162 SUMMARY Studies on the Synthesis, Structure Elucidation of Methyl 1-Alkyl-2-N- Substituted Acetamido-1H-Benzimidazole~5-Carboxylate Derivatives and Their Antimicrobial Effects In this study, at first methyl 4-chloro-3-nitro benzoate (2) was prepared by the esterification of 4-chloro-3-nitro benzoic acid (1), then the chlorine atom of 2 was changed to amines by using the aromatic nucleofilic substitution reactions, following this, reduction of 3a-e afforded methyl 3-amino-4-N- alkylsubstituteamino benzoate derivatives (4a-e). The reaction of 4a-e with ethyl 0-amino-fJ-ethoxyacrylate hydrochloride gave ethyl 5-methoxycarbonyl- 1-alkylsubstituted-1H-benzimidazole-2-acetate derivatives (5a-e). The final products methyl 1-alkyl-2-N-substituted acetamido-1H-benzimidazole-5- carboxylate derivatives (6a-u) were prepared by the amidification of 5a-e ester compounds. A serious of new 31 compounds (1 1 intermediates and 20 final products) were prepared. The purity of these compounds were controlled by TLC and melting points of unhygroscopic derivatives were determined. The chemical structure of the compounds were elucidated by their IR, 1H- NMR, Mass spectral data and their elemantel analysis. The in vitro antibacterial and antifungal activity of the synthesized compounds against £ coli, S. aureus, B. subtilis and C. albicans were evaluated by the Macrodilution Broth and Disc Diffusion Techniques. Only compounds 6r and 6s were exhibited antibacterial activity against S. aureus with the MIC value of 50 ııg/ml.
