Özet

Amaç: Eksenatid, metformin ve kestane balının, Diabetes Mellituslu (DM) sıçanlarda ağırlık, kan glukozu, fruktozamin, kan lipit düzeyleri, karaciğer ve pankreas enzimleri üzerine etkilerini değerlendirmektir. Materyal ve Metod: Wistar Albino erkek sıçanlar (n=60) 10 gruba ayrılıp, 8 grupta Streptozosin ile DM oluşturuldu (kontrol ve bal grubu hariç). Gruplara bal, eksenatid ve metformin ayrı ayrı ve birlikte verildi. Deney başlangıcı ve 28 günlük deney sonu sıçanların ağırlıkları ölçüldü. Deney sonunda sıçanların serum glukoz, fruktozamin, total kolesterol, trigliserit, yüksek dansiteli lipoprotein (HDL), düşük dansiteli lipoprotein (LDL), aspartat aminotransferaz (AST), alanin aminotransferaz (ALT), amilaz ve lipaz değerleri ölçüldü. Bulgular: DM'li sıçanların hepsinde ağırlık kaybı gözlendi (p<0,05). Bal verilen diyabetik grupta, kontrol diyabet grubuna göre kan glukoz ve fruktozamin değerleri değişmedi. Kontrol DM grubuna göre bal verilen DM'li sıçanlarda total kolesterol, trigliserid ve LDL değerleri düştü (p<0.01), HDL değeri arttı (p<0.05). DM'li sıçanlarda eksenatid ve metformin tek başlarına kan glukozu ve fruktozamin değerlerini değiştirmedi. Beraber verildiklerinde glukoz, fruktozamin, trigliserit, LDL, AST ve ALT değerlerini düşürürken (p<0.01), amilaz seviyesini arttırdılar (p<0.01). Metformin, tek başına diyabetik sıçanlarda trigliserit, LDL ve AST düzeyini düşürürken (p<0.01), amilaz değerini arttırdı. Eksenatid diyabetik sıçanlarda tek başına trigliserit (p<0.01) ve LDL (p<0.05) değerini düşürürken, HDL ve amilaz değerini arttırdı (p<0.01). Sonuç: Bal, DM'li gruplarda lipid profiline olumlu tesir gösterirken, kan glukoz regülasyonu, karaciğer ve pankreas enzim düzeyleri üzerinde belirgin etkisi gözlenmedi. Eksenatid ve metformin kombinasyonu kan glukozu ve fruktozamin değerleri, kan lipit profili, AST, ALT ve amilaz değerleri üzerine olumlu etkileri dikkate alınabilir.

Aim: The aim of this study was to evaluate the effects of exenatide, metformin and chestnut honey on weight, blood glucose, fructosamine, blood lipid levels, liver and pancreatic enzymes in rats with Diabetes Mellitus (DM). Material and Method: Wistar Albino male rats (n=60) were divided into 10 groups and in 8 groups DM induced with Streptozosin (excluding control and honey group). Honey, exenatide and metformin were given to these groups separately and in combination. The weights of the rats were measured at the beginning and end of the 28-days experiment. At the end of the experiment, serum glucose, fructosamine, total cholesterol, triglyceride, high density lipoprotein (HDL), low density lipoprotein (LDL), aspartate aminotransferase (AST), alanine aminotransferase (ALT), amylase and lipase levels were measured from venous blood. Results: Weight loss was observed in all diabetic rats (p<0.05). Glucose and fructosamine levels did not change in DM group treated honey compared to DM control group. Total cholesterol, triglyceride and LDL values decreased and HDL values increased. Exenatide and metformin did not alter glucose and fructosamine values when they were applied separately to the diabetic rats. But when they were used together, they reduced glucose, fructosamine, triglyceride, LDL, AST and ALT (p<0.01) while they increased the amylase levels (p<0.01). Metformin decreased the triglyceride, LDL and AST in diabetic rats (p<0.01) and increased the amylase when it was applied separately. When the diabetic rats were treated only with exenatide, it decreased the triglyceride (p<0.01) and LDL (p<0.05) and increased HDL and amylase (p<0.01) levels. Conclusion: Honey showed positive effects on lipid profile in groups with DM, while no significant effect was observed on blood glucose regulation, liver and pancreatic enzyme levels. When exenatide and metformin were used in combination, their positive effects on blood glucose, fructosamine, lipid profile, AST, ALT and amylase levels may be considered.
