Özet

Bu çalışma kapsamında Cyanus depressus (M. Bieb.) Soják bitkisinin fitokimyasal özelliklerinin araştırılması ve insektisidal aktivitesi üzerinde incelemeler yapılması amaçlanmıştır. C.depressus'un toprak üstü kısımlarının iki farklı ekstraksiyon yöntemi (Soxhlet ve Ultrasonik) ile farklı polaritelere sahip üç farklı çözücü (metanol, n-hekzan, etil asetat) kullanılarak hazırlanan ekstrelerinin öncelikle insektisidal aktivitesine topikal aplikasyon yoluyla laboratuvar koşullarında 25 ˚C sıcaklık ve % 60 nemde Tribolium castaneum (Herbst) (Coleoptera:Tenebrionidae) ve Sitophilus oryzae (L.) (Coleoptera: Curculionidae) isimli böcekler üzerinde incelenmiş olup metanol ve n- hekzan ile hazırlanmış olan ekstrelerin insektisidal etkiye sahip olduğu görülmüştür. Çalışmada kullanılan C. depressus bitkisinin toprak üstü kısımlarından Soxhlet ve ultrasonik ekstraksiyon yöntemleri ile hazırlanan metanollü ekstrelerinin, içeriğindeki fenolik bileşikler ters faz YPSK yönemi ile kalitatif olarak analiz edilmiştir. Her iki ekstre içerisinde gallik asit, klorojenik asit ve luteolin ve varlığı tespit edilmiştir. Ayrıca, bu ekstrelerin farklı mobil fazlar içerisinde İTK incelemeleri sonucunda içeriğindeki etken madde grupları araştırılmıştır. İTK analizinde kullanılan çözücü sistemlerinden en uygun çözücü sistemi seçilerek YPİTK analizleri yapılmıştır.

In this study, it was aimed to investigate the phytochemical properties of Cyanus depressus (M. Bieb.) Soják and to examine its insecticidal activity. The insecticidal activity of the extracts of the aerial parts of C. depressus, which are prepared by two different extraction methods (Soxhlet and Ultrasonic) and three different solvents with different polarities (methanol, n-hexane, ethyl acetate), are first added to the insecticidal activity of the extracts under laboratory conditions at a temperature of 25 ˚C and a temperature of 25 ˚C. It was examined on insects named Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) and Sitophilus oryzae (L.) (Coleoptera: Curculionidae) at 60% humidity, and it was found that the extracts prepared with methanol and n-hexane had insecticidal effects. The phenolic compounds in the methanolic extracts prepared by Soxhlet and ultrasonic extraction methods from the above-ground parts of the C. depressus plant used in the study were analyzed qualitatively by reverse phase YPSK method. Gallic acid, chlorogenic acid and luteolin were found in both extracts. In addition, as a result of TLC examinations of these extracts in different mobile phases, the active substance groups in their contents were investigated. HPTLC analyzes were performed by choosing the most suitable solvent system from the solvent systems used in TLC analysis.
