Özet

7. ÖZET Bu çalışmada bronkodilatör aktivite göstermesi beklenen on yeni 7- substitüe teofilin türevinin sentezlenmesi amaçlanmıştır. 2- naftiloksi ve 4 - kumariloksi alkil halojenürler ile reaksiyonundan 2 - naftol ve 4-hidroksi kumarin ile uygun dihaloalkanlar hazırlanmıştır. Sonra bu bileşikler sodyum teofillin ile DMF içinde 60 °C ısıtılarak 7-(2- naftiloksi) alkil ve 7-(4- kumariloksi) alkil teofilin yapısındaki bileşikler elde edilmiştir. Bileşiklerin fiziksel sabitleri ve Rf değerleri saptanmış. UV, ER, *H - NMR, Mass spektrum verileri ve elamenter analiz sonuçlan ile bileşiklerin yapılan kanıtlanmıştır. Sentezlenen bileşiklerin bronkodilatör aktiviteleri erkek ve dişi kobayların nefes borularının izole organ banyosunda mcelerımiştir. Bütün bileşikler dimetilsülfoksit içinde çözülmüş, asetil kolin ile oluşturulan kontraksiyonlar aminofilin ile karşılaştırmalı olarak kobay nefes borusu dokusunda sentez edilen bileşikler denenmiştir. Bileşik 8a en aktif bileşik olarak gözlenmiştir. 8. SUMMARY The synthesis often new 7 - substituted theophylline derivatives, which were expected to possess bronchodilator activity, was carried out in this study. First (2 - naphtyl) and ( 4 - coumaril)alkyl halogenurs were prepared by using 2 - naphtol and 4 - hydroxycoumarin with corresponding dihaloalkanes. Sodium theophylline was then refluxed at 60°C with those alkyl halogenurs in dry DMF to obtain the compounds having the 7-(2-naphtyloxy)alkyl and 7~(4- coumariloxy) alkyl theophylline structures. Physical properties and Rf values of the compounds were determined and UV, IR, ^-NMR, Mass spectral data and micro analysis results verified the structure of the target compounds. Bronchodilator activities of the target compounds were investigated on tracheas of male and female guinea pigs in isolated organ bath. All compounds were dissolved in dimethyl sulfoxide. The contractions induced by 10'3 M Acetyl choline were recorded after the samples were incubated with various concentrations of the compounds or aminophylline. The results were expressed as means ± SE. Among the compounds 8a was the most active compound.
