminde kritik bir rol oynar. Çalışmamızın melatoninin çinko taşıyıcı proteinler aracılığıyla çinkonun sindirim sisteminden emilimini düzenlediğini gösteren bulguları Pubmed taramalardan yola çıkılarak ilk ve orijinal bulgu olarak takdim edilebilir.

It has been noted that the melatonin hormone released from the pineal gland might increase the absorption of zinc in the digestive system. The objective of the present study is to explore the relationship between levels of metallotionin, ZnT2 (zinc transport protein), ZIP2 (ZRT and IRT-like protein), ZIP4 and tissue zinc levels, which comprise a basic mechanism in the absorption of zinc, in the parts of the small intestines (duodenum, jejunum, and ileum) of rats whose pineal glands were removed (px), which were supplemented with melatonin after pinealectomy (px + mel), and which were supplemented with melatonin without touching the pineal gland (mel). The study was carried out at the Experimental Medicine Research and Application Center of Selcuk University and registered Wistar type adult male rats supplied by the same Center. After the study protocol was approved by the ethics committee of the Center, a total of 36 rats were allocated to groups as follows: Group 1 (n:6), Control: The group which was not subjected to any procedure and fed by a normal diet. Group 2 (n:6), Pinealectomy: The group which was pinealectomized and fed by a normal diet. Group 3 (n:6), Pinealectomy + Melatonin: The group which was pinealectomized and then supplemented with 5 mg/kg/day subcutaneous melatonin for 4 weeks. Group 4 (n:10), Melatonin: The group supplemented with 5 mg/kg/day subcutaneous melatonin for 4 weeks. At the end of the four-week study period, the animals were decapitated to collect small intestine samples (duodenum, jejunum and ileum) to be used in analyses. The portion of the samples to be used in immunohistochemical analyses were placed into containers containing 10% formol and the samples to be utilized in tissue zinc analyses were stored at -80oC until the time of analysis. The percentages of ZnT2, ZIP2, ZIP4 and metallotionin were determined using the immunohistochemical method, and tissue zinc levels were studied with atomic absorption spectrophotometry. The lowest ZnT2 levels in the duodenum, jejunum, and ileum were found in the pinealectomized group (Group 2) (p<0,05). ZIP-2 percentages in the duodenum and ileum of Group 2 (Px) were established significantly lower than those in all other groups. Jejunum ZIP-2 values in Group 2 (Px) were lower than the levels in the control group (Group 1) (p<0,05), but not different than those in other groups. The lowest ZIP-4 levels in the duodenum and jejunum were identified in the pinealectomized group (Group 2) (p<0,05). Group 2 (Px) also had significantly lower duodenum and ileum metallotionin percentages than all other groups (p<0,05). The highest ileum metallotionin levels, on the other hand, were found in the melatonin-supplemented Group 4 (p<0,05). The results of the study indicate that reduced levels of ZnT2, ZIP-2, ZIP-4, and metallotionin, especially in the duodenum after pinealectomy are almost restored to control values after melatonin supplementation. It can thus be concluded that, melatonin hormone released from the pineal gland affecting the levels of ZnT-2, ZIP-2, ZIP-4 and metallotionin levels in the rat small intestines, plays a critical role in the absorption of zinc in the digestive system. In consideration of the med-line reviews, the results of the study suggesting that melatonin regulates absorption of zinc in the digestive system through zinc-transporting proteins can be presented as original findings reported for the first time.
