Özet

Fermantasyon oranları, demleme koşulları (sıcaklık, süre), çay bitkisinin anatomik kısmı ve tüketim formlarına (servis şekli) göre hazırlanan çay infüzyonlarının içerdiği kafein ve L-theanin miktarlarının belirlenmesi amacıyla bu çalışma planlanmıştır. Bu doğrultuda farklı fermantasyon oranlarına sahip yeşil, beyaz, sarı, oolong, siyah ve Pu-erh çay olmak üzere altı çay türünün; çay bitkisinin farklı anatomik kısmından elde edilen (yaprak/filiz) siyah çayların; yeşil, beyaz ve siyah çayların farklı tüketim formlarının (poşet/dökme) infüzyonları 80°C ve 100°C sıcaklıklarında 2, 5 ve 10 dakika demleme sürelerinde deneysel demleme prosedürü ile hazırlanmıştır. Çay infüzyonlarındaki kafein ve L-theanin miktarlarının kromotografik analizi ters fazlı yüksek performanslı sıvı kromotografisi (RP-HPLC) ile analiz edilmiştir. Farklı fermantasyon oranlarına sahip aynı koşullarda demlenen çay infüzyonları için sarı çay; yeşil, beyaz ve oolong çaylara kıyasla önemli düzeyde yüksek kafein miktarlarına sahipken (p<0,001) siyah ve Pu-erh çaylar ile benzer miktarda kafein içerdiği saptanmıştır (p>0,001). Aynı demleme koşulları altında L-theanin miktarları ise en yüksek siyah ve yeşil çay infüzyonlarında tespit edilmiş ve bu çayların diğer çay türleri ile aralarında önemli bir fark bulunmuştur (p<0,001). Farklı demleme sıcaklıklarında demlenen çay türleri içerisinde sadece oolong çayında 80°C ve 100°C sıcaklıkta demleme sonucunda kafein miktarları birbirlerinden önemli bir şekilde farklı iken (p<0,05) tüm çay türleri için farklı demleme sıcaklıklarında L-theanin miktarları arasında önemli bir fark bulunmamıştır (p>0,05). Farklı demleme sürelerinin tüm çay türlerinde hem kafein hem L-theanin miktarlarına etki ettiği ve özellikle 2 dakika demleme süresi ile diğer sürelerdeki belirlenen miktarlar arasındaki farkın önemli olduğu tespit edilmiştir (p<0,05). Farklı anatomik kısımların kullanılması ile hazırlanan siyah çay infüzyonlarında yaprak çayların kafein miktarları filiz çay infüzyonlarının kafein miktarlarından yüksektir (p<0,001). Yeşil çayların 80°C demleme sıcaklığında 5 ve 10 dakika demleme sonunda bardak poşet çay infüzyonlarında tespit edilen kafein miktarları dökme çay infüzyonlarından yüksek bulunurken (p<0,05) siyah çaylarının ise tüm koşullarda bardak poşet çay infüzyonlarındaki kafein miktarı demlik poşet ve dökme çay infüzyonlarının içerdiği kafein miktarından önemli derecede yüksektir (p<0,001). Beyaz çayların kafein miktarı ile yeşil, beyaz ve siyah çayların L-theanin miktarları farklı tüketim formu kullanılarak hazırlanan infüzyonlarda önemli bir farklılık göstermemiştir (p>0,05). Sonuç olarak çayın fermantasyon oranları, demleme koşulları (sıcaklık, süre), anatomik kısımları ve çayın tüketim formlarının (servis şekli) çay infüzyonlarının kafein ve L-theanin miktarlarına etki edebileceği belirlenmiştir.

This study was planned to determine the amounts of caffeine and L-theanine contained in tea infusions prepared according to fermentation rates, brewing conditions (temperature, time), anatomical parts of the tea plant, and consumption forms (serving form). In this respect, infusions of six tea types, including green, white, yellow, oolong, black, and Pu-erh tea, with different fermentation rates; black teas obtained from different anatomical parts of the tea plant (leaf or sprout); and different consumption forms of green, white, and black teas (bags or bulk), were prepared with experimental brewing procedures at 80°C and 100°C temperatures for 2, 5, and 10 minutes brewing times. Chromatographic analysis of caffeine and L-theanine amounts in tea infusions was performed by reversed-phase high-performance liquid chromatography (RP-HPLC). For tea infusions brewed under the same conditions with different fermentation rates, yellow tea was found to contain significantly higher amounts of caffeine compared to green, white, and oolong teas (p<0,001), while black and Pu-erh teas contained similar amounts of caffeine (p >0,001). Under the same brewing conditions, the highest amounts of L-theanine were found in black and green tea infusions, and a significant difference was found between them and other tea types (p<0,001). Among the tea types brewed at different brewing temperatures, only the caffeine amounts of oolong tea were significantly different from each other as a result of brewing at 80°C and 100°C (p<0,05), while no significant difference was found between the L-theanine amounts at different brewing temperatures for all tea types (p>0,05). It was determined that different brewing times affected both caffeine and L-theanine amounts in all tea types, and especially the difference between the amounts determined in 2 minutes of brewing time and other times was significant (p<0,05). In black tea infusions prepared by using different anatomical parts, the caffeine content of leaf teas was higher than the caffeine content of sprout tea infusions (p<0,001). While the caffeine content of green teas was higher than that of bulk tea infusions at the end of 5 and 10 minutes of brewing at 80°C brewing temperature (p<0,05), the caffeine content of black teas was significantly higher than the caffeine content of teabags and bulk tea infusions in all conditions (p<0,001). The caffeine content of white teas and the L-theanine content of green, white, and black teas did not show a significant difference in the infusions prepared using different consumption forms (p>0,05). As a result, it was determined that fermentation rates of tea, brewing conditions (temperature, time), anatomical parts, and consumption forms of tea (serving method) may affect the caffeine and L-theanine amounts of tea infusions.
