ÖZET Yüksek Lisans Tezi BİTKİ EKSTRESİ İÇEREN NAZAL FORMÜLASYON ÇALIŞMASI ve İN VİTRO-İN VtVO DEĞERLENDİRİLMESİ FUNDA ÖZTAN Anadolu Üniversitesi Sağlık Bilimleri Enstitüsü Farmasötik Teknoloji Anabilim Dalı Danışman: Prof.Dr. Yasemin YAZAN 2000 Nazal yol uzun yıllar boyunca lokal etki sağlamak için kullanılmıştır. Nazal formülasyonlara gittikçe artan ilgi, nazal yolun uygulama kolaylığı ve nazal mukozanın istenen etki için uygun şartlara sahip olması nedeniyledir. Ecballium elaterium Türkiye'de yaygın olarak bulunan ve sıkma yoluyla elde edilen suyu. halk arasında sinüzit tedavisinde kullanılan bir bitkidir. Sıkma ile elde edilen meyve suyunun kararlı olmaması nedeniyle, bu çalışmada nazal formülasyon çalışmaları yapılmıştır. Ecballium elaterium meyvelerinin suyu alındıktan sonra süzgeç kağıdından süzülmüş ve liyofîlize edilerek toz şeklinde nazal formülasyonlarda etkin madde olarak kullanılmıştır. Formülasyon çalışmalarında, farklı özellikte maddeler içeren alü değişik fonnülasyonun ön çalışmaları yapılmıştır. Bu ön formülasyonlar, renk ve kıvam değişikliği, zamanla çökelti veya faz ayrışması olup olmadığına göre gözlenmiş, santrifujleme testi yapılmış ve bu incelemelerin sonuçlarına göre değerlendirilerek en uygun olan formülasyon seçilmiştir. Seçilen formülasyon, Methocel®, Carbopol®, Vit E, benzalkonyum klorünbenzil alkol karışımı, toz ekstre ve distile su kullanılarak hazırlanmıştır. Bu fonnülasyonun fiziksel özellikleri değerlendirilmiş, üzerinde santrifüj ve stabilite testleri, pH ölçümü, reolojik analiz ve in vivo histolojik çalışmalar yapılmıştır. İn vivo histolojik çalışmalarda, formülasyonlann sıçan nazal mukozasındaki etkileri incelenmiştir. Yapılan fiziksel, kimyasal ve in vivo çalışmalar sonucunda, hazırladığımız nazal fonnülasyonun kararlı olduğu, pH'sının burun mukozası pH'sına uygun olduğu, ve histolojik olarak etkin olduğu bulunmuştur.

ABSTRACT Master of Science Thesis NASAL FORMULATION of a PLANT EXTRACT and m viTRO-m vrvo evaluation FUNDA ÖZTAN Anadolu University Graduate School of Health Sciences Pharmaceutical Technology Program Supervisor: Prof.Dr. Yasemin YAZAN 2000 Nasal route has been used to obtain local activity for long years. There is an increasing interest in the nasal formulations because of the ease of administration and the appropriate conditions that nasal mucosa has for required effects. Ecballium elaterium is a plant found widespread in Turkey and whose juice obtained by pressing the fruits has been used for the treatment of sinusitis. Nasal formulation studies were performed in this study because of the instability of the fruit juice. Following the obtention of the fruit juice of Ecballium Elaterium, it was filtered through filter paper and liyofilized. This powder was used as the active material in nasal formulations. Six different formulations containing materials with different properties were preformulated during formulation studies. These preformulations were observed according to changes in colour, viscosity, formation of sedimentation or phase separation and centrifugation tests were performed. The results were evaluated and the most suitable formulation was selected. The selected formulation was prepared using Methocel®, Carbopol®, Vlt E, benzalchonium chloride:benzyl alcohol mixture, powder extract and distilled water. The physical properties of this formulation were evaluated and, centrifugation tests, stability tests, pH measurements, rheological analysis, and in vivo histological studies were performed on this formulation. The affects of the formulation on rats' nasal mucosa were examined by in vivo histological studies. According to the results of the physical, chemical and in vivo studies, it was found that the nasal formulation prepared was stable, its pH was suitable to the pH of the nasal mucosa and it was affective histologically.
