SUMMARY The mechanisms underlying opiate dependence and withdrawal are highly complex and still remain poorly understood. In animal experiments to elucidate the subject, various pharmacologic manipulations were applied and it was seen that both of them can be inhibited or intensified. In the present study, the effects of the thyroid states on the naloxone-precipitated abstinence syndrome in morphine dependent rats have been investigated. For this purpose Wistar Albino female rats were divided into four groups: One group of rats were induced hypothyroid state by adding 0.1 % propylthiouracil to their drinking water for three weeks. The rats in the other group were injected intraperitoneal^ with I- thyroxine (25 (jq/rat) twice a week for three weeks to induce hyperthyroid state. There were two control groups at the same period. All the animals were fed regular food ad libitum. At the end of this period, two pellets each one containing 75 mg morphine base were implanted subcutaneously on the back of rats under light ether anesthesia. They were injected with naloxone (2 mg/kg) by intraperitoneal^ 72 hours later from pellet implantation and were observed for 15 minutes in order to evaluate the characteristic abstinence syndrome signs. These are flying, jumping, wet-dog shakes, teeth-chattering, defecation, diarrhoea and ptosis. 43In comparison with the control groups hypothyroidy significantly attenuated the signs of naloxone-precipitated morphine abstinence syndrome; in contrast hyperthyroidy intensified the signs of the syndrome in general. Furthermore a close correlation have been found between the changes in plasma 'corticosterone level yin rectal temperature and in weight loss during naloxone-precipitated morphine abstinence syndrome with the changes in the signs of this syndrome. Further studies are required to clarify the mechanisms involved in morphine abstinence syndrome during the altered thyroid states. 44
